Biological Functions of the COOH-Terminal Amino Acids of the $\alpha$-Subunit of Tethered Equine Chorionic Gonadotropin

  • Glycoprotein hormones have a common $\alpha$-subunit that is involved in the signaling pathway together with G protein, adenylcyclase and cAMP induction; however, it is an unclear how this common structure is related to hormonal action. To determine the biological functions of the COOH-terminal amino acids in the $\alpha$-subunit of these glycoprotein hormones, a tethered-molecule was constructed by fusing the $NH_2$-terminus of the $\alpha$-subunit to the COOH-terminus of the $\beta$-subunit of equine chorionic gonadotropin (eCG). The following deletion mutants were created by PCR; Ile was inserted at position 96 to form ${\Delta}96$, Lys was substituted at position 95 to form ${\Delta}95$, His was inserted at position 93 to form ${\Delta}93$ and Tyr was substituted at position 87 to form ${\Delta}87$. Each mutant was transfected into CHO-K1 cells. Tethered-wt eCG, and ${\Delta}96$, ${\Delta}95$, and ${\Delta}93$ mutants were efficiently secreted into the medium but the ${\Delta}87$ mutant was not secreted. Interestingly, the RT-PCR, real-time PCR, and northern blot analyses confirmed that the RNA was transcribed in the ${\Delta}87$ mutant. However, the ${\Delta}87$ mutant protein was not detected in the medium or the intracellular fraction of the cell lysates. The LH- and FSH-like activities of the recombinant proteins were assayed in terms of cAMP production using rat LH/CG and rat FSH receptors. The metabolic clearance rate (MCR) was determined by injecting rec-eCG (2 IU) into the tail vein. The ${\Delta}95$ and ${\Delta}93$ mutants were completely inactive in both the LH- and FSH-like activity assays. The ${\Delta}96$ mutant showed slight activity in the LH-like activity assay. In comparison to the wild type, the activity of the ${\Delta}96$ mutant in the FSH-like activity assay was the highest among all the mutants. The MCR assay in which rec-eCG was injected showed a peak at 10 min in all the treatment groups, which disappeared 4 h after injection. These results imply a direct interaction between the receptor and the COOH-terminal region of the a-subunit. The data also reveal a significant difference in the mechanism by which the eCG hormone interacts with the rLH and rFSH receptors. The COOH-terminal region of the $\alpha$-subunit is very important for the secretion and functioning of this hormone.

  • These studies were carried out to find the proper conditions for in vitro maturation and fertilization of bovine follicular oocytes and culture methods capable of further developing early embryos. For these objectives, the cleavage rate of oocytes matured and fertilized in vitro was investigated under medium supplemented with hormones and estrous cow serum and season of oocytes collection as well as different cumulus cell stage before insemination. Finally, 2~8 cell embryos were cultured in in vitro and in vitro culture system to investigate developmental capacity into morula. 1. Cleavage rate of oocytes matured in vitro was 27%(20/73) for A(LH+FSH+estradil-17$\beta$+10% FBS), 38%(27/71) for B(LH+10% ECS) and 27%(15/56) for C(10% ECS), respectively. Supplement B showed more higher rate and 4~8 cell embryos were also obtained much more in this group(67%, 18/27). In vitro maturation rate of follicular oocytes cultured in TCM 199 supplemented withLH and 10% ECS was 88%(75/85). 2. Cleavage rate(15%, 10/65) of oocytes collected in summer was significantly lower than in fall(47%, 42/89). 3. Cleavage rate(15%, 10/65) of oocytes with partially removed cumulus cell before insemination was more higher than that(44%, 27/62) of oocytes with intact cumulus cell. 4. The frequency of development from early cleaved embryos into morula was 6%(4/65), 12%(4/33) for co-culture of cumulus cell monolayer and bovine oviduct epithelial cell monolayer, respectively and 25%(6/25) in ligated rabbit oviduct.

