Plant Regeneration through Somatic Embryogenesis from Mature Zygotic Embryos of Ginseng(Panax ginseng C. A. Meyer) and Flowering of Plantlets (인삼(Panax ginseng C. A. Meyer)의 성숙한 배로부터 체세포 배발생을 통한 구분화 및 유식물체의 개화)

  • Mature zygotic embryos dissected from ginseng(Panax ginseng C. A. Meyer) seeds were cultured on Murashige and Skoog's (MS) medium containing various concentrations of 2, 4-dichlorophenoxyacetic acid(2, 4-D) and kinetin. Somatic embryos were induced directly from cotyledonary tissue or from intervening callus. The induction frequency of somatic embryos was up to 55%. Upon transfer to half-strength MS medium supplemented with 1 mg/1 6-benzyladenine(BA) and 1 mg/1 GA3, most somatic embryos developed into plantlets. Over 50% of the plantlets flowered after 4 weeks of culture and then a few bore immature fruits in vitro. Therefore, it is suggested that the juvenility of the ginseng tissue which give rise to somatic embryos does not interfere with in vitro flowering of their regenerated plantlets.

Factors Affecting Sporulation of a Mycoherbicide, Epicoccosorus nematosporus, on the Lesion of Eleocharis kuroguwai

  • Effects of temperature and dew period on sporulation of a mycoherbicide, Epicoccosorus nematosporus, on the lesion of its host, Eleocharis kuroguwai were determined. Conidia formation was first observed after 10 days on plants incubated for either 12 or 16 h in a dew chamber at 28$^{\circ}C$; 16 h dew period resulted in more conidia formation. As the dew period was decreased to less than 8 h, fewer conidia formed. Conidial production was most abundant at 28$^{\circ}C$ and produced as much as 3.3$\times$10$^4$conidia per lesion, while 0.1$\times$10$^3$and 2.3$\times$10$^3$conidia per lesion were produced at 16$^{\circ}C$ and 36$^{\circ}C$, respectively. Alternating temperature regimes, i.e., 30/15, 30/20, 28/20, and 28/15$^{\circ}C$ (day/night) were much better than constant temperature, i.e., 30/30, 28/28/, and 20/2$0^{\circ}C$ for sporulation. In the second sporulation, there were as much as 3.1$\times$10$^4$conidia per lesion (ca. <50% of the first sporulation). Then, sporulation dropped sharply to 6.2$\times$10$^2$conidia per lesion in the third sporulation. Results of this study suggest that temperature combined with dew period is the primary limiting factor in the use of E. nematosporus as a mycoherbicide off, kuroguwai.

Identification Factor Development of Particulate Matters Emitted from Coal-fired Power Plant by FE-SEM/EDX Analysis (FE-SEM/EDX 분석법을 이용한 석탄화력발전소에서 배출되는 입자상물질의 확인자 개발)

  • Coal-fired power plants emit various Particulate Matter(PM) at coal storage pile and ash landfill as well as the stack, and affect the surrounding environment. Field Emission Scanning Electron Microscopy and Energy Dispersive X-ray analyzer(FE-SEM/EDX) were used to develop identification factor and the physico-chemical analysis of PM emitted from a power plant. In this study, three samples of pulverized coal, bottom ash, and fly ash were analyzed. The pulverized coal was spherical particles in shape and the chemical composition of C-O-Si-Al and C/Si and C/Al ratios were 200~300 on average. The bottom ash was spherical or non-spherical particles in shape, chemical composition was O-C-Si-Al-Fe-Ca and C/Si and C/Al ratios were $4.3{\pm}4.6$ and $8.8{\pm}10.0$. The fly ash was spherical particles in shape, chemical composition was O-Si-Ai-C-Fe-Ca and C/Si and C/Al ratios were $0.5{\pm}0.2$ and $0.8{\pm}0.5$.

Molecular Characterization and Infectious cDNA Clone of a Korean Isolate of Pepper mild mottle virus from Pepper

  • A Korean isolate of Pepper mild mottle virus (PMMoV-Kr) was isolated from a diseased hot pepper plant and its biological and molecular properties were compared to that of PMMoV-J and PMMo V -So The genomic RNA of PMMoV-Kr consists of 6,356 nucleotides. The nucleotide and amino acid sequences identities of four viral proteins and two noncoding regions among PMMoV-Kr, PMMoV-S and PMMoV-J were $96.9\%\;to\;100.0\%\;and\;97.5\%\;to\;98.6\%$, respectively. Full-length cDNA amplicon of PMMoV-Kr was directly amplified by RT-PCR with a set of 5'-end primer anchoring T7 RNA promoter sequence and 3'-end virus-specific primer. Capped transcript RNAs from the full-length cDNA clone were highly infectious and caused characteristic symptoms of wild type PMMoV when mechanically inoculated to systemic host plants such as Nicotiana benthamiana and pepper plants.

