Production and Characterization of Cholesterol Oxidase from Streptomyces sp. No.4 (방선균으로부터 Cholesterol Oxidase의 생산 및 특성)

  • An actinomycetes strain No.4 which produce the cholesterol oxidase(EC 1.1.3.6), was isolated from soil and identified as Streptomyces sp. based on taxonomic studies. The conditions of cholesterol oxidase production and enzymatic properties were investigated. The optimum composition of medium for production of the enzyme was 1% soluble starch, 2% corn steep liquor, 0.1% $KH_2PO_4$, 0.1% $NaNO_3$ and 0.05% $MgSO_4$ (pH 7.0). The optimum pH and temperature of the cholesterol oxidase were pH 6.0~7.5 and $37^{\circ}C$, respectively. The enzyme was stable in the range of pH 6.0~9.0. The isoelectric point determined by multichambered electrofocusing unit was in the range of pH 6.0~6.5.

  • The conditions for the $\beta$-fructofuranosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at 50$\circ$C for 36 hrs with batch culture in the optimal medium containing 1.0% sucrose, 0.6% yeast extract, 0.1% each of KH$_{2}$PO$_{4}$, K$_{2}$HP0$_{4}$, NaH$_{2}$PO$_{4}$, and initial pH 9.5, and the final enzyme activity under the above condition was 102 units per ml of cell free extract.

Effect of High Concentrations of Sodium or Chloride Salts in Soil on the Growth of and Mineral Uptake by Tomatoes (토양에의 고농도 Na 및 Cl 염류가 토마토의 생육 및 무기성분 흡수에 미치는 영향)

  • This study was conducted to investigate the effect of high concentration of sodium salts and chlorides in soil on the growth of tomato and the uptake of minerals. The growth inhibition rates of plant height and dry weight were different depending on salts, but they were not related to the electric conductivities (EC) and acidities (pH) in the soil solution. The orders of growth inhibition were Cl, SO$_4$, CO$_3$, PO$_4$>NO$_3$ in the sodium salts series, and Na, K, Mg, NH$_4$>Ca in the chlorides. The growth inhibition rates of the sodium salts series tended to be larger than those of the chloride series. Yield was lower 30%~10% in the sodium salt and chloride series than in the control. Chlorophyll content, photosynthetic rate and stomatal conductance were lower in the sodium salts and chloride series than in the control. Mineral concentration was lower in sodium salts and chlorides than in control. The nitrate absorption was inhibited in all salts except for NaNO$_3$ and NH$_4$Cl, and specially in NaCl and Na$_2$SO$_4$ treatments of the sodium salts and in KCl treatment of chloride series. K concentration was reduced NaCl and Na$_2$SO$_4$ treatments compared with the other salts. In the sodium salt series, calcium and magnesium concentration were decreased antagonistically when sodium concentration was increased.

  • A microorganism capable of producing high level of extracelluar cyclomaltodextrin glucanotransferase(EC 2.4.1.19; CGTase) was isolated ’rom soil. The isolated strain No. 239 was identified as Bacillusstearothermophilus. The maximal CGTase production (about 7.0 unitslml) was observed in medium containing2% soluble starch, 0.5% defatted soybean meal, 0.1% NaH_2PO_4.2H_2O$ and 0.015% $ CaC_l2 $ with initial pH 7.0. The strain was cultured at $55^{\circ}C$ for 48 hr with reciprocal shaking. At 0.83% substrated concentration potato starch was the optimum substrate with 50.1% conversion to cyciodextrin (CD)after the reaction at $65^{\circ}C$ for 24 hr (CGTase 10 unitlg starch). Using soluble starch as substrate (5% substrate concentration, CGTase 10 unitlg starch), the maximum conversion of 40% was obtained at11 hr reaction, and the ratio of $\alpha-, \beta-$ and $\gamma$-CD production at this time were 1.0:1.3:0.4, respectively., respectively.

  • The time-lag-effect of alkali salts on the gelation of sodium alginate-$CaSO_4{\cdot}$1/2 $H_2O$ is compared with Miyake's data, and then the formation rate of the elastics measured by the continuous method (an improved Schwedoff's method) and the change of rigidity with metallic oxides are studied as follows: (1) The gelation processes of sodium alginate and $CaSO_4{\cdot}$1/2 $H_2O$-aqueous sol are studied by measuring, continuously the increases of tensions ofsamples. (2) The time-lag-effect of $Na_3PO_4$ on the formation rate of the elastic gel is larger than that of $Na_2CO_3$, but the difference between the effects of the two alkali salts on the rate is found not so greater than predicted in Miyake's data. (3) Any regularities of the effect on the rate by metallic oxides are not observed. The increasing effects of the rates of $SiO_2$ and MgO are relatively large, and that of ZnO is relatively small. However, $Al_2O_3$, $Sb_2O_3$ and $TiO_2$show some decreasing effects. As a result it is noted that the regularities do not depend on the effect of oxide species and their amounts. (4) It is not found proportionality between the rigidity and the gelation rate. However, the increasing effect of the rigidity with the addition of metallic oxides can be observed. The rigidity increasing rate of MgO is the largest of them.