사람 LH 호르몬유전자를 도입한 배아주세포에 의한 카이미라 생쥐 생산 (The Production of Chimeric Mice by Embryonic Stem Cell Carrying Human Luteinizing Hormone Gene)

  • 최근 의약적으로 유용한 단백질을 대량 생산키 위한 실현 가능한 방법이 유전자변환 가축의 이용과 관련되어 발전되어 왔다. 이러한 유전자 변환동물은 이종의 단백질을 유즙속으로 분비시키는 생체반응기로서 이용되고 있다. 이러한 전략적 목적을 위해 현재 유전자 변환동물의 생산을 위한 이용에 있어 여러 가지 방법들이 보고되고 있다. 그러나 ES 세포의 사용이 이러한 방법들 사이에서 가장 실질적인 것으로 추정되고 있다. 본 실험에서는 유전자 구축을 위해 사람 황체 호르몬(human luteinizing hormone; hLH)의 전사를 유도하기 위해 각각 2.2 및 0.5 kb의 토끼 $\beta$-casein pronoter 단편을 이용하여 생쥐의 유선에 hLH를 발현시키도록 조절하고 발현이 thynidine kinase(TK) pronoter에 의해 좌우되는 neo 유전자를 selectable marker로서 plasnid속에 삽입하였다. 그 결과 생긴 구축 유전자는 각각 pCas 2.2와 pCas 0.5로 명명하였다. 구축된 유전자로 2$\times$107의 TT-2 ES세포를 170V, 550$\mu$F로 100$\mu$g의 선상 plasmid에 의해 electroporation 시켰다. 감염된 colony들은 250$\mu$g/$m\ell$ G418을 함유하는 ESM 배양액에서 선별 7일 이후에 회수하여 성공적으로 감염된 ES세포는 PCR 및 Southern blot에 의해 확인되었고 그들 중 나머지는 trypsin 처리 후 각각 미세조작과 공배양 기술을 사용하여 ICR 생쥐의 8세포기 수정란 속에 도입하였다. 결국 24시간 동안 37$^{\circ}C$, 5% $CO_2$에서 배양된 배반포를 chimera의 생산을 위해 위임신 유기된 G418 선발처리 이후 400 및 275개의 ES 세포 colony가 생존하였으며, 3개의 ES 세포으 colony 의 genome 속에 임의적으로 plamid가 삽입된 것을 Southern blot에 의해 확인되었다. 총 13 chimera 생쥐가 3 colony로부터 생산되었으나 germ-line chimera는 현재 조사중이다. chimera 생산빈도는 공배양 기술보다 주입방법에서 현저히 높았다.

돼지에 있어서 분만전후(分娩前後)의 혈청(血淸) 성(性) Hormone 수준(水準)에 관(關)한 연구(硏究) (Studies on the Sex Hormone Level is Serum of Puerperal Sow)

  • 돼지에 있어서 분만전후(分娩前後)의 혈청중(血淸中) peptide 및 steroid hormone 수준(水準)의 변화(變化)를 구명(究明)하기 위(爲)하여 8두(頭)의 임신돈(妊娠豚)을 대상(對象)으로 분만전(分娩前) 20일(日)부터 분만후(分娩後) 20일(日)까지 혈청중(血淸中)의 FSH, LH, prolactin, estradiol-$17{\beta}$, progesterone 및 cortisol의 농도변화(濃度變化)를 radioimmunoassay 방법(方法)에 의(依)하여 측정(測定)하였다. 혈청(血淸) FSH의 농도(濃度)는 분만전후(分娩前後)에 큰 변화(變化)가 없었으며, $8.1{\pm}1.8mIU/ml$에서 $9.0{\pm}2.6mIU/ml$의 사이에 있었다. LH농도(濃度)는 분만(分娩) 20일전(日前)에 $2.6{\pm}0.3mIU/ml$이던것이 분만일(分娩日)에는 $3.9{\pm}1.1mIU/ml$로 증가(增加)하였으며, 분만후(分娩後) 2일(日)에는 $3.2{\pm}0.9mIU/ml$로 감소(減少)되었고, 그 후(後)로는 거의 비슷한 수준(水準)을 유지(維持)하였다. Prolactin의 농도(濃度)는 분만일(分娩日)에 $68.5{\pm}9.5ng/ml$로 최고수준(最高水準)을 나타냈으며, 분만당일(分娩當日) 전후(前後)에 비교적(比較的) 높은 수준(水準)을 나타냈다. Estradiol-$17{\beta}$는 분만전(分娩前) 6일(日)에 $205.0{\pm}29.5pg/ml$로 증가(增加)하였고, 분만일(分娩日)에는 $425.0{\pm}35.6pg/ml$로 급증(急增)하였으나 분만후(分娩後) 2일(日)에는 $59.5{\pm}7.8pg/ml$로 다시 감소(感少)했다. Progesterone 농도(濃度)는 분만전(分娩前) 20일(日)부터 6일(日)까지는 $18.4{\pm}1.6{\sim}20.2{\pm}2.1ng/ml$사이에 있었으나, 분만전(分娩前) 2일(日)부터 감소(減少)하기 시작(始作)하여 분만일(分娩日)에는 $2.6{\pm}0.8ng/ml$까지 감소(減少)하였고, 분만후(分娩後) 2일(日)에는 $0.9{\pm}0.3ng/ml$에 도달(到達) 하였으며, 그 후(後)로는 거의 같은 수준(水準)을 유지(維持)하였다. Cortisol은 분만일(分娩日)에 $86.5{\pm}10.5ng/ml$로 최고수준(最高水準)을 나타냈다.

