Lithspermic acid-A slows down the inactivation kinetics of cardiac $Na^+$ current by intracellular $Ca^{2+}$-dependent mechanisms

  • Salviae Miltiorrhizae Radix has been used for treatment of cardiovascular diseases in oriental medicine. To investigate the possible involvement of cardiac ion channel in this effect, we examined electrophysiological effects of the extract of Salviae Miltiorrhizae Radix on action potentials and ionic currents in rat ventricular myocytes. The extracts of Salviae Miltiorrhizae Radix were fractionated into nine fractions, and the effect of each fraction on action potential was tested. The fraction containing monomethyl lithospermic acid-A (LSA-A) induced a significant prolongation of action potential duration (APD). LSA-B which is a major component of Salviae Miltiorrhizae Radix, however, did not cause a significant effect. In voltage clamp experiments, the effects of LSA-A on K currents, Ca currents and Na currents were tested. Neither K currents nor L-type Ca currents were affected by LSA-A. On the contrary, LSA-A significantly slowed down the inactivation kinetics of the Na current with no effect on the fast component of the inactivation process. The amplitude of the peak current and the voltage-dependence of activation were not changed by LSA-A. The effect of LSA-A on Na current was abolished when high concentration of $Ca^{2+}$ buffer (10 mM BAPTA) was included in the pipette solution or when Ca2+ current was blocked by nicardipine (1 $\mu$M) in the bath solution.n.

  • This paper presents a compensation method of unbalanced phase currents in interleaved bi-directional converters. Phase currents in interleaved bi-directional converter are apt to be unbalanced due to circuit parameter error and switch operation difference. This problem causes the switch element failure and the reduced efficiency of the converter. Therefore, it is necessary that a certain balance control algorithm is provided in interleaved bi-directional converter system. In this paper, a balance control algorithm based on the circular chain control method is proposed. Further, in order to reduce the number of phase current sensors, this paper shows a simple method in which phase currents can be extracted indirectly through a DC-link current sensor in both charging and discharging modes. The validity and the effectiveness of the proposed phase currents balance control algorithm are illustrated through the simulation results.

One-Cycle Control Strategy for Dual-Converter Three-Phase PWM Rectifier under Unbalanced Grid Voltage Conditions

  • In this paper, a dual-converter three-phase pulse width modulation (PWM) rectifier based on unbalanced one-cycle control (OCC) strategy is proposed. The proposed rectifier is used to eliminate the second harmonic waves of DC voltage and distortion of line currents under unbalanced input grid voltage conditions. The dual-converter PWM rectifier employs two converters, which are called positive-sequence converter and negative-sequence converter. The unbalanced OCC system compensates feedback currents of positive-sequence converter via grid negative-sequence voltages, as well as compensates feedback currents of negative-sequence converter via grid positive-sequence voltages. The AC currents of positive- and negative-sequence converter are controlled to be symmetrical. Thus, the workload of every switching device of converter is balanced. Only one conventional PI controller is adopted to achieve invariant power control. Then, the parameter tuning is simplified, and the extraction for positive- and negative-sequence currents is not needed anymore. The effectiveness and the viability of the control strategy are demonstrated through detailed experimental verification.

Characterization of Acetylcholine-induced Currents in Male Rat Pelvic Ganglion Neurons

