Background: To investigate the relationship of five TP53 polymorphisms (p.P47S, p.R72P, PIN3 ins16bp, p.R213R and r.13494g>a) with the esophageal cancer (EC) risk in North Indians. Materials and Methods: Genotyping of p.P47S, p.R72P, PIN3 ins16bp, p.R213R and r.13494g>a polymorphisms of TP53 in 136 sporadic EC patients and 136 controls using polymerase chain reaction and PCR-RFLP. Results: The frequencies of genotype RR, RP and PP of p.R72P polymorphism were 16.91 vs 26.47%, 58.82 vs 49.27% and 24.27 vs 24.27% among patients and controls respectively. We observed significantly increased frequency of RP genotype in cases as compared to controls (OR=1.87, 95% CI, 1.01-3.46, p=0.05). The frequencies of genotype A1A1, A1A2 and A2A2 of PIN3 ins16bp polymorphism were 69.12 vs 70.59%, 27.20 vs 25% and 3.68 vs 4.41% among patients and controls. There was no significant difference among genotype and allele distribution between patients and controls. The frequencies of genotype GG, GA and AA of r.13494g>a polymorphism were 62.50 vs 64.70%, 34.56 vs 30.15% and 2.94 vs 5.15% among patients and controls respectively. No significant difference between genotype and allele frequency was observed in the patients and controls. For p.P47S and p.R213R polymorphisms, all the cases and controls had homozygous wild type genotype. The RP-A1A1-GG genotype combination shows significant risk for EC (OR=2.01, 95%CI: 1.01-3.99, p=0.05). Conclusions: Among the five TP53 polymorphisms investigated, only p.R72P polymorphism may contributes to EC susceptibility.
Background: The purpose of this study was to evaluate the potential association of five (p.P47S, p.R72P, PIN3 Ins16bp, p.R213R and r.13494g>a) polymorphisms of TP53 with the risk of developing breast cancer in North Indian Punjabi population. Methods: We screened DNA samples of 200 sporadic breast cancer patients (197 females and 3 males) and 200 unrelated healthy, gender and age matched individuals for the polymorphisms. Results: For the p.P47S polymorphism, we observed the PP genotype in 99.5% of the patients and PS genotype in only 1 patient. All the controls had the wild type PP genotype. The frequency of RR, RP and PP genotype of p.R72P was 23.5% vs 33.5%, 51.5% vs 45.5% and 25% vs 21% in patients and controls respectively. Heterozygous (RP) genotype was increased in breast cancer patients as compared to controls (51.5 vs 45.5%) and showed 1.61 fold significantly increased risk for breast cancer (OR=1.61, 95% CI, 1.01-2.58, p=0.04). In breast cancer patients the frequencies of A1A1, A1A2 and A2A2 genotypes of PIN3 Ins16bp polymorphism were 67%, 26% and 7% respectively whereas in controls the genotype frequencies were 68.5%, 27.5% and 4% respectively, with no significant difference. For p.R213R (c.639A>G), all individuals had homozygous wild type genotype. The frequencies of GG, GA and AA genotypes of TP53 r.13494g>a polymorphism were 62 vs 67.5%, 33 vs 28% and 5 vs 4.5% in patients and controls respectively, again without significant difference. We observed that RP-A1A1 genotype combination of p.R72P and PIN3 Ins16bp and RP-GG combination of p.R72P and r.13494g>a polymorphism showed significant risk of breast cancer (OR=1.65, 95%CI: 0.98-2.78, p=0.05; OR=1.72, 95%CI: 1.01-2.92, p=0.04). Conclusion: The results of present study indicated that among the five TP53 polymorphisms investigated, the p.R72P polymorphism, and the RP-A1A1 and RP-GG genotype combination contribute to breast cancer susceptibility in North Indians.

