The objective of this research was to investigate the antioxidant and antimicrobial activities of muscadine grape extracts. Three different cultivars of muscadine grapes including Higgings, Jumbo, and Noble were selected. The skin/pulp and seed parts of three selected muscadine grape cultivars were used for extraction. The total phenolic contents of muscadine grape extracts were expressed as gallic acid equivalents (GAE). The antioxidant activity of muscadine grape extracts were determined by scavenging activity of diphenylpicrylhydrazyl (DPPH) radical and expressed as effective concentration ($EC_{50}$), which represented the concentration of the extract exhibiting 50% DPPH radical scavenging. The antimicrobial activity against E. coli K12 was determined and expressed as the minimum inhibition concentration (MIC). The seed extracts exhibited greater total phenolic contents than the skin/pulp extracts, ranging from 231.24 to 294.81 mg/mL GAE. The seed extracts exhibited greater antioxidant activities than the skin/pulp extracts ($EC_{50}$ of Higgins seed extract=0.026 mg/mL). However, the skin/pulp extracts exhibited greater antimicrobial activities than the seed extracts, exhibiting the minimum inhibition concentration (MIC) in Higgins skin/pulp extract (MIC=4.0 mg/mL). This research indicated that the seed part and skin/pulp parts of the muscadine grapes possessed antioxidant activity and antimicrobial activity, respectively. Therefore, it was concluded that muscadine grapes possess the potential to be utilized as functional foods or nutraceuticals.

This study was carried out to investigate the anti-microbial of various herbal extracts on six types of bacteria related to skin diseases and effects of Saussurea lappa extract on dermatitis and acne. The results are followed ; 1. In the experiment on P. acnes using paper disk methods, Eugenia caryophyllata extract made 19mm clear zone, Saussurea lappa extract and Morus alba extract made 13mm and MIC of Morus alba extract and Eugenia caryophyllata extract was below 0.01$\%$. 2. In the experiment on S. aureus, extract of Eugenia caryophyllata, Poncirus trifoliata and Rubus coreanus made 9-10mm clear zone. 3. In the experiment on C. xerosis, Sesamum indicum extract made 16mm clear zone and MIC of that was 0.3$\%$. 4. In the experiment on C. albicans, extract of Cinnamomum cassia, Eugenia caryophyllata and Asparagus cochinchinensis made 9-10mm clear zone. 5. In the experiment on P. ovale, Cinnamomum cassia extract made 25mm clear one, MIC of that was 0.05$\%$. 6. In the experiment on T. mentagrophytes, Cinnamomum cassia extract made 26mm clear zone, extract of Eugenia caryophyllata, Rhizoma kaempferiae, Piper longum, Saussurea lappa and Zingiber officinale made 18-22mm clear zone, MIC of all extracts was below 0.02$\%$. 7. Saussurea lappa extract inhibited 85$\%$ and 28$\%$ of NO production at 50㎍/㎖ and 5 ㎍/㎖ 8. Saussurea lappa extract inhibited activity of COX Ⅱ over 50$\%$ at 10㎍/㎖ 9. Saussurea lappa extract had not radical scavenging activity and Saussurea lappa extract didn't inhibit manifestation of IL-1${\beta}$, IL-6 and TNF-${\alpha}$, and activity of 50- reductase I.
CRAMP, a 37-amino acid cationic antimicrobial peptide was recently deduced from the cDNA cloned from mouse femoral marrow RNA. In order to investigate the structure-activity relationship and functional region of CRAMP, CRAMP and its 18-mer overlapping peptides were synthesized by the solid phase method. CRAMP showed broad spectrum antibacterial activity against both Gram-positive and Gram-negative bacterial strains (MIC: 3.125-6.25 μM) but had no hemolytic activity until 50 μM. CRAMP was found to have a potent anticancer activity (IC50: 12-23 μM) against two human small cell lung cancer cell lines. Furthermore, CRAMP was found to display faster bactericidal rate in B. subtilis rather than E. coli in the kinetics of bacterial killing. Among 18-meric overlapping fragment peptides, only CRAMP (16-33) displayed potent antibacterial activity (MIC: 12.5-50 μM) against several bacteria with no hemolytic activity. Circular dichroism (CD) spectra anal-ysis indicated that CRAMP and its analogues will form the amphipathic α-helical conformation in the cell membranes similar to other antimicrobial peptides, such as cecropins and magainins.