  • The pelvic ganglia provide autonomic innervations to the various urogenital organs, such as the urinary bladder, prostate, and penis. It is well established that both sympathetic and parasympathetic synaptic transmissions in autonomic ganglia are mediated mainly by acetylcholine (ACh). Until now, however, the properties of ACh-induced currents and its receptors in pelvic ganglia have not clearly been elucidated. In the present study, biophysical characteristics and molecular nature of nicotinic acetylcholine receptors (nAChRs) were studied in sympathetic and parasympathetic major pelvic ganglion (MPG) neurons. MPG neurons isolated from male rat were enzymatically dissociated, and ionic currents were recorded by using the whole cell variant patch clamp technique. Total RNA from MPG neuron was prepared, and RT-PCR analysis was performed with specific primers for subunits of nAChRs. ACh dose-dependently elicited fast inward currents in both sympathetic and parasympathetic MPG neurons $(EC_{50};\;41.4\;{\mu}M\;and\;64.0\;{\mu}M,\;respectively)$. ACh-induced currents showed a strong inward rectification with a reversal potential near 0 mV in current-voltage relationship. Pharmacologically, mecamylamine as a selective antagonist for ${\alpha}3{\beta}4$ nAChR potently inhibited the ACh-induced currents in sympathetic and parasympathetic neurons $(IC_{50};\;0.53\;{\mu}M\;and\;0.22\;{\mu}M,\;respectively)$. Conversely, ${\alpha}-bungarotoxin$, ${\alpha}-methyllycaconitine$, and $dihydro-{\beta}-erythroidine$, which are known as potent and sensitive blockers for ${\alpha}7$ or ${\alpha}4{\beta}2$ nAChRs, below micromolar concentrations showed negligible effect. RT-PCR analysis revealed that ${\alpha}3$ and ${\beta}4$ subunits were predominantly expressed in MPG neurons. We suggest that MPG neurons have nAChRs containing ${\alpha}3$ and ${\beta}4$ subunits, and that their activation induces fast inward currents, possibly mediating the excitatory synaptic transmission in pelvic autonomic ganglia.

Voltage-Dependent Inactivation of Calcium Currents in the Mouse Eggs

  • Inactivation properties of Ca current in the unfertilized eggs of mouse were studied by using the whole cell voltage clamp technique and single microelectrode voltage clamp technique. Membrane potential was held at -80 mV and step depolarization was applied from -50 mV to 50 mV for $200{\sim}500\;ms$. Peak of inward Ca currents was $-2{\sim}-4\;nA$ at a membrane Potentials from -20 mV to 0 mV and outward currents were not observed within the membrane voltage range studied $(-50{\sim}50\;mV)$. Inward currents were fully inactivated within 200 ms after the onset of step depolarization. As the membrane became depolarized, time constant of inactivation (${\tau}$) was decreased but remained around $20{\sim}30\;ms$ beyond 10 mV. When $Ca^{2+}$ was used as a charge earlier, inactivation of inward $Ca^{2+}$ current also occured and time course of inactivation was similar to that of $Ca^{2+}$ currents as charge carrier. In the bathing solution containing high potassium $(131\;mM\;K^+)$, process of inactivation was not changed except a parallel decrease of value for the entire range of membrane potential. Steady-state inactivation of the $current(h_{\infty})$ obtained from the double pulse experiment showed the voltage-dependent change. These results suggested that inactivation of Ca currents in the unfertilized eggs of mouse was voltage-dependent.

Involvement of Thromboxane $A_2$ in the Modulation of Pacemaker Activity of Interstitial Cells of Cajal of Mouse Intestine

  • Although many studies show that thromboxane $A_2\;(TXA_2)$ has the action of gastrointestinal (GI) motility using GI muscle cells and tissue, there are no reports on the effects of $TXA_2$ on interstitial cells of Cajal (ICC) that function as pacemaker cells in GI tract. So, we studied the modulation of pacemaker activities by $TXA_2$ in ICC with whole cell patch-clamp technique. Externally applied $TXA_2\;(5{\mu}M)$ produced membrane depolarization in current-clamp mode and increased tonic inward pacemaker currents in voltage-clamp mode. The tonic inward currents by $TXA_2$ were inhibited by intracellular application of GDP-${\beta}$-S. The pretreatment of ICC with $Ca^{2+}$ free solution and thapsigargin, a $Ca^{2+}$-ATPase inhibitor in endoplasmic reticulum, abolished the generation of pacemaker currents and suppressed the $TXA_2$-induced tonic inward currents. However, chelerythrine or calphostin C, protein kinase C inhibitors, did not block the $TXA_2$-induced effects on pacemaker currents. These results suggest that $TXA_2$ can regulate intestinal motility through the modulation of ICC pacemaker activities. This modulation of pacemaker activities by $TXA_2$ may occur by the activation of G protein and PKC independent pathway via extra and intracellular $Ca^{2+}$ modulation.