A comprehensive collection of proteins senses local changes in intracellular $Ca^{2+}$ concentrations ($[Ca^{2+}]_i$) and transduces these signals into responses to agonists. In the present study, we examined the effect of sphingosine-1-phosphate (S1P) on modulation of intracellular $Ca^{2+}$ concentrations in cat esophageal smooth muscle cells. To measure $[Ca^{2+}]_i$ levels in cat esophageal smooth muscle cells, we used a fluorescence microscopy with the Fura-2 loading method. S1P produced a concentration-dependent increase in $[Ca^{2+}]_i$ in the cells. Pretreatment with EGTA, an extracellular $Ca^{2+}$ chelator, decreased the S1P-induced increase in $[Ca^{2+}]_i$, and an L-type $Ca^{2+}$-channel blocker, nimodipine, decreased the effect of S1P. This indicates that $Ca^{2+}$ influx may be required for muscle contraction by S1P. When stimulated with thapsigargin, an intracellular calcium chelator, or 2-Aminoethoxydiphenyl borate (2-APB), an $InsP_3$ receptor blocker, the S1P-evoked increase in $[Ca^{2+}]_i$ was significantly decreased. Treatment with pertussis toxin (PTX), an inhibitor of $G_i$-protein, suppressed the increase in $[Ca^{2+}]_i$ evoked by S1P. These results suggest that the S1P-induced increase in $[Ca^{2+}]_i$ in cat esophageal smooth muscle cells occurs upon the activation of phospholipase C and subsequent release of $Ca^{2+}$ from the $InsP_3$-sensitive $Ca^{2+}$ pool in the sarcoplasmic reticulum. These results suggest that S1P utilized extracellular $Ca^{2+}$ via the L type $Ca^{2+}$ channel, which was dependent on activation of the $S1P_4$ receptor coupled to PTX-sensitive $G_i$ protein, via phospholipase C-mediated $Ca^{2+}$ release from the $InsP_3$-sensitive $Ca^{2+}$ pool in cat esophageal smooth muscle cells.
Brain, heart, liver, lung, kidney and thymus etc. 12 organs were removed and homogenized from Dawley-Sprague rats after suffocation. After fractionation of the tissue cytosols, enzymatic activities of the key enzymes in metabolic inositol phosphates cycle, PLC, IPSK and Ins(1,4,5) P35-phosphatase, were measured respectively. Hybridoma monoclones producing anti-lP3K murine monoclonal antibodies were obtained by the fusion of SP2/Ag 0-14 and spleen cells of mouse immunized with purified 53KDa IPSK, screening and cloning procedures. 18 cloned hybridoma cells were obtained, background due to nonspecific binding was very low with 10 clones. These Abs were purified from ascitic fluids by using affi-gel 15, and determined subtype of Abs. When immunoreactivities for rat tissues IP3K were exercised by adding the mixed Abs of 19Gl and 19G2b, they showed an overall similarity with noncompetitive inhibition. Brain tissue has high sensitivity for anti-lP3K Ab, whereas heart tissue has very low activity. In kinetic parameters Km value was 1.58 mM and Vmx value was 5.41umol/min/ml, respectively Only one form of 40 KDa IPSK was detected in heart tissues, however rat brain contains at least three immunologically distinct IP3K (53, 51 and 40 KDa) in western blot analysis. Of them 53 KDa protein was major enzyme in enzymatic activity. Northern blot analysis with 32P-labeled CDNA probe which encodes 1.8 Kb IPSK gene was performed. These results suggest that IPSK are regulated at transcriptional level during rat tissue development.
As the mechanism underlying glucose metabolism regulation by oxymatrine is unclear, this study investigated the effects of oxymatrine on pyroptosis in INS-1 cells. Flow cytometry was employed to examine cell pyroptosis and reactive oxygen species (ROS) production. Cell pyroptosis was also investigated via transmission electron microscopy and lactate dehydrogenase (LDH) release. Protein levels were detected using western blotting and interleukin (IL)-1β and IL-18 secretion by enzyme-linked immunosorbent assay. The caspase-1 activity and DNA-binding activity of nuclear factor kappa B (NF-κB) and nuclear factor (erythroid-derived 2)-like 2 protein (Nrf2) were also assessed. In the high glucose and high fat-treated INS-1 cells (HG + PA), the caspase-1 activity and LDH content, as well as Nod-like receptor family pyrin domain containing 3, Gsdmd-N, caspase-1, apoptosis-associated speck-like protein containing a CARD, IL-1β, and IL-18 levels were increased. Moreover, P65 protein levels increased in the nucleus but decreased in the cytoplasm. Oxymatrine attenuated these effects and suppressed high glucose and high fat-induced ROS production. The increased levels of nuclear Nrf2 and heme oxygenase-1 (HO-1) in the HG + PA cells were further elevated after oxymatrine treatment, whereas cytoplasmic Nrf2 and Keleh-like ECH-associated protein levels decreased. Additionally, the elevated transcriptional activity of p65 in HG + PA cells was reduced by oxymatrine, whereas that of Nrf2 increased. The results indicate that the inhibition of pyroptosis in INS-1 cells by oxymatrine, a key factor in its glucose metabolism regulation, involves the suppression of the NF-κB pathway and activation of the Nrf2/HO-1 pathway.
