A Kinetic Study of Phosphate Absorption by Rice Roots (벼에 의한 인산흡수의 기작에 관한 연구)

  • Phosphate absorption from a Na2H32PO4 solution by Oryza sativa L. was studied in order to elucidate kinetic mechanisms of ion transport. The rates of phosphate absorption from different concentraitons indicated the presence of dual mechanisms in root tips, one in the low (1$\times$10-6 to 8$\times$10-5M) and the other in the high (1$\times$10-4 to 8$\times$10-3M). A phosphate compensation point of phosphate transport was revealed with a 1$\times$10-6M solution of Na2H32PO4. The kinetic model that ion transport involves an exchange reaction of absorption and desorptin is prosposed as follows: where C represents an ionic-specific organic carrier in the membrane; M, Mo and Mi are the mineral ions, M-outside and M-inside; MC is a carrier-ion complex; and the K's represent rate constants. In this model, the Mi velocity, v, is given by: {{{{v= {dMi} over {dt}= {(K1K3Mo-K2K4Mi) Ct} over {(K2+K3)+K1Mo+K4Mi} }} where Ct is equal to C+MC, and t is time.

  • Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in response to changes in the sucrose content $(2{\sim}8%)$, nitrogen content $(0{\sim}250mM\;NH_4NO_3)$, phosphate content $(0{\sim}12mM\;NaH_2PO_4)$, and the potassium content $(0{\sim}87.2 mM\; KCl)$ of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62mM $NH_4NO_3$ 1.2 mM $NaH_2PO_4$, and 0.5mM KCl.

  • Nanorod graphitic carbon nitride (g-C3N4) was synthesized by reacting melamine (C3H6N6) with trithiocyanuric acid (C3H3N3S3) in distilled water for 10 h at room temperature. The resulting mixture was calcined at 550℃ for 2 h in an electric furnace under an air atmosphere. Nanorod g-C3N4/Ag3PO4 composites were prepared by adding nanorod graphitic carbon nitride (g-C3N4) powder, silver nitrate (AgNO3), ammonia (NH3·H2O, 25.0-30.0%), and sodium hydrogen phosphate (Na3HPO4) to distilled water. The samples were characterized via X-ray diffraction, scanning electron microscopy, and Fourier-transform infrared spectroscopy. The photocatalytic activities of the nanorod g-C3N4/Ag3PO4 composites were demonstrated via the degradation of organic dyes, such as methylene blue and methyl orange, under blue light-emitting diode irradiation and evaluated using UV-vis spectrophotometry.

  • This study was carried out to investigate the efficient in vitro mass propagation methods for juvenile sporophytes of Matteuccia struthiopteris. Chopped segments of pinnae, petiole and rhizome were cultured on 1/2MS with 0.1% activated charcoal. Among these explant sources only rhizome segments produced young sporophytes, regenerating vigorously on 1/2 MS medium. Adjusting sucrose concentration to 2% and supplement to $50mgL^{-1}$ $NaH_2PO_4$ in 1/2MS medium proved to be more efficient for plant regeneration. Various combinations of growth regulators such as kinetin, BA, NAA, and IBA were added to the growing media, and the best sporophyte regeneration was obtained by $1{\mu}M$ kinetin. The BA addition resulted in vigorous proliferation of meristematic tissues, but without differentiation to sporophytes. Three types of culture methods, solid using agar, liquid stationary, and liquid shaking culture, were employed with or without activated charcoal. The addition of 0.1% activated charcoal to modified 1/2MS media (2% sucrose, $50mgL^{-1}$ $NaH_2PO_4$, $1{\mu}M$ kinetin, pH 5.8 and 0.8% agar) yielded highest sporophyte regeneration in liquid shaking culture.

  • Struvite의 최적 생성조건은 NH$_4$-N의 초기농도에 관계없이 모두 NH$_4^+$, PO$_4^{3-}$ 및 Mg$^{2+}$가 등몰비이고 pH 10.5이었다. Struvite 침전반응에서 NH$_4$-N 및 PO$_4$-P 제거율에 미치는 struvite seeding 효과는 습윤상태의 struvite를 seeding할 경우 그 효과가 거의 없었으나 건조된 struvite를 seeding할 경우 NH$_4$-N의 제거율은 증가되지만 PO$_4$-P의 제거율은 감소되는 경향을 나타냈다. 이는 건조과정에서 struvite의 NH$_4$-N가 휘산되어 소실되었기 때문이다. Mg와 P원으로서 struvite의 재이용을 위한 적정 건조온도는 100$^{\circ}C$ 이하였으며 그 이상 온도에서는 struvite가 $NH_4MgPO_4\cdot6H_2O$형에서 MgPO$_4$형으로 상전이점에 따라 struvite seeding에 의한 NE$_4$-N의 제거율이 현저히 감소되었다. 건조된 struvite는 초기 NH$_4$-N의 몰농도 대비 50%를 seeding할 경우 60% 이상의 NH$_4$-N를 제거하였으며, seeding량을 150%로 증가시킬 경우 90% 이상의 NH$_4$-N 제거율을 얻을 수 있었다. 그러나 struvite를 반복 재사용할 경우 재사용 횟수에 비례하여 NH$_4$-N의 제거율은 감소하는 경향을 보여 재사용 횟수가 제한적임을 알 수 있었다.

