Synthesis of Ultra-fine Calcium Phosphate Powders from Ca(OH)2 Suspension and Various Phosphoric Aqueous Solutions (Ca(OH)$_2$ 현탁액과 각종 인산 수용액으로부터 인산칼슘 초미분말의 제조)

  • Ultra-fine calcium phosphate powders were synthesized by the reaction of Ca(OH)2 suspension with various phosphoric aqueous solutions such as (NH4)2HPO4, H4P2O7 and H3PO4, and the characterization of powders was examined for each synthetic condition. When (NH4)2HPO4 and H3PO4 were used, hydroxyapatite powders with poor crystallinity were obtained. In the case of H4P2O7, amorphous calcium phosphate was obtained up to 0.3 mol/ι Ca(OH)2 suspension, but above the concentration, poor crystalline hydroxyapatite was produced. Crystalline phases of powders heat-treated at 80$0^{\circ}C$ were hydroxyapatite, $\beta$-tricalcium phosphate and $\beta$-tricalcium phosphate for the case of (NH4)2HPO4, H4P2O7 and H3PO4, respectively. SEM observation revealed that the shapes of synthesized powders were vigorously agglomerated spherical with the size below 100 nm, but TEM observation revealed that primary shapes of particles were rod for (NH4)2HPO4 and H3PO4 and were sphere for H4P2O7. There was no dependence of the concentration of Ca(OH)2 suspension. In the case that reaction temperature and pH of the suspension were raised, the inclination to the hydroxyapatite were remarkable. The amorphous calcium phosphate synthesized in this experiment contained water about 20% , and was crystallized to $\beta$-tricalcium phosphate at 69$0^{\circ}C$.

  • The matrix tablet containing sodium alginate and $CaHPO_4$ can release drugs in a controlled fashion from hydrogel with gelling and swelling due to their interaction as water penetrates the matrices of the tablet. The purpose of this study was to evaluate release characteristics of the matrix tablet varying the amount of sodium alginate, $CaHPO_4$ and other excipients such as chitosan, hydroxypropyl methylcellulose (HPMC) and $Eudragit^{\circledR}$ RS100 in the simulated gastric and intestinal fluid. The practically soluble ibuprofen was used as a model drug. The release profiles of matrix tablet in the gastric fluid as a function of sodium alginate/$CaHPO_4$ ratio was not pronounced because of low solubility of drug and stability of alginate matrices. However, release rate of drug from the matrix tablet in the intestinal fluid was largely changed when sodium alginate/$CaHPO_4$ ratio was increased, suggesting that the ratio of sodium alginate/$CaHPO_4$ was an important factor to control the gelling and swelling of the matrix tablet. The incorporation of other excipients into the matrix tablet also influenced the release rate of drug. The chitosan and HPMC decreased the release rate of drug. No release of drug was occurred when $Eudragit^{\circledR}$ RS100 was added into the tablet. The retarded release of matrix tablet when excipients were added resulted from the hindrance of swelling and gelling of the matrix tablet containing sodium alginate and $CaHPO_4$. The hardness and bulk density of the matrix tablet was not correlated with release rate of drug in the study. From these findings, the ratio of sodium alginate and $CaHPO_4$ in the matrix tablet in addition to incorporation of excipients could be very important to control the release rate of drug in dosage form design.

  • $\alpha$-tricalcium phosphate($\alpha$-TCP) powders were synthesized and their hydration properties were investigated in Tris. solution. Two kinds of $\alpha$-TCP powder samples were prepared; the one is reaction product of CaHPO4.2H2O and CaCO3, and another is that of hydroxyapatite(HAp) and $\beta$-Ca2P2O7. They were satisfied with Ca/P mole ratio 1.5 and were heated at 150$0^{\circ}C$ for 5 hours. In the hydration of $\alpha$-TCP samples the powder which was synthesized from HAp and $\beta$-Ca2P2O7 was hydrated faster than that from CaHPO4.2H2O and CaCO3. The hydration reaction of $\alpha$-TCP powder transformed rapidly into HAp accompanying setting and hardening. It was realized that the hydration reaction of $\alpha$-TCP was due to the solution-precipitation mechanism and the hydrates from the reaction were Ca-deficient HAp having funtional group HPO42-.