Improvement of Photoheterotrophic Hydrogen Production of Rhodobacter sphaeroides by Removal of B800-850 Light-Harvesting Complex

  • The photoheterotrophic $H_2$ production of Rhodobacter sphaeroides was significantly increased through disruption of the genes coding for uptake hydrogenase and poly-${\beta}$-hydroxybutyrate (PHB) synthase (Lee et al., Appl. Microbiol. Biotechnol. 60: 147-153, 2002). In this work, we further removed the B800-850 light-harvesting (LH) complex from the strain and found an increase in $H_2$ production at the light-saturating cell growth (${\ge}10$ Watts $[W]/m^2$). Neither the mutant nor the wild-type produced more $H_2$ at the brighter light. Accordingly, light does not appear to be limited for the $H_2$ production by the presence of B800-850. However, increase in the level of the spectral complexes resulted in decrease of $H_2$ production. Thus, although the B875 is essential for light harvesting, the consumption of cellular energy for the synthesis of B800-850 and the surplus LH complexes may reduce the energy flow into the $H_2$ production of R. sphaeroides.

Cacao Bean Husk 추출물로부터 Glucosyltransferase 저해 활성 물질의 분리 및 구조 동정 (Purification and Structure Determination of the GTase Inhibitor from Cacao Bean Husk Extract)

  • 충치 예방 물질의 탐색 연구 일환으로, Cacao bean husk(CBH) 추출물로부터 얻어진, gluco-syltransferase(Gtase) 저해 활성 분획을 분리하였 다. 이를 다시 MCI-gel CHP-20와 Sephadex LH 20의 column chromatography법으로 분리, 정제하였으며, 이로부터 얻어진 2개의 GTase 활성 저해 물질에 대한 구조 해석을 행하였다. 분리.정제된 2개의 GTase 저해 활성 물질은 anisaldehyde-$H_2SO_4$ 및 FeCb에서 각각 갈색 및 청색 반응을 하며, 또 TLC상에서 dimer의 flavan-3-ol $R_r$값을 나타내는 단일 물질이었다. NMR(PMR, C CMR) 빛 mass spectrum으로 두 정제 분획에 대 한 화학 구조를 분석한 결과, G Tase 저해능을 나타 내는 단물질은 (-)-epicatechin-($\beta\rightarrow8$)-catechin 으로 결합된 procyanidin B-1 ($C_{30}H_{26}O_{12}$) 및 cate chin의 2량체인 procyanidin B一3($C_{30}H_{26}O_{12}$)이었다.

