Characteristics of Fumarate Reductase from Enterococcus faecalis RKY1 (Enterococcus faecalis RKY1 이 생산하는 Fumarate Reductase의 특성)

  • An oxygen-sensitive fumarate reductase has been purified from the cytosol fraction of the Enterococcus faecalis RKY1 grown anaerobically on a defined medium containing glycerol and fumarate. A major portion of the purification was performed with employing Triton X-100 and reducing agents by Phenyl-sepharose CL-4B DEAE-sepharose and Dephadex G-150 The final activity was 0.42 unit/mg. The deduced molecular mass of active band was 66 kDa. The optimal pH and temperature for the activity were 7.0 and 38$^{\circ}C$ respectively. The enzyme activity was not affected by 1mM metal ions such as bacl2 $.$2H2O HgCl2 MnCl2$.$4H2O ZnCl2 CuCl2$.$2H2O Mgcl2$.$6H2O FeSo4$.$7H2O and by EDTA. Partially purified enzyme ws yellow in color ; spectroscopic study indicated the presence of flavins as a cofactor.

Mercury recovery from aqueous solutions by polymer-enhanced ultrafiltration using a sulfate derivative of chitosan

  • The sulfatation of chitosan, by reaction with chlorosulfonic acid under controlled conditions, allowed increasing the pH range of chitosan solubility. The biopolymer was characterized using FTIR and $^{13}C$-NMR spectroscopy, elemental analysis and titration analysis and it was tested for mercury recovery by polymer enhanced ultrafiltration (PEUF). In slightly alkaline conditions (i.e., pH 8) mercury recovery was possible and at saturation of the polymer the molar ratio $-NH_2$/Hg(II) tended to 2.6. Polymer recycling was possible changing the pH to 2 and the polymer was reused for 3 cycles maintaining high metal recovery. The presence of chloride ions influences metal speciation and affinity for the polymer and "playing" with metal speciation allowed using the PEUF process for mercury separation from cadmium; at pH 11 the formation of hydroxo-complexes of Hg(II) limits it retention. Cake formation reveals the predominant controlling step for permeation flux.

  • A chloroform membrane system containing a given mixture of benzylaza-12-crown-4 and oleic acid is introduced for the selective and efficient transport of $Pb^{2+}$ ion. The transport was capable of moving metal ions ‘up-hill’. In the presence of ${S_2O_3}^{2-}$ion as a suitable metal ion acceptor in the receiving phase, the amount of lead ion transport across the liquid membrane after 150 minutes is (95.0 ${\pm}$ 1.7)%. The selectivity and efficiency of lead transport from aqueous solution containing $Tl^+,\;Ag^+,\;Mg^{2+},\;Ca^{2+},\;Co^{2+},$ $Ni^{2+},\;Cu^{2+},\;Zn^{2+},\;Hg^{2+}$ and $Cd^{2+}$were investigated. In the presence of thiosulfate as a suitable masking agent in the source phase, the interfering effects of $Ag^+\;and\;Cu^{2+}$ were diminished drastically.

  • Agar degrading enzyme-agarase-was purified from the culture fluid of Cytophaga so/ ACLJ-18, by acetone precipitation, DEAE-Cellulose, Sephadex G-100 and CM-Sephadex C25 column chromatographies. The molecular weight of purified agarase was estimated to be 24,700 dalton by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for agarase activity were 7.0 and 40$^{\circ}C$, respectively. this agarase was stable in the pH range of 6.5 - 8.0 and 40$^{\circ}C$, and required 0.35M NaCl for optimum activity. And this agarase was inhibited by metal ions such as Ba2+, Cu2+, Co2+, Mn2+, Hg2+, Zn2+, and showed specificity on agar.

  • Our works performed for preparation of oligosaccharides from carrageenan, seaweed polysaccharide, and one active strain for carrageenan was isolated from sea water and identified to Pseudomonas alcaligenes. Carrageenan degrading enzyme was purified from the culture fluid of isolated strain-Pseudomonas alcaligenes JCL-43, by DEAE-Cellulose, Sephadex G-100, Q-Sepharose and CM Sepharose CL-6B column chromatography. Two enzyme-F-I, F-II- was identified this purifying process, and the molecular weight of the purified carrageenase were estimated to be 23.6kDa and 30.2kDa, respectively. The optimum pH and temperature for two carrageenase activity were 7.0 and 4$0^{\circ}C$. These enzymes were stable in the pH range of 6.0~7.5 and lower than 5$0^{\circ}C$, and required 1.5% NaCl for optimum activity. And these carragennase were inhibited by metal ions such as Cu2+, Zn2+, Hg2+, but increased by Ba2+ and Ca2+, and showed specificity on -carrageenan.