  • We introduced a novel sustainable slow-releasing agrochemical formulation, a biopolymer bound to silica, for controlling plant diseases. The formulation was obtained through the following process. Curdlan, sodium silicate ($Na_2SiO_3$) and isopropyl alcohol were dissolved in DDW (Deionized-distilled water). The resultant solution was then irradiated using a $^{60}Co$ ${\gamma}$-irradiator (150 TBq of capacity; ACEL, Canada) at KAERI. The resultant solution was treated with phosphorous acid ($H_3PO_3$). Finally, we obtained a novel biopolymer-silica microsized formulation containing phosphorous acid ($H_3PO_3$) from the solution. The morphology of the complex was characterized using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). SEM and TEM images revealed that the curdlan-silica formulation has a particle size ranging from 1 to $3{\mu}m$ with high stability. We also detected that $H_3PO_3$ was distributed within the formulation through energy dispersive X-ray spectroscopy (EDX) analysis. $H_3PO_3$ was sustain-released from the formulation in water. Based on our results, it seems effectively that one or two applications of the formulation during a cropping season will assist in controlling various plant diseases.

  • Nanotube morphology change of Ti-Ta-Zr alloy as Zr content increase has been researched using various experimental methods. Ti-Ta-Zr(3, 7 and 15 wt%) alloys were prepared by arc melting and nano-structure controlled for 24 hr at $1000^{\circ}C$ in argon atmosphere. Formation of oxide nanotubes are conducted by anodizing a Ti-Ta-Zr alloy in $H_3PO_4$ electrolytes with small amounts of fluoride ions at room temperature. Electrochemical experiments were carried out with conventional three-electrode configuration with a platinum counter electrode and a saturated calomel reference electrode. The samples were embedded with epoxy resin, leaving a square surface area of $10mm^2$ exposed to the anodizing electrolyte, 1.0M $H_3PO_4$ containing 0.8wt% NaF.

  • In order to investigate on hydroxyapatite precipitation phenomena on nanotubular Ti-29Nb-xHf ternary alloys, Ti-29Nb-xHf alloys contained (0% to 15%) Hf were manufactured using arc melting furnace. Formation of nanotubular structure was achieved by an electrochemical method in 1M $H_3PO_4$ electrolytes containing 0.8%wt. % NaF. Electrochemical deposition was carried out using cyclic and voltammetry(CV) method at $85^{\circ}C$ in $5mM\;Ca(NO_3)_2+3mM\;NH_4H_2PO_4$. HA coating on nanotube formed Ti-29Nb-xHf ternary alloys showed a good wettability.

  • 1) For the purpose of preparation of food and fodder yeasts from nonedible seaweed, two suitable Candida yeasts have been isolated from seaweed compost. 2) They had the ability of fermenting galactose, sucrose and glucose, and could not ferment maltose and mannit, but could assimilate mannit. 3) NaCl concentration from 1 to 2% had no remarkable effect on growth of yeast and the optimum pH was $4{\sim}5$. 4) In the acid hydrolyzate of brown seaweed (Ecklonia cava Kjellman, Sargassum fulvellum AGARDH) an amorphous deposit was produced during storage after neutralization of media and its removal always delayed yeast growth, but addition of $(NH_4)_2SO_4$ and $NaH_2PO_4$ to media could increase the assimilation of reducing sugar and yeast yield. 5) $Co^{60}$ gamma ray irradiation (dose rate : 1 Mrad/hr, BNL shipboard irradiator) of seaweed had not so much effect on the hydrolysis of carbohydrates and nitrogen compounds in seaweed but could increase the yeast production from seaweed hydrolyzate. 6) The yeast yield was $7{\sim}8$ g of dry yeast per 100 g of seaweed by cultivation with jar fermentor.

  • An experiment was conducted to determine for the optimal phosphorus supplemention to carp feed which contains $50\%$ soybean meal, with $32\%$ protein content to obtain maximum growth of the Israeli strain of common carp (Cyprinus carpio). The experimental fish were between 100 g and 1000 g in body weight. The results, obtained were as follows: the optimum amount of phosphorus added to the experimental feed was $1.6\%$ as $NaH_2PO_4{\cdot}2H_2O$ which brings up the total amount of available phosophorus to $0.504\%$ of the feed. This amount is slightly less than the amount, $0.6-0.7\%$ which was previously reported for the Japanese native strain of carp which weighted around 4.5g. Thus there seems to be some difference for the requirement of phosphorus between the sizes of carp.
