Effects of Mercuric Chloride on the Differentiation Cerebral Neuron of Chick Embryo (II) (계배 대뇌의 신경세포 분화에 미치는 수은의 영향 (II))

  • To investigate the effects of mercuric chloride ($HgCl_2$) on the differentiation of the cerebral neuron of chick embryo 9 days, the ultrastructural changes in nerve cells injected with a various doses of mercuric chloride were observed with transmission electron microscope. The enzyme activity of the some dehydrogenases, cerebral proteins and adenosine triphosphate (ATP) were also analyzed. The results obtained are as follows: The ultrastructural changes in 0.5 and 1.0mg-injected groups were undetectable, but in 2.0mg-injected group, the nuclear envelops were very irregular and mitochondria, were swelled and destroyed partly. The number of polypeptide bands separated by SDS-PAGE in the normal group were 37 bands. According to the in creased dose of mercuric chloride, contends of the bands were increased in 7 bands. The activities of dehydrogenases were declined by increasing the dose of mercuric chloride. Lactate dehydrogenase (LDH) activity failed to 78% in 1.0mg-injected group and greatly to 68% in 2.0 mg-injected group. Malate dehydrogenase (MDH) activity failed to 81% in 2.0 mg-injected group. On the other hand, succinate dehydrogenase (SDH) activity decreased to 80% in 1.0 mg-injected group and greatly to 63% in 2.0 mg-injected group. ATP content in 1.0 mg-injected group was increased slightly and in 2.0 mg-injected group was increased greatly.

  • These studies were conducted an attempt to investigate effects of 'Paeryungtang' and 'Kamipaeryungtang' water extracts on diuretic, antipyretic, antiinflammatory and analgesic actions. The results of these studies were summarized as follows; Increase in urinary volume, urinary $Na^+$ excretions were significantly recognized in normal rat. Increase in urinary volume, urinary $Na^+\;and\;Cl^-$ excretions were significantly shown in rat with 2mg/kg $HgCl_2$-induced acute renal failure. Antipyretic, anti-inflammatory and analgesic effects of 'Paeryungtang' and 'Kamipaeryungtang' were recognized in mice, rats and rabbits.

  • The paramagnetic organoruthenium(III) complexes $({\eta}^5-C_5Me_5)RuCl_2(PR_3) (PR_3 = PMe_3,\;PEt_3,\;PiPr_3,\;PCy_3,\;PMe_2Ph,\;PMePh_2,\;PPh_3,\;P(p-C_6H_4CH_3)_3$, DPPE, DPPB, Py) (2a∼2k) were synthesized by the reaction of $[({\eta}^5-C_5Me_5)RuCl_2]_2$ (1) with 1 equivalent of the corresponding phosphines $(PR_3)$. The effective magnetic moment ((${\mu}_{eff} = 1.65∼2.07 B.M.$)) derived from the magnetic susceptibility measurements of the complexes (2a∼2k) were consistent with the presence of a "single" unpaired electron in the molecule. Treatment of dichlororuthenium (III) complex ({\eta}^5-C_5Me_5)RuCl_2(PR_3)$ (2) (i) with KBr in acetone afforded the dibromoruthenium (III) complex $({\eta}^5-C_5Me_5)RuBr_2(PR_3) (PR_3 = PPh_3)$, (ii) with sodium amalgam in diethylether led to the bis(phosphine) derivatives $({eta}^5-C_5Me_5)RuCl(PR_3)_2 (PR_3 = PMe_3,\;PMePh_2)$, and (iii) with carbonmonoxide gave to the carbonyl derivatives $({\eta}^5-C_5Me_5)RuCl(PR_3)(CO) (PR_3 = PMe_3,\;PPh_3)$.