  • A study was carried out to investigate the control effect of sodium and potassium phosphates, sodium citrate and three different salts mixtures on kimchi fermentation when they were added into half-fermented kimchi in the concentration range of $0.001{\sim}0.01\;M$. The salts mixtures added were sodium phosphates mixture(CA-A), addition of $NaNO_2$, Ca-EDTA and BHA to CA-A(CA-B) and substitution of BHA with sodium citrate in CA-B. The results showed that sodium phosphates and sodium citrate significantly inhibited the kimchi fermentation while potassium phosphate had little effect. The order of control effect was $Na_3PO_4-Na_2HPO_4-sodium\;citrate-NaH_2PO_4-K_2HPO_4-KH_2PO_4$. Among the salts mixtures, CA-A showed the most reducing effect in the fermentation rate followed by CA-C and CA-A. The mixture of CA-C could extend the time of holding pH $4.2{\sim}4.4$ by approximately 6 times at $4{\sim}25^{\circ}C$ when it was compared to control. The microbial growth study of total and Leuconostoc mesenteroides also showed a very significant decrease in their numbers.

Effect of Ca^{++} and Cu^{++} Removal from Molasses on Yest cell Growth and RNA Accumulation. (당밀로부터 Ca^{++} 및 Cu^{++} 이온 제거가 효모 생육 및 RNA축적에 미치는 영향)

  • When Saccharomyces cerevisiae MTY62, a high-RNA content yeast, was cultivated by fed-batch mode feeding molasses and com steep liquor, the cell density less than 45g-DCW/L and the RNA content less than 140mg/g-cell were obtained, indicating that unknown compounds inhibiting the cell growth and RNA accumulation are contained in the molasses. Therefore, in order to obtain higher levels of cell density and RNA content, $Ca^{++}$, $Cu^{++}$and $K^{+}$ ions in molasses were removed by pretreatments of molasses with various agents such as IonClear BigBead, $Na_2$$HPO_4$, $H_2$$SO_4$, citric acid, $K_2$$HPO_4$, and EDT A. Among them, IonClear BigBead, $Na_2$$HPO_4$, and EDTA gave the highest $Ca^{++}$ removal efficiency of about 60-90%. In the batch culture with pretreated molasses, the cell concentration of 18.6g-DCW/I and RNA concentration of 3127 mg/I, maximum specific growth rate of 0.459$h^{-1}$ , and specific consumption rate of reducing sugar of 1.28g-sugar/g-cell-h were obtained, which are about 10%, 17%,47%, and 36% higher levels, respectively, over the batch culture with untreated molasses.

Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus (차가버섯의 균사체 성장에 대한 배지성분의 영향)

  • Effect of temperature($22-32^{\circ}C$), pH(5-7) and medium composition on the mycelial growth for the submerged culture of Inonotus obliquus. The concentrations of glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ and $CaCl_2$ were examined in the ranges of 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L and 0-0.5g/L, respectively. The maximum mycelial growth of Inonotus obliquus was obtained for $26-27^{\circ}C$ and pH 6. The concentrations of glucose, yeast extract and $CaCl_2$, which gave the maximum mycelial growth of Inonotus obliquus, were 70 g/L, 5 g/L and 0.1 g/L, respectively. In the cases of starch, peptone and $K_2HPO_4$, the mycelial growth of Inonotus obliquus increased with increasing the concentrations. However, as the concentration of $MgSO_4{\cdot}7H_2O$ increased, the mycelial growth of Inonotus obliquus decreased. The medium for maximum mycelial growth of Inonotus obliquus consisted of (per 1 L): glucose, 70 g; peptone, 5-20 g; starch, 10 g; yeast extract, 5 g; $K_2HPO_4$, 2 g and $CaCl_2$, 0.1 g.

Stabilization of Heavy Metals-contaminated Soils Around the Abandoned Mine area Using Phosphate (인산염을 이용한 휴.폐광산 주변 중금속 오염토양의 안정화처리에 관한 연구)

  • The objective of this study was to evaluate the efficiency of $(NH_4)_2HPO_4$, $Na_2HPO_4{\cdot}12H_2O$, $CaHPO_4{\cdot}2H_2O$, $Ca(H_2PO_4)_2{\cdot}H_2O$ and $H_3PO_4$ for the stabilization of soils contaminated with multi-metals containing Pb, Cd and As. The application rate of stabilizers to soils was determined based on $PO_4/Pb_{total}$ molar ratio of 0.5, 1, 2, 4. The results of Korea Standard Test and TCLP (EPA Method 1311) showed the reduction of metal leachabilities below the regulatory limits for Pb and Cd when $H_3PO_4$ and $Ca(H_2PO_4)_2{\cdot}H_2O$ were applied. However, stabilization efficiency for Cd was low and in case of As leaching concentration increased rather. It is considered that $PO_4$ reacted effectively $Pb^{2+}$ due to leaching Pb under low pH condition created by adding $H_3PO_4$. Accordingly Pb was stabilized by dissolution and precipitation of hydroxypyromorphite. From the change of metals fraction using sequential extraction procedure when $H_3PO_4$ applied as a stabilizer, we confirmed that residual fraction increased more than 60% and this result was accorded with XRD analysis that detected only hydroxypyromorphite peak in $H_3PO_4$.