  • The effect of the alkali, alkali earth metal elements on selective catalytic reduction(SCR) catalyst deactivation behavior were investigated in terms of microstructure, surface area, pore volume and De-NOx test. Poisoned SCR catalyst were manufactured by injection of $K_2CO_3$, $Na_2CO_3$, $Ca(CH_3COO)_2{\cdot}H_2O$, $C_4H_6MgO_4{\cdot}4H_2O$, $H_3PO_4$ solutions in the new SCR catalyst at $350^{\circ}C$ for 6 hours. New and poisoned catalysts surface were similar. But specific surface area, pore volume decrease from Na, Mg, K, Ca, P compared to new SCR catalyst. Especially, Na poisoned catalyst surface area and pore size extremely decreased by $10.20m^2/g$, $0.061cm^2/g$. De-NOx test results of new and poisoned catalysts at $150{\sim}450^{\circ}C$ indicated that alkali metal (K, Na) poisoned SCR catalysts have the lowest De-NOx efficiency, alkali earth metal poisoned SCR catalysts (Ca, Mg) De-NOx efficiency are higher than alkali metal poisoned SCR catalysts. P poisoned SCR catalyst De-NOx efficiency is similar new SCR catalyst. It were considered that physical deactivation of SCR catalyst was affected by SCR catalyst surface area and pore volume change.

Electrochemical synthesis of nanosized hydroxyapatite/graphene composite powder

  • Electrochemical synthesis was employed to prepare a novel hydroxyapatite/graphene (HAP/Gr) composite powder suitable for medical applications as a hard tissue implant (scaffold). The synthesis was performed in a homogeneous dispersion containing Na2H2EDTA·2H2O, NaH2PO4 and CaCl2 with a Ca/EDTA/PO43− concentration ratio of 0.25/0.25/0.15M, along with 0.01 wt% added graphene nanosheets, at a current density of 137 mA cm−2 and pH value of 9.0. The field emission scanning electron microscopy and transmission electron microscopy observations of the composite HAP/Gr powder indicated that nanosized hydroxyapatite particles were uniformly placed in the graphene overlay. Raman spectroscopy, Fourier transform infrared spectroscopy and X-ray diffraction confirmed graphene incorporation in the HAP/Gr powder. The electrochemically prepared HAP/Gr composite powder exhibited slight antibacterial effect against the growth of the bacterial strain Staphylococcus aureus.

  • This study was performed to compare the anticarious effect of the different fluoride adsorbed(Naf, $NH_4F, Na_2PO_3F, SnF_2, TiF_4$) on synthetic hydroxyapatite and enamel. The amount of fluoride adsorbed in synthetic hydroxyapatite under various concentrations and pH of these fluoride solutions was measured by specific electrode. Enamel samples treated with 5 kinds of 1,000ppm fluoride solutions for 10 minutes were evaluated for fluoride uptake and enamel soubility. The results were as follows. 1. The adsorption of fluoride on synthetic hydroxyapatite increased gradually by the concentration of the fluoride solution, In 1,000ppm fluoride solution, the adsorption of fluoride on synthetic hydroxyapatite treated with NH4F and NaF solutions at pH 4.0 was relatively higher than that of other fluoride solutions. In NH4F and NaF solutions, the adsorption of fluoride on powdered enamel was higher at pH 4.0 solution than at pH 7.0 solution. 2. Fluoride uptake from NH4F solution was relatively high. But that from $Na_2PO_3F$ solution was lower than those from other fluoride solutions. 3. Fluoride solutions were significantly effective on enhancing acid resistance. $NH_4F$ solution was relatively more effective than others on enhancing acid resistance. 4. $SnF_2 and TiF_4$ solutions had the same effect on fluoride adsorption, fluoride uptake, and enamel solubility.

Small Metal Ion Effect on The Harvest of Perilla Leaves in Aquiculture (수경재배 들깻잎의 수확량에 미치는 미량금속 이온 효과)

  • The perillas were cultivated to investigate for the small metal ion effect on the harvest of perilla loaves in aquicul-ture system in the constant flow rate. The perillas were und-ergrown at the condition of low concentration of KNO$_3$and NH$_4$ H$_2$ PO$_4$as below 270ppm and 152ppm respectively. The high concentration of the metal and non-metal ca-tions of $Ca^{2+}$, $K^+,NH^{4+},Mn^{2+},Zn^{2+},Cu^{2+},Na^+,Mo^{+6}$ are influenced to the growth of plant length of perillas at the earlier time. The low concentration of the metal cations of $Ca^{2+}, K^+, NH^{4+}, NO_3^-, H_2PO_4^-$ are influenced to lower growth of perillas. The concentration of the cations of $Ca^{2+}, K^+$and $NH_4^+$ and the anions of $NO_3^-$and $H_2PO_4^-$ are affected the growth of leaf length and width of leaf of perillas. The spectoscopic analytical results showed that the perillas were growing rapidly in the period of 6 days from June 7 to June 12 by high amount of $Mg^{2+}$ ion with accumulation inside perillas. The crude protein, the crude fat and the hydrocarbon are accumulated in the leaves of perillas by binding the inorganic with amino acids to provide the nutritions needed for growth of perillas.

Optimization of Major Culture Elements on Growth and Shikonin Production in the Lithospermum erythrorhizon Hairy Root Culture

  • The effects of basal media, carbon, nitrogen, phosphate and some major macro elements on growth and shikonin production in Lithospermum erythrorhizon hairy root culture were studied. Among examined media, growth of hairy root cultured in B5 liquid medium was rapid, whereas shikonin production was high in MS liquid medium. Under B5 basal medium, sucrose concentration for optimal growth and shikonin production was 9% and 4% respectively. The growth and shikonin production on pH changes in B5 medium resulted little effect in pH 5.8 to pH 8.8 ranges, whereas growth was decreased dramatically in both above 8.8 and under 5.8. Nitrogen source and concentration effected on the growth and shikonin production. The highest growth rate was in B5 medium (50 mM $KNO_3$ and 1 mM $NaH_2PO_4)$, whereas the highest shikonin production was in the condition supplemented with 5 mM $KNO_3$ and 10 mM $NaH_2PO_4$.