  • The silver selenide electrode has been prepared and its properties as an indicating electrode for silver ion have been investigated. Epoxy resin was used as a filler of silver selenide electrode. Silver metal plate was directly connected with the membrane of the electrode and the silver paste was used as its binder. The sintered electrode was more sensitive and stable than the pressed electrode, and the silver selenide electrode more sensitive than the silver sulfide electrode to silver ion. The linear relationship between the electrode potential and logarithmic concentration of silver ion has been observed down to 10-6 M for the electrode. Several heavy metal ions except mercuric ion did not interfere this linearity, but halide, cyanide, and thiocyanate ions did intensively interfere owing to the formation of silver compounds and complexes. This electrode has been applied to the potentiometric titration for determining halide ion. It is concluded that interferences from ,$CN^-, SCN^-, S^-, I^-, Br^-, Cl^- and Hg^{2+}$ ions are detrimental to the practical use of the electrodes for measuring pAg.

Properties of Crude Trehalase from Agaricus bisporus (양송이 중의 조(粗) Trehalase의 분리와 그 성질)

  • In order to study the trehalase (EC 3. 2. 1. 28) from mushroom, Agaricus bisporus Lange Sing., the crude trehalase preparation was separated by fractionation of mushroom extracts with ammonium sulfate between 0.4 and 1.0 saturation, and its properties were examined. Mushroom trehalase showed optimum pH 6.0, and optimum temperature $40^{\circ}C$. The enzyme was stable at pH range between 5.0 and 7.0, and at temperature below $50^{\circ}C$. The activities of crude trehalase had proportional relations with enzyme concentrations below 490.2 mg % of protein and with substrate concentration below $2.6{\times}10^{-3}M$, showing a Km value of 0.760 mM. The enzyme was inhibited by some metal ions such as $Sn^{2+}$, $Ca^{2+}$, $Hg^{2+}$, $Cd^{2+}$, $Cu^{2+}$, $Mn^{2+}$, $Zn^{2+}$, $Al^{3+}$, and $Fe^{3+}$, while $Ag^{+}$, $Ba^{2+}$, and $Mg^{2+}$ demonstrated remarkable increasing effects on the enzyme activity.

  • Polarographic studies of reduction of Ni(Ⅱ)-CN complex on Hg-electrode have indicated that $Ni(CN)_4^{2-}$ is reduced by two paths, via one electron process Ni(CN)42- + e [1]↔[2] Ni(CN)43- =(eq) Ni(CN)2- + 2CN- and via two electron process Ni(CN)42- + 2e [3]→ 1/2[Ni(CN)33-]2 + CN- of which reduction [1] must be faster than reduction [3]. At very dilute cyanide concentration (0.004 to 0.01 M) cathodic wave is practically responsible for reaction [1] and two cyanide ions appear to contribute to the reaction. As increasing cyanide ion concentration the rate of oxidation reaction [2] catalysed by Hg increases and reaction [1] and [2] approach to equilibrium. Therefore, reaction [3] represents the cathodic wave at high concentration of cyanide (above 0.2 M). This mechanism can also explain the fact that limiting current at $[CN^-]$ = 8 M is approximately twice of that at 0.004 M CN.

  • Colorimetric sensors were usually used to detect specific metal ions using selective color change of solutions. While almost organic dye in colorimetric sensors detected single molecule, dithizone (DTZ) solution could be separately detected above 5 kinds of heavy metal ions by the change of clear color. Namely, DTZ could be used as multi-colorimetric sensors. However, DTZ was generally used as aqueous type and paper/pellet-type DTZ was not reported yet. Therefore, in this work, polystyrene (PS) was prepared to composite with DTZ and then DTZ/PS pellet was obtained, which was used to selectively detect 10 kinds of heavy metal ions. When 10 ppm of Hg and Co ions was exposed in DTZ/PS pellets, clear color change was revealed. It is noted that DTZ/PS pellet could be used in detecting of heavy metal ion as dry type.

Biochemical Properties of NAD(P)H-Quinone Oxidoreductase from Saccharomyces cerevisiae

  • The NAD(P)H-quinone oxidoreductase (EC 1. 6. 99. 2) was purified from S. cerevisiae. The native molecular weight of the enzyme is approximately 111 kDa and is composed of five identical subunits with molecular weights of 22 kDa each. The optimum pH of the enzyme is pH 6.0 with 1,4-benzoquinone as a substrate. The apparent $k_m$ for 1,4-benzoquinone and 1,4- naphthoquinone are 1.3 mM and $14.3\;{\mu}M$, respectively. Its activity is greatly inhibited by $Cu^{2+}$ and $Hg^{2+}$ ions, nitrofurantoin, dicumarol, and Cibacron blue 3GA. The purified NAD(P)H-quinone oxidoreductase was found capable of reducing aromatic nitroso compounds as well as a variety of quinones, and can utilize either NADH or NADPH as a source of reducing equivalents. The nitroso reductase activity of the purified NAD(P)H-quinone oxidoreductase is strongly inhibited by dicumarol.