Effect of Calcium Compounds from Oyster Shell Bouind Fish Skin Gelatin Peptide in Calcium Deficient Rats (어피 젤라틴 펩티드와 결합한 굴껍질 유래 칼슘 화합물이 칼슘 결핍 흰쥐에 미치는 영향)

  • To utilize the oyster shell containing a lot of calcium, we investigated the bioavailability of calcium compounds from oyster shell. First, calcium oxide was prepared by burning of oyster shell at $1200^{\circ}C$. Its purity was approximately $98.5\%$. Calcium compounds, $CaCl_2$, and $CaHPO_4$, from the calcium oxide were prepared by chemical reaction. Effect on calcium absorption by the calcium compounds from oyster shell was improved using fish skin gelatin peptides.(FSGP), which was prepared by enzymatic hydrolysis of fish skin gelatin for 4hr with tuna pyloric caeca crude enzyme (TPCCE). in vitro experiment, calcium absorption by addition of FSGP in a mixture solution of calcium and phosphate was higher approximately $70\%$ than that by control. in vivo using calcium deficient rats, a group taken the diets with $3\%$ FSGP and $CaHPO_4$ was significantly improved amount of calcium and ash in femur and strength of femur. These results suggest that calcium compounds from oyster shell and FSGP could be used as an effective dietary calcium source.

Isolation and physiological characteristics of cellulolytic bacteria (섬유소 분해세균의 분리 및 생리적인 특성)

  • Three hundred and one cellulolytic bacterial were isolated from the 148 screening sources such as decomposed wood, soil, compost and leaf mold. Among them, strain KL-6 was found to have the highest of cellulase activity, and identified as species belonged to the genus Cellulomonas. Strain KL-6 was decompose up to 90% of the filter paper (whatman No. 1) substrate within 50 hours, and showed the colony halo formation (11 cm). The activities of CMCase (67 unit/ml), FPase (70 unit/ml) and ${\beta}-glucosidase$ (0.68 unit/ml) were obtained when this strain was cultured for 50 hrs at $30^{\circ}C$. Glucose was not found in detectable amounts at the FP medium. The optimum composition of nutrient medium for the cell growth by strain KL-6 was sucrose 0.5%, yeast extract 0.1%, $(NH_4)_2HPO_4\;0.1%$, $K_2HPO_4\;0.1%$, $MgSO_4{\cdot}7H_2O\;0.01%$, $CaCl_2\; 0.01%$, NaCl 0.6%, $CaCO_3\;0.1%$ and the optimum pH and temperature were 7.0 and $30^{\circ}C$, respectively.

  • The formation and dissolution of hydroxides, carbonates and hydroxyapatite (HAp), which depend on the pH of solution, are important factor for the preparation of homogeneous and fine HAp, $Ca_{10-x}(HPO_4)_x(PO_4)_{6-x}(OH)_{2-x}(x=0)$, ceramic powder from the $Ca-PO_4-H_2O$ system. Since the solubility of each complex ion is a linear function of pH, the solubility diagram can be obtained by plotting the logarithmic molar concentrations calculated from the values of the equilibrium constants and solubility products for hydroxides, carbonates, and hydroxyapatite. The optimum pH condition for the formation of single phase $Ca_{10-x}(HPO_4)_x(PO_4)_{6-x}(OH)_{2-x}(x=0)$ powder in $Ca-PO_4-H_2O$ system at $25^{\circ}C$ was estimated as $10.5{\pm}0.5$ through the theoretical consideration. The HAp powder dried at $80^{\circ}C$ showed a fine agglomerated particles with a size of 75 nm. The HAp powder calcined at $1,000^{\circ}C$ consisted of nearly homogeneous particles with a size of 450 nm. Even though the dried HAp particles consisted of agglomeration, mechanical properties were superior due to fine microstructure after sintering.