To investigate the antimicrobial activities and safety of natural naringin, it was isolated with methanol from peels of Citri fructus. Its hydrolysate, naringenin was obtained by hydrolysis of naringin. In the antimicrobial activities of two components against eleven species of bacteria and eleven species of Fungi were examined by serial dilution method. Its result appeared to the minimal inhibitory concentration(MIC) and the antimicrobial activities of naringin and naringenin were compared. Naringenin showed considerably high order of activities against bacteria. There were no effect against Fungi $(MIC>100{\mu}g/ml)$. In the safety tests of naringin, examined for 50% lethal dose, Blood clinical chemical tests and organ tissue tests. The results showed that 50% lethal does in mice was 1,650 mg/kg. The experiments of administration in rats showed that there were no changes in blood clinical chemical future and organ tissue as control.

A natural habitat bacterium, Enterobacter cloaceae K41 was isolated from fresh water plant root and identified. This strain was used to investigate heavy metal resistance. The optimal growth conditions of the bacterium were LB medium containing$1\%$ yeast extract, $1\%$ lactose, $1\%$ NaCl, pH 7.0, at $37^{\circ}C$, and for 24 hours on a shaker. The minimal inhibitory concentration (MIC) of heavy metals against E. cloaceae KCTC2519 and E. cloaceae K41 was compared. The MIC of E. cloaceae K41 was 150 ppm in Cu, 50 ppm in Cd whereas that of the standard strain was 50 ppm in Cu but no growth was observed either Cd or two mixed heavy metal solution. The presence of plasmid was cleared from the isolated strain whereas no possession from the standard strain. The plasmid from E. cloaceae K41 was transformed into E. coli $DH5{\alpha}$. The MIC of transformed strain increased resistance 7 times in Cu and 6 times in Cd by insertion of this plasmid. The metal adsorption of the transformant was increased 1.3 times in Cu and 1.5 times in Cd indicating the plasmid was responsible for heavy metal resistance.
The actinomycete strain KH-6l4 possessed strong antifungal activity, especially antagonistic to the rice blast fungus, Pyricularia oryzae. Diaminopimelic acid (DAP) type and morphological and physiological characteristics, examined by scanning electron microscopy (SEM), indicated that KH-614 belonged to the genus Streptomyces. Antifungal agent produced by this strain was found to be most active, when the strain was cultured in the presence of glucose, polypeptone, and yeast extract (PY) medium for 6 days at $27^{\circ}C$. Based on the spectral report data, MS and NMR, the antifungal agent was identified as cyclo(L-leucyl-L-prolyl). According to the antimicrobial activity test measured by minimal inhibitory concentration (MIC), the cyclo(1eu-pro) exhibited the activity against Candida albicans IAM 4905, Mucor ramannianus IAM6218, Rhizoctonia solani IFO 6218, Aspergilus fumigatus ATCC 42202, Glomerella cingulata IFO 9767, Trichophton mentagrophytes ATCC 18749, and Trichophyton rubrum ATCC 44766, the order of MIC values were 50, 12.5, 5, 50, 25, 5, $5\;\mu\textrm{g}/ml$, respectively. Specifically, cyclo(1eu-pro) was one of the most effective elements against Pyricularia oryzae IFO 5994 with the MIC value of $2.5\;\mu\textrm{g}/ml$, thus indicating that cyclo(leu-pro) is a potential antifungal agent.
