Local Difference of Na-Pump Activity in the Rabbit Sinoatrial Node (토끼 동방결절 부위에 따른 Na-Pump활동도의 차이에 관한 연구)

  • Electrophysiological difference of the central and peripheral area of the sinoatrial node in the rabbit was studied by glass microelectrode technique. Effects of $K^+,\;Na^+,\;Cs^+,$ adrenaline and ouabain on the action potential of the two areas were investigated, and transient hyperpolarization ($K^+-induced$ hyperpolarization) which developed following readmission of potassium after having pre-treated with $K^+-free$ Tyrode solution for 10 minutes was analyzed. The results obtained were as follows ; 1) The frequency of the spontaneous action potential recorded in the periphery of the SA node was faster than the central area. Reduction by $Cs^+$ and increase by O mM $K^+$, $10^{-6}M$ adrenaline and $10^{-6}M$ ouabain in the frequency of action potential were noticed more prominently in the peripheral than the central area. On the contrary, the frequency in the central area was more decreased than the Peripheral area by 13 mH $K^+$ and 1 mM $Co^{2+}$. 2) The amplitude of the K+_induced hyperpolarization was very small in the central area but large in the peripheral area. Transient hyperpolarization was abolished by ouabain and low sodium, and decreased by cooling the tissue $(17^{\circ}C)$. 3) By changing the concentration of $Ca^{2+}$ in the perfusate, the amplitude and the rate of transient hyperpolarization were increased in the high $Ca^{2+}$ concentration. It could be concluded that the central area of the SA node is less susceptible to the inhibition of Na-Pump and more susceptible to Ca-blocker and high concentration of $K^+$. The Na-Pump activity of the central area measured by means of transient hyperpolarization is found to be much less active than that of the peripheral area.

  • Glibenclamide, a sulfonylurea derivative, has been used in tile treatment of type II diabetes mellitus. Recent studies provided evidence that glibenclamide, in addition to blocking ATP-sensitive $K^{+}$ channels, also affected Na$^{+}$-K$^{+}$ pumps and L-type $Ca^{2+}$ channels in noncardiac cells. The effect of glibenclamide on the cardiac muscle is not clearly known. In the present study, the effects of glibenclamide on intracellular Na$^{+}$ concentration ([Na$^{+}$]$_{i}$ ), twitch tension, $Ca^{2+}$ transient, and membrane potential were investigated in isolated guinea-pig ventricular myocytes. Glibenclamide at concentration of 200 $\mu$M increased [Na$^{+}$]$_{i}$ by 3.9$\pm$0.4 mM (mean $\pm$ SE, n=12), decreased twitch tension by 36.1 $\pm$ 4.0% (mean $\pm$ SE, n=8), reduced $Ca^{2+}$ transient by 24.4$\pm$5.1% (mean $\pm$ SE, n=3), slightly depolarized diastolic membrane potential, and did not change action potential duration. To determine whether inhibitions of Na$^{+}$-K$^{+}$ pumps and L-type $Ca^{2+}$ channels are responsible for the increase of [Na$^{+}$]$_{i}$ and the decrease of twitch tension, we tested effects of glibenclamide on Na$^{+}$-K$^{+}$ pump current and L-type $Ca^{2+}$ current. Glibenclamide decreased Na$^{+}$-K$^{+}$ pump current and L-type $Ca^{2+}$ current in a concentration-dependent manner.t in a concentration-dependent manner.

Effects of External $Ca^{2+}$ ana the Inhibition of Na-pump on the Vanadate-induced Contraction in the Isolated Human and Rat Uterine Smooth Muscle (사람 및 흰쥐의 자궁근에서 Vanadate에 의한 수축에 미치는 외부 Calcium 및 Na-pump억제의 영향)