Illustration of Disease Suppression of Anthracnose on Cucumber Leaves by Treatment with Chlorella fusca (오이 잎에서 Chlorella fusca 처리에 의한 오이탄저병 발생 억제 기작)

  • Chlorella is known as chlorophyceae which can live autotrophically by photosynthesis, promote the growth of plants and suppress some plant diseases. However, a few researches in inhibition mechanism of plant diseases by chlorella have been carried out. In this study cucumber leaves pre-treated with Chlorella fusca suspension were investigated whether anthracnose by Colletotrichum orbiculare is suppressed or not. Furthermore, in order to illustrate how the algae can restrain the antracnose, the infection structures of C. orbiculare were observed on the cucumber leaves pre-treated with the algae. Consequently, appressorium formation rate was apparently reduced in the cucumber leaves pre-treated with C. fusca compared to untreated control one. Also, the numbers of conidia found at the inoculation sites were significantly reduced compared to untreated one. On the other hand, on the leaves pre-treated with $Benomyl^{(R)}$ appressorium formation were decreased remarkably and numbers of conidia were also reduced similar with those pre-treated with C. fusca. Based on these results, it was revealed that occurrence of anthracnose can be suppressed by C. fusca pre-treatment and suggested that biochmical or structural hinderance by C. fusca resulting in the decline of appressorium formation on the leave surfaces may play an important role in the disease suppression.

Effects of Temperature and Light intensity on Growth and Yield of Condonopsis lanceolata (온도 및 光條件이 더덕의 생육과 수량에 미치는 영향)

  • The present study was performed to obtain a basic data of cultivation for Condonopsis lanceolata. Various temperatures, light, and DIF were treated during the whole plant growth. The growth of aerial part was most remarkable between $20^{\circ}C$ and $25^{\circ}C$, but inhibited by means of shading treatment. The fresh weight of subterranean part was heaviest(16.6g) at $15^{\circ}C$. Also, plant height increased when a constant temperature and +DIF were treated. Leaf width and length was not related to temperature and DIF treatment. The fresh weight of subterranean part increased in +DIF-treated sample at $25^{\circ}C$.

  • In $\lambda$-Zap II vector system, a cDNA library was constructed for the developing secondary xylem mRNA from hybrid poplar, Populus nigra x maximowiczii. A cDNA clone of 1.5 kb in size, pOMTB1.4 encoding a lignin-bispecific O-methyltransferase was screened by plaque hybridization using a probe of 540 bp cDNA amplified by polymerase chain reaction from the cDNA library and identified by nucleotide sequencing. Its nucleotide sequence contains one open reading frame of 366 amino acids. The deduced amino acid sequence in comparison with that of Populus tremuloides showed the differences of 9 amino acids and revealed 85-99% homology among alfalfa, poplar and aspen.

Estimation of nuclear DNA content of various bamboo and rattan species

  • We determined the nuclear DNA content (genome size) of over 35 accessions each of bamboo and rattan species from Southeast Asia. The 2C DNA per nucleus was quantified by flow cytometry. The fluorescence of nuclei isolated from the leaves and stained with propidium iodide was measured. The genome size of the bamboo species examined was between 2.5 and 5.9 pg DNA per 2C nucleus. The genome size of the rattan species examined ranged from 1.8 to 10.5 pg DNA per 2C nucleus. This information will be useful for scientists working in diverse areas of plant biology such as biotechnology, biodiversity, genome analysis, plant breeding, physiology and molecular biology. Such data may be utilized to attempt to correlate the genome size with the ploidy status of bamboo species in cases where ploidy status has been reported.

Effects of Shelf Temperature on the Quality of Red Pepper (Capsicum annuum L.) after Low Temperature Storage (홍고추의 저온저장 후 모의 유통온도가 품질에 미치는 영향)

  • The study was conducted to investigate quality changes of red pepper by different shelf temperatures after storage at $7^{\circ}C$ for 30 days. Red pepper fruits produced high volume of $C_2H_4$ and $CO_2$ when alternating temperatures exceeded $10^{\circ}C$. Total sugar contents were not different among all the treatments. In contrast, sucrose content decreased about half at higher shelf temperature conditions than at $7^{\circ}C$. Capsaicinoid content was not influenced by alternating temperature, but ascorbic acid content tended to decrease by high shelf temperature after storage at $7^{\circ}C$. Decay did not occur under $13^{\circ}C$, while it occurred in 22.2% of hits at $15^{\circ}C$ and 68.8% at room temperature, respectively. From this results, it is suggested that the optimum shelf temperature of red pepper after low temperature storage should be under $13^{\circ}C$ and that fruits of red pepper stored at $7^{\circ}C$ for 30 days could be sold within 10 days on the shelf.