본 연구에서는 60대 이상 한국 여성 노인을 대상으로 하여 체내 나트륨 대사에 밀접하게 관여하는 ADD1 Gly460Trp, AGT Met235Thr, ACE Ins/Del 유전자형의 분포를 살피고 각각의 유전자 다형성 혹은 유전자형의 조합과 수축기 및 이완기 혈압과의 관계, 그리고 식이 나트륨 섭취 수준이 유전자-혈압과의 관계에 미치는 영향에 대해 파악하고자 수행 되었으며 그 결과를 요약하면 다음과 같다. 본 연구에서 분석된 유전자형들의 분포는 1) ADD1 유전자-Gly/Gly:Gly/Trp:Trp/Trp=16.5:49.5:34.0, 2) AGT 유전자-Met/Met:Met/Thr:Thr/Thr=4.6:31.2:64.2, 3) ACE 유전자-Ins/Ins:Ins/Del:Del/Del=34:49.5:16.5이었다. AAD1 Gly460Trp, AGT Met235Thr, ACE Ins/Del 각각의 유전자형은 본 연구대상자들의 수축기 및 이완기 혈압을 유의하게 변화시키지 않았으나, ACE Del/Del형과 ADD1 Trp/Trp형을 동시에 보유하고 있는 경우 다른 유전자형을 가진 대상자들에 비해 수축기 혈압이 유의적으로 높았으며(p=0.01), ACE Del/Del형과 AGT Met allele을 동시 에 보유하고 있는 경우 다른 그룹들에 비해 높은 이완기 혈압을 가지고 있는 것으로 나타났다(p<0.001).식이 나트륨 섭취량의 상대적인 수준에 따라 전체 대상자를 두 그룹으로 나누었을 때, 나트륨 섭취량이 낮은 그룹에서만 ADD1 Gly460Trp유전자형에 따른 평균 수축기 혈압의 차이가 관찰되었고(p=0.03), 나트륨 섭취량이 높은 그룹에서는 ADD1유전자형별 평균 수축기 및 이완기 혈압에 유의적인 차이가 없었다. 이상의 결과는 한국 여성 노인에 있어 혈압의 증가에 ADD1, AGT 및 ACE유전자 다형성이 복합적으로 관여함을 제시한다. 또한, 이들 유전자 다형성에 대한 혈압의 표현형이 식이 나트륨 섭취 수준에 따라 변화함을 규명하였다. 이러한 결과로 볼 때, 앞으로 유전자-질병간의 연관성을 밝히기 위한 연구들에서 단일 유전자보다는 다양한 유전자들에 대한 분석이 복합적으로 이루어져야 하며, 식이 요인 등 주요 환경 요인에 대한 분석이 반드시 함께 수행되어야 할 것으로 생각된다.
Kinesin-1은 2개의 장쇄(KHCs, 또는 KIF5s)와 2개의 단쇄(KLCs)가 결합한 복합체로 되어 있다. 본 연구에서 효모 two-hybrid system을 이용하여 중추신경계의 신경세포에서 주로 발현되는 KIF5A와 결합하는 단백질을 탐색한 결과 phosphatidylinositol-3,5-bisphosphate ($PI(3,5)P_2$)의 5번 위치 인산을 제거하는 탈인산화효소 Fig4(Sac3)를 분리하였다. KIF5A는 Fig4의 C-말단과 결합함을 효모 two-hybrid assay로 확인하였다. Fig4는 KIF5A의 C-말단과 결합하지만, 두 개의 다른 장쇄인 KIF5B와 KIF5C 그리고 KLC1와는 결합하지 않았다. 단백질 간 결합을 glutathione S-transferase pull-down assay와 공동면역침강으로 추가 검증하였다. 생쥐의 뇌 파쇄액을 KIF5A 항체로 면역 침강한 결과 Fig4가 같이 침강하였다. 이러한 결과들은 kinesin-1이 Fig4와 결합한 단백질 복합체 혹은 운반체를 세포 내에서 운반함을 시사한다.