Antioxidant Principles of Nelumbo nucifera Stamens

  • In our ongoing study to identity antioxidants from natural sources, the antioxidant activity of Nelumbo nucifera stamens was evaluated for their potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals, inhibit total reactive oxygen species (ROS) generation, in kidney homogenates using 2 ,7 -dichlorodihydrofluorescein diacetate (DCHF-DA), and scavenge authentic peroxynitrites ($ONOO^-$). A methanol (MeOH) extract of the stamens of N. nucifera showed strong antioxidant activity in the $ONOO^-$system, and marginal activity in the DPPH and total ROS systems, so were therefore fractionated with several organic solvents, such as dichloromethane ($CH_2 Cl_2$), ethyl acetate (EtOAc) and n-butanol (n-BuOH). The EtOAc soluble fraction, which exhibited strong antioxidant activity in all the model systems tested, was further purified by repeated silica gel and Sephadex LH-20 column chromatographies. Seven known flavonoids [kaempferol (1), kaempferol 3-Ο-$\beta$-D-glucuronopyranosyl methylester (2), kaempferol 3-Ο-$\beta$-D-glucopyranoside (3), kaempferol 3-Ο-$\beta$-D-galactopyranoside (4), myricetin 3 ,5 -dimethylether 3-Ο-$\beta$-D-glucopyranoside (5), kaempferol 3-Ο-$\alpha$-L-rhamnopyranosyl-(1$\rightarrow$6)-$\beta$-D-glucopyranoside (6) and kaempferol 3-Ο-$\beta$-D-glucuronopyranoside (7)], along with $\beta$-sitosterol glucopyranoside (8), were isolated. Compound 1 possessed good activities in all the model systems tested. Compounds 2 and 7 showed scavenging activities in the DPPH and $ONOO^-$ tests, while compounds 3 and 4 were only active in the $ONOO^-$ test. Conversely, compound 8 showed no activities in any of the model systems tested.

[ $\beta$ ]-Secretase (BACE1) Inhibitors from Sanguisorbae Radix

  • In the course of screening anti-dementia agents from natural products, two $\beta$-secretase (BACE1) inhibitors were isolated from the ethyl acetate soluble fraction of Sanguisorbae Radix by the activity-guided purification using silica gel, Sephadex LH-20, and RP-HPLC. They were identified as 1,2,3-trigalloyl-4,6-hexahydroxydiphenoyl-$\beta$-D-glucopyranoside (Tellimagrandin II, 1) and 1,2,3,4,6-pentagalloyl-$\beta$-D-glucopyranoside (2) and were shown to non-competitively inhibit $\beta$-secretase (BACE1) with the $IC_{50}$ values of $3.10{\times}10^{-6}M\;and\;3.76{\times}10^{-6}M$, respectively. The Ki values of 1 and 2 were $6.84{\times}10^{-6}M\;and\;5.13{\times}10^{-6}M$. They were less inhibitory to asecretase (TACE) and other serine proteases such as chymotrypsin, trypsin, and elastase, suggesting that they were relatively specific inhibitors of BACE1.

Synthesis, Characterization and Antimicrobial Activity of Zirconium (IV) Complexes

  • Heteroleptic complexes of zirconium (IV) derived from bulky Schiff base ligands containing a sulphur atom and oximes of heterocyclic ${\beta}$-diketones of the general formula ZrLL' (where $LH_2=RCNH(C_6H_4)SC:C(OH)N(C_6H_5)N:CCH_3$, $R=-C_6H_5$, $-C_6H_4Cl(p)$ and $L^{\prime}H_2=R^{\prime}C:(NOH)C:C(OH)N(C_6H_5)N:CCH_3$, $R^{\prime}=-CH_2CH_3$, $-C_6H_5$, $-C_6H_4Cl(p)$ were prepared by the reactions of zirconium tetrachloride with disodium salts of Schiff bases ($L\;Na_2$) and oximes of heterocyclic ${\beta}$-diketones ($L^{\prime}\;Na_2$) in 1:1:1 molar ratio in dry refluxing THF. The structures of these monomeric zirconium (IV) complexes were elucidated with the help of elemental analysis, molecular weight measurements, spectroscopic (IR, NMR and mass) studies. A distorted trigonal bipyramidal geometry may be suggested for these heteroleptic zirconium (IV) complexes. The ligands (bulky Schiff base ligands containing a sulphur atom and oximes of heterocyclic ${\beta}$-diketones) and their heteroleptic complexes of zirconium (IV) were screened against A. flavus, P. aeruginesa and E. coli.

  • Six compounds were isolated from the EtOAc and $Et_2O$ fractions of the bark of Rhus chinensis by repeated column chromatography with $SiO_2$ and Sephadex LH-20. The structures were determined by instrumental analysis using MS and NMR spectrophotometer as: gallic acid (1), methyl gallate (2), 6, 7-dimethoxycoumarin (3), orcinol-${\beta}$-D-glucoside (4), scopoletin (5), semialactone (6). Among these compounds, 6,7-dimethoxycoumarin (3) was isolated from this plant for the first time. To measure the antioxidant activity, the DPPH radical scavenging activity test was performed. Gallic acid (1) showed the strongest activity, while orcinol-${\beta}$-D-glucoside (4), semialactone (5) and scopoletin (6) had the low activities.