Degradation Properties of ZnO Surge Arresters Due to Lightning Impulse Currents (뇌임펄스전류에 의한 ZnO 피뢰기의 열화특성)

  • This paper describes the degradation properties of ZnO surge arresters impressed by lightning impulse currents. To investigate the deterioration behaviors of ZnO surge arresters due to lightning surges, the 8/20[${\mu}s$], 2.5[kA] standard lightning impulse currents were injected to the ZnO surge arrester under test. The power frequency AC and DC leakage currents flowing through the ZnO surge arresters with and without the injection of lightning impulse currents were measured. As a result, the leakage currents are increased and the asymmetry of the AC leakage current is pronounced as the number of injection of the impulse current increases. The ZnO grain of the surge arrester without the injection of lightning surges are uniform but the ZnO grain of the ZnO surge arrester with the injection of lightning impulse currents are deformed. Also, it was found that the decrease of the $Bi_2O_3$ due to the lightning impulse current leads to the lack of grain boundary layer and the current concentrated by the lack of grain boundary layer play an important role to degrade nonlinear property of ZnO surge arrester blocks.

Relationship of Bogi, Boyang, Onri herbs activated ion currents and excitatory neurotransmitter, glutamate, receptor (보기, 보양, 온리약에 의해 유발된 이온전류와 흥분성 신경전달물질인 glutamate 수용체와의 관계)

  • The effects of Bogi, Boyang and Onri herbs on glutamate receptor using nystatin perforated patch clamp under voltage clamp were investigated and the following results was obtained. Ginseng radix and Astragali radix were chosen as Bogi herbs. Cervi cornu and Boshniakiae herba were used as Boyang herbs. Aconiti tuber and Zingiberis rhizoma were selected as Onri herbs. 1. $10^{-6}M$ of glycine brought about the increase of the ion currents by Astragali radix. Cervi cornu. Boshniakiae herba and Aconiti tuber to a minute extent that has no experimental meaning. 2. It was assumed that Bogi herb had no effect on glutamate receptor since the ion currents of Ginseng radix and Astragali radix that are Bogi herbs had not been inhibited to a great extent by D-AP5 and CNQX. 3. Boyang herbs were seem to have meaningful relationship with non-NMDA receptor since the ion currents by Cervi cornu and Boshniakiae that are Boyang herbs were inhibited by CNQX but not inhibited to a meaningful extent by D-AP5. 4. D-AP5 had not a significant effect on the ion currents activated by Onri herbs such as Aconiti tuber and Zingiberis rhizoma. CNQX had a significant effect on the ion currents of Aconiti tuber but no effect on the ion currents of Zingiberis rhizoma. Therefore, it seemed that Aconiti tuber had a relationship with non-NMDA receptor while Zingiberis rhizoma had no relationship with glutamate receptor. In summary, Bogi, Boyang and Onri herbs had different effects through different mechanisms on glutamate receptor. Hence it was believed that. through experiments on acetylcholine receptor new classification could be made.

Wave Modeling considering Water Level Changes and Currents Effects (수위변화와 흐름효과를 고려한 파랑모델링)

  • In this study, wave model was conducted on the presence or absence of water level changes and currents effects in coastal waters coexisting with waves and currents, then the results were compared. The flow field applied the results of the RIAMOM model and the wave model applied the SWAN model. Among ECMWF, NCEP and JMA, wind data applied JMA data sets which agreed well with the observed data comparatively. Numerical simulation was conducted for 8 months from January to August 2016. For each case, the deviation of wave height was calculated for the high wave of more than 2.5 m for comparison with observed data. As a result, the deviation of wave height was not significant both considering water level changes and currents effects or not at wave observation stations installed in deep waters. However, a significant deviation of wave height of 5~10% was obtained depending on water level changes and currents effects at the comparison point in shallow waters.

  • This paper presents the RTDS training course on using passive filters for control of harmonic currents. The course can show harmonic currents in porer system, which is occurred by function generators, with Fourier analysis function of RTDS and the effect of passive filters implemented in RTDS to eliminate harmonics. In addition to, With Jeju-Haenam HVDC system, we have simulated the effect of passive filters on harmonic currents occurred by massive power conversion system of HVDC.