It is now generally accepted that a phosphoinositide cycle is involved in the transduction of a variety of signals in plant cells. In animal cells, the hydrolysis of phosphatidyl-4,5-bisphosphate catalysed by phosphatidylinositol - specific phospholipase C yields to D-myo-inositol - 1,4,5-trisphosphate and diacylglycerol, which are well known second messengers. The binding of InsP$_3$to a receptor located on the endoplasmic reticulum triggers a calcium release from the endoplasmic reticulum. We have detected and partially characterised key components of phosphoinositide signaling. First, tobacco microsomal fraction and plasma membrane PI-PLC. Consecutively, using a radioligand binding assay we have identified a $Ca^{2+}$ -dependent high affinity InsP$_3$binding site in microsomal membrane fraction vesicle preparation and then we have measured inositol-1,4,5-trisphosphate induced calcium release from tobacco microsomal fraction. These findings suggest that phosphoinositide signaling system is present and operates in the tobacco suspension culture.e.
큰가리비 치패의 대체 먹이원을 개발하기 위해 각기 다른 먹이원을 공급하여 성장, 생존율 및 체내 조성을 조사하였다. 생존율은 PTE + PHY 실험구에서 90.0%로 가장 높게 나타났다. 각장과 각고는 PHY에서 가장 높게 나타났으나 PTE +PHY 실험구와의 유의적인 차이는 없었다. 반면, PTE와 OTE 단독구는 유의적으로 가장 낮게 나타났다. 한편, 각폭 및 전중량도 동일한 경향을 보여 PHY 단독 실험구와 PTE+PHY 혼합 실험구에서 유의적으로 가장 높게 나타난 반면 PTE와 OTE 단독구는 가장 낮은 값을 보였다. 한편, 체내 지방산 분석에서 DHA 함량은 기존 먹이인 미세조류 단독 실험구 (PHY) 와 경제적인 먹이원인 INS 실험구에 비해 지질강화원인 PTE와 OTE가 혼합된 실험구에서 더욱 높은 것으로 나타났으며, 이러한 높은 값에 기인하여 n-3 PUFA도 높았다. 또한, 총 단백질 함량은 주요 단백질원인 PHY와 INS가 들어간 실험구가 55.5-65.2%의 함량을 보인반면 지질영양강화원만 들어간 PTE, OTE 단독 실험구는 각각 44.8%, 47.0%로 가장 낮은 함량을 나타내었다. 필수아미노산 함량도 동일한 경향을 보여 단순 지질강화원 실험구에서는 낮았다. 한편, 체내 핵산 분석에서 RNA 값은 PHY 단독 실험구와 PTE+PHY 혼합구에서 0.76으로 가장 높게 나타났으나 PTE, OTE 단독구는 각각 0.35, 0.32로 가장 낮게 나타났다. DNA 값도 PHY 단독 실험구와 PTE + PHY 혼합구에서 3.95으로 가장 높게 나타난 반면 PTE, OTE 단독구는 유의적으로 가장 낮았다. 이러한 경향에 의해 RNA/DNA ratio 값도 PHY와 INS가 들어간 단독구와 몇몇 혼합구가 PTE와 OTE 단독구 보다 높았다. 이상의 결과를 통해서 미세조류 단독 공급구인 PHY에 PTE를 혼합해 준 PTE+PHY 혼합 공급구는 큰가리비 치패의 성장, 생존율 및 체내 조성을 향상시켜 주는 것으로 나타났다. 본 실험을 통해 새롭게 대체 개발된 PTE+PHY 혼합구는 이매패류 양식에서 차지하는 미세조류 생산비용의 절감 효과를 기대해 볼 수 있는 것으로 경제적인 측면에서 매우 유용한 결과라 할 수 있다.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