  • The effects of external $Ca^{2+}$ ana the inhibition of Na-pump on vanadate-induced contraction in isolated human and rat uterine smooth muscle were studied and the following results were observed. 1) Vanadate induced contraction in rat uterine muscle and showed maximal contraction at concentration of $5{\times}10^{-4}$M, and the contractile response to vanadate was more sensitive in human than rat uterine muscle. 2) Vanadate-induced contraction was not completely inhibited by $Ca^{2+}$ removal from PSS and the response to $Ca^{2+}$ removal was more sensitive in human than rat uterine muscle. 3) Vanadate-induced contraction decreased with increasing concentration of verapamil, but even in the presence of $3{\times}10^{-5}M$ verapamil which inhibited 100 K-induced contraction completely. about 40% of maximal contraction remained, and its amplitude was similar to that of contraction in $Ca^{2+}$-free solution. 4) Vanadate-induced contraction was increased by the inhibition of Na-pump and this increase also could be observed in the presence of $3{\times}10^{-5}M$ verapamil. 5) After pretreatment with $Ca^{2+}$-free PSS containing ouabain Vanadate-induced contraction was not increased, but the contractile response of these tissues to the addition of external $Ca^{2+}$ was remarkably increased in the presence of vanadate. 6) $3{\times}10^{-5}$M verapamil inhibited vanadate-induced $Ca^{45}$ influx completely, but after pretreatment with ouabain vanadate could induce remarkable $Ca^{45}$ influx even in the presence of verapmil. 7) With increasing the time of pretreatment with ouabain or $K^+$-free solution, the degree of increase in contraction by vanadate was more remarkable. 8) $10^{-4}M$ papaverine stowed a considerable inhibition of the increase in the vanadate-induced contraction by pretreatment with ouabain. 9) Acetylcholine-induced contraction increased with lengthening the duration of Na-pump inhibition even in the presence of verapamil. Considering above results it seems that the uterine muscle of human is more sensitive to vanadate than that of rat, and both internal and external $Ca^{2+}$ is utilized in vanadate·induced contraction. In the case of Na-pump inhibition several smooth muscle contracting agents seems to induce $Ca^{2+}$ influx which is not inhibited by verapamil. This $Ca^{2+}$ influx seems to be inhibited by papaverine and to be associated with membrane potential, although its precise characteristics is not certain.

  • 세포의 흥분성과 막전위의 조절에 있어서 $K^{+}$ channel의 역할이 크다는 사실이 인정 됨으로서 (Rudy, 1988) $K^{+}$ channel 개방 약물의 약리학적 연구와 임상적 응용에 대한 관심은 높아졌다. Cromakalim, nicorandil 및 pinacidil등이 혈관 평활근을 특히 예민하게 이완시킨다. 작용기전으로서는 세포의 원형질 막을 통한 $K^{+}$ 전도의 항진과 $K^{+}$ outward current의 증가가 막 과분극을 일으킨다. 이러한 결과는 막흥분에 의한 voltage-dependent $Ca^{++}$ channel을 닫게하고 세포내 free $Ca^{++}$의 농도를 감소시켜 혈관의 흥분성과 수축력의 감소를 야기하여 근이완을 야기한다. 한편, 평활근 세포막의 $Na^{+}$-$K^{+}$ ATPase도 활성화하면 electrogenic pump를 가동하여 막 과분극을 일으키고 막 흥분성을 저하시킨다. $Na^{+}$-$K^{+}$ pump는 세포 바깥의 $K^{+}$과 세포안의 $Na^{+}$농도에 의하여 활성화한다.

Moderate Elevation of Extracellular $K^+$ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries: Role of Na Pump and Ba-Sensitive Process