The aggregation behavior of nystatin (NYS) in the presence of pluronic F127, triblock copolymer of poly (ethylene oxide) (PEO) and poly (propylene oxide) (PPO), was measured and correlated with hemolytic activity. Antifungal activity was also studied using Saccharomyces cerevisiae as a model strain. The critical aggregation concentrations (CAC) of the drug were 50.1, 108.0, 134.2, 154.3, and $217.9\;{\mu}M$ at 0.1%, 0.5%, 1.0%, 1.5%, and 2.0% pluronic F127 solution, respectively. The levels of NYS required to start lysis of erythrocytes were about 80, 100, 125, 150, and $200\;{\mu}M$ at 0.1%, 0.5%, 1.0%, 1.5%, and 2.0% pluronic F127 solution, respectively. It was $50\;{\mu}M$ in the absence of the polymer. Minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) of NYS-pluronic F127 lyophilizate were same at $3\;{\mu}g/ml$, while MIC and MFC of pure NYS are $3\;{\mu}g/ml$ and $12\;{\mu}g/ml$, respectively. By modulating the aggregation behavior of NYS, pluronic F127 was able to reduce the toxicity of the drug without compromising the MIC and MFC.

An antibacterial component was isolated from the roots of evening primrose(Oenothera odorata Jacquin). The component was purified by repeated preparative TLC of the chloroform-extract with solvent system of toluene(7):ethyl acetate(4):formic acid(O.8). NMR spectrum indicated that it is a steroid compound containing radical(-C=C-C=0) and aromatic structures. Its antibacterial activity against strains of the genus Streptococcus was relatively high, the MIC value was 50 acccording to the Hoechst Manual-345, as compared with the other genera such as Staphylococcus and Escherichia.
Extensive acquirement of drug resistance to traditional antibacterial agents poses a serious problem to eel aquaculturists. To collect the basic information for new drug development in the future, we assessed the in vitro antibacterial efficacy of 14 new quinolones with 75 isolates of Edwardsiella tarda from local aquaculture tanks of Anguilla japonica. Of all tested quinolones under development or marketed for human use, DU-6859 was most potent with its $MIC_{50}$ value of $0.05{\mu}g$/ml in broth microdilution assay. The drugs whose $MIC_{50}$ values ranged from 0.2 to $0.78{\mu}g$/ml were T-3762, Bay-y3118, ciprofloxacin, norfloxacin, ofloxcin and tosufloxacin. The weakest group of drugs, with their $MIC_{50}$ being 1.56-$3.13{\mu}g$/ml, were difloxacin, sparfloxacin, fleroxacin, Q-35, amifloxacin, lomefloxacin and enoxacin. The number of resistant strains, when arbitrarily defined with their MICs of $\geq6.25{\mu}g$/ml, was : 3 to T-3762, 3 to Bay-y3118, 44 to difloxacin, 16 to sparfloxacin, 13 to ciprofloxacin, 19 to fleroxacin, 36 to Q-35). 31 to amifloxacin, 5 to norfloxacin, 13 to ofloxacin, 31 to lomefloxacin, 41 to enoxacin, 12 to tosufloxacin and 0% to DU-6859, respectively. This information can be taken into consideration for the future development of fisheries antibacterial quinolones.
Ten bacteriocin-producing Enterococcus faecium strains with urease activity ($1.10-6.2nkat.mL^{-1}$) were isolated from the rumen of 2-8 weeks old calves. All strains were resistant aginst disodium arsenate at a minimal inhibition concentration - MIC $5g.L^{-1}$ and mercury chloride ($MIC=10-20mg.L^{-1}$). Eight strains were resistant against silver nitrate ($MIC=40-50mg.L^{-1}$) and three against antibiotics used. The resistance against six antibiotics was found in A23 strain. Values of adherence index ranged from 5.02 to 20.4 enterococci adhered per one epithelial cell of rumen wall. All isolates produced bacteriocins which inhibited the growth at least of one of five indicator organisms. The EF1 strain with a good affinity to the epithelial cell ($15.2{\pm}1.2$) produced bacteriocin substance with antimicrobial activity against grampositive and gramnegative indicator bacteria.