  • The effects of thermo-solutal convection on mercurous chloride system of ${Hg_2}{Cl_2}$, and Ne during physical vapor transport are numerically investigated for further understanding and insight into essence of transport phenomena, For $10\;K{\le}{\Delta}T{\le}30\;K$, the growth rate slowly increases and, then is decreased gradually until ${\Delta}T$=50 K, The occurrence of this critical point near at ${\Delta}T$=30 K is likely to be due to the effects of thermo-physical properties stronger than the temperature gradient corresponding to driving force for thermal convection. For the range of $10\;Torr{\le}P_B{\le}300\;Torr$, the rate is second order-exponentially decayed with partial pressures of component B, $P_B$. For the range of $5{\le}M_B{\le}200$, the rate is second order-exponentially decayed with a function of molecular weight of component B, $M_B$. Like the case of a partial pressure of component B, the effects of a molecular weight arc: reflected through the binary diffusivity coefficients, which are intimately related with suppressing the convection flow inside the growth enclosure, i,e., transition from convection to diffusion-dominant flow mode as the molecular weight of B increases. The convective mode is near at a ground level, i,e., on earth (1 $g_0$), and the convection is switched to the diffusion mode for $0.1\;g_0{\le}g{\le}10^{-2}g_0$, whereas the diffusion region ranges from $10^{-2}g_0$ up to $10^{-5}g_0$.

Purification and Properties of Cyclodextrin glycosyltransferase from Bacillus stearothermophilus KY-126 (Bacillus stearothermophilus KY-126가 생산하는 Cyclodextrin glycosyltransferase의 정제 및 특성)

  • A bacterial strain No. KY-126, which produced extracellular cyclodextrin glycosyltransferase(CGTase), was isolated from soil and identified as Bacillus stearothermophilus KY-126. The enzyme was purified by the treatments of ammonium sulfate precipitation, DEAF-Sephadex, Sephadex G-100 column chromatography. The optimal pH and temperature for the enzyme activity were pH 5.5 and $65^{\circ}C$, respectively. And the enzyme was stable at pH values from 6.0 to 11.0 at $55^{\circ}C$ for 30 min and stable up to $60^{\circ}C$ for 30 min.. The enzyme was inhibited by $HgCl_{2}$. The molecular weight of the enzyme was estimated to be 67,000 by using SDS-PAGE. The maximum conversion from starch to cyclodextrin (CD) by CGTase was 43% and obtained at 6 hr reaction and the ratio of ${\alpha}-,\;{\beta}-,\;{\gamma}-$, CD production at this time was 2.9 : 2.1 : 1.0.

Mercury-Specific Effects on Photosynthetic apparatus of Barley Chloroplasts Compared with Copper and Zinc Ions (구리${\cdot}$아연과 비교한 보리 엽록체의 광합성 기구에 미치는 수은 이온의 특이한 효과)

  • To find heavy metal-specific effects on the photosynthetic apparatus of higher plants, we investigated effects of $CuCl_2$, HgCl_2$ and $ZnCl_2$ on electron transport activity and chlorophyll fluorescence induction kinetics of chloroplasts isolated from barley seedlings. Effects on some related processes such as germination, growth and photosynthetic pigments of the test plants were also studied. Germination and growth rate were inhibited in a concentration-dependent manner by these metals. Mercury was shown to be the most potent inhibitor of germination, growth and biosynthesis of photosynthetic pigments of barley plants. In the inhibition of electron transport activity, quantum yield of PS II, and chlorophyll fluorescence induction kinetics of chloroplasts isolated from barley seedlings, mercury chloride showed more pronounced effects than other two metals. Contrary to the effects of other two metals, mercury chloride increased variable fluorescence significantly and abolished qE in the fluorescence induction kinetics from broken chloroplasts of barley seedlings. This increase in variable fluorescence is due to the inhibition of the electron transport chain after PS ll and the following dark reactions. The inhibition of qE could be attributed to the interruption of pH formation and do-epoxidation of violaxathin to zeaxanthin in thylakoids by mercury. This unique effect of mercury on chlorophyll fluorescence induction pattern could be used as a good indicator for testing the presence and/or the concentration of mercury in the samples contaminated with heavy metals.

Studies on the Utilization of Phenolic Substance by Yeast (효모에 의한 phenol 성 물질의 자화에 관한 연구)

  • Phenol utilizing yeast No. 558 isolated from soil sewage sediment was able to use substantial amount of phenol as the sole carbon source, and the biomass productivity by this organism was very excellent. This organism could grow well in 1000 ppm of phenol concentration, the maxim-um specific growth rate obtainable at pH 5.0, 3$0^{\circ}C$ was 0.27/hr., and the biomass yield coefficient Y vs. consumed phenol was 3.2. Maximum production rate of biomass was observed at 35$^{\circ}C$, pH 3.5 to pH 4.5, and the addition of the 0.005~0. 01% yeast extract was the most effective. Addition of HgCl$_2$ and phenyl hydrazine, inhibitors of oxide-reductase, in the phenol containing cultural liquid caused this organism no-growth at the concentration of 10$^{-5}$ M, 10$^{-3}$ M respectively. This organism could utilize not only phenol but catechol, resorcinol and benzidine.