  • Cerebral blood vessels relax when extracellular $K^+$ concentrations $([K^+])_e$ are elevated moderately $(2{\sim}15$ mM, $K^+-induced$ vasorelaxation). We have therefore studied the underlying mechanism for this $K^+-induced$ vasorelaxation in the isolated middle cerebral arteries (MCAs). The effects of ouabain and $Ba^{2+}\;on\;K^+-induced$ vasorelaxation were examined to determine the role of sodium pump and/or Ba-sensitive process (possibly, inward rectifier K current) in the mechanism. Mulvany myograph was used to study 24 rats, 18 rabbits, and 10 humans MCAs $(216{\pm}3\;{\mu}m,\;347{\pm}7\;{\mu}m,\;and\;597{\pm}39\;{\mu}m$ in diameter when stretched to a tension equivalent to 55 mmHg). High $K^+$ (125 mM) and $PGF_{2{\alpha}}\;(1{\sim}10\;{\mu}M)$ induced concentration-dependent contractions in all 3 species, while histamine $(10{\sim}50\;{\mu}M)$ evoked contraction only in the rabbits and induced relaxation in the rats and humans. Addition of $K^+\;(2{\sim}10\;{\mu}M)$ to the control solution induced vasorelaxations. These effects were inhibited by the pretreatment with both ouabain $(10\;{\mu}M)$ and $Ba^{2+}\;(0.1{\sim}0.3\;mM)$ in the rat, but only with ouabain $(10\;{\mu}M)$ in the rabbit and human. These results suggest that $K^+-induced$ vasorelaxation occurs via the stimulation of electrogenic Na pump in the rabbit and human MCAs, while in the rat MCAs via the activation of both Na pump and Ba-sensitive process.

The Influence of NaCl and Carbonylcyanide-m-Chlorophenylhydrazone on the Production of Extracellular Proteases in a Marine Vibrio Strain

  • In general, the salinity of the ocean is close to 3.5% and marine vibrios possess the respiratory chain-linked Na$\^$+/ pump. The influence of sodium chloride and the proton conductor carbonylcyanide m-chlo-rophenylhydrazone (CCCP) on the production of extracellular proteases in a marine Vibrio strain was examined. At the concentration of 0.5 M, sodium chloride minimally inhibited the activity of extra-cellular proteases by approximately 16%, whereas at the same concentration, the producton of extra-cellular proteases was severely inhibited. On the other hand, the production of extracellular proteases was completely inhibited by the addition of 2 ${\mu}$M CCCP at pH 8.5, where the respiratory chain-linked Na$\^$+/ pump functions.

  • Plasma membrane Na$^{+}$-K$^{+}$ ATPase (pump) is an essential component to maintain asymmetrical ion distribution across cell membrane. The Na$^{+}$-K$^{+}$ ATPase was discovered by Jens C. Skou in 1957 and since then physiological and biochemical properties of the enzyme have been extensively studied. Jens C. Skou was awarded the 1997 Nobel Prize in chemistry for his discovery of the Na $^{+}$ - $K^{+}$ ATPase.(omitted)

Effects of Insulin and IGFS on Growth and Functional Differentiation in Primary Cultured Rabbit Kidney Proximal Tubule Cells -Growth and membrane transport-

  • The purpose of this study was to compare effects of insulin and IGFs on growth, apical membrane enzyme activities and membrane transport systems of primary cultured rabbit kidney proximal tubule cells. Results were as follows: 1. Insulin and IGF-I produced significant growth stimulatory effects at $5{\times}10^{-10}M.\;IGF-II(5×10^{-10}\;M)$ did not stimulate significant cell growth. 2. Insulin stimulated the phosphorylation of a 97 KD protein. It was difficult to determine whether this band represents insulin and/or the IGF-I receptor. 3. The activities of apical membrane enzymes (alkaline phosphatase, leucine aminopeptidase, and ${\gamma}-glutamyl \;transpeptidase)$ were observed to be diminished after the cells were placed in the culture environment. 4. The uptake of ${\alpha}-MG,$ Pi and Na was significantly increased in cells incubated with insulin or IGF-I, IGF-II had no effect on the uptake of these substrates. 5. Na-pump activity, as assayed by Rb uptake, was significantly increased in cells treated with insulin or IGFs. In conclusion, insulin and IGF-I exert stimulatory effects on growth and membrane transporter(glucose, Na, Pi, and Na-pump) activities in primary cultured rabbit kidney proximal tubule cells. IGF-II had no effect on cell growth and membrane transporter(glucose, Na and Pi) activities.

  • Ascending aortic aneurysms are usually treated with graft replacement under cardiopulmonary bypass. However, if a candidate for off-pump coronary artery bypass grafting has an enlarged ascending aorta, surgeons may consider wrapping it without cardiopulmonary bypass. Here, we report a 78-year-old female who underwent successful wrapping of the ascending aorta concomitant with off-pump coronary artery bypass grafting, using a new wrapping technique that involves multiple bootstraps.