Importance of convection during physical vapor transport of Hg2Cl2 in the presence of Kr under environments of high gravitational accelerations

  • Special attention in the role of convection in vapor crystal growth has been paid since some single crystals under high gravity acceleration of $10g_0$ appear considerably larger than those under normal gravity acceleration ($1g_0$). With increasing the gravity acceleration from $1g_0$ up to $10g_0$, the total molar flux for ${\Delta}T$ = 30 K increases by a factor of 4, while for ${\Delta}T$ = 90, by a factor of 3. The maximum molar fluxes for three different gravity levels of $1g_0$, $4g_0$ and $10g_0$, appear approximately in the neighborhood of y = 0.5 cm, and the molar fluxes show asymmetrical patterns, which indicate the occurrence of either one single or more than one convective cell. As the gravitational level is enhanced form $1g_0$ up to $10g_0$, the intensity of convection is increased significantly through the maximum molar fluxes for ${\Delta}T$ = 30 K and 90 K. At $10g_0$, the maximum total molar flux is nearly invariant for for ${\Delta}T$ = 30 K and 90 K. The total molar flux increases with increasing the gravity acceleration, for $1g_0{\leq}g_y{\leq}10g_0$, and decreases with increasing the partial pressure of component B, a noble gas called as Kr (Krypton), $P_B$. The ${{\mid}U{\mid}}_{max}$ is directly proportional to the gravity acceleration for 20 Torr $P_B{\leq}300$ Torr. As the partial pressure of $P_B$ (Torr) decreases from 300 Torr to 20 Torr, the slopes of the ${{\mid}U{\mid}}_{max}s$ versus the gravity accelerations increase from 0.29 sec to 0.54 sec, i.e. by a factor of 2. The total molar flux of $Hg_2Cl_2$ is first order exponentially decayed with increasing the partial pressure of component B, $P_B$ (Torr) from 20 Torr up to 300 Torr.

Purification and Properties of Extracellular Protease from Streptomyces rimosus (Streptomyces rimosus가 생산하는 Protease의 정제와 특성)

  • Extracellular neutral pretense of Streptomyces rimosus producing oxytetracycline was purified by ammonium sulfate fractionation, DEAE Sephadex A-50 chromatography and Sephadex G-100 gel filteration, and was showed single band on the cathodic gel electrophoresis. The optimum pH and temperature of the enzyme were pH 8.0 and 6$0^{\circ}C$, respectively. The enzyme was activated about 80% in the presence of Co$^{2+}$ ion, and strongly inhibited by Hg$^{2+}$, Fe$^{2+}$ and chelatig agent, EDTA. Molecular weight of the enzyme was estimated to be 12, 000. The Km value of the enzyme of casein as a substrate was 2.7$\times$10$^{-4}$M.

Salinity and Heavy Metal Contents of Solar Salts Produced in Jeollanamdo Province of Korea (전라남도 지역에서 생산된 천일염의 염도 및 중금속 함량)

  • To assess the safety of solar salts of Jeollanamdo province where most of Korean solar salts are produced, 433 samples of solar salts in total were collected and analyzed for their contents of sodium chloride and heavy metals (lead, cadmium, arsenic and mercury). Sodium chloride contents ranged from 80 to 85% in 63% of solar salts analyzed. Lead was not detected in 77% of solar salt samples and showed a value of below the maximum permissible limit (2 ppm) in other samples. Cadmium was not detected in most samples (78%), and the other samples where cadmium was detected showed a value far below the maximum permissible limit (0.5 ppm) except for three ones. Arsenic was detected in only four samples, but their concentrations were far below the permissible limit (0.1 ppm). Mercury was not detected in all samples. There were some differences between producing areas in the levels of sodium chloride and lead of solar salts, but the contents of cadmium, arsenic and mercury did not show significant regional differences. These results clearly indicate that solar salts produced from Jeollanamdo province are safe in the aspects of lead, cadmium, arsenic and mercury contents.
