Objectives: Tanshinone I is a bioactive constituent in Salvia miltiorrhiza. At present, the anti-obesity effect and mechanism of tanshinone I are not fully understood. Here we investigated the effect of tanshinone I on lipid accumulation in 3T3-L1 preadipocytes and zebrafish. Methods: Lipid accumulation and triglyceride (TG) content in 3T3-L1 cells were determined by Oil Red O staining and AdipoRed assay, respectively. The expression and phosphorylation levels of adipogenic/lipogenic proteins in 3T3-L1 cells were evaluated by Western blotting. The messenger RNA (mRNA) expression levels of adipogenic/lipogenic markers and leptin in 3T3-L1 cells were measured by reverse transcription polymerase chain reaction (RT-PCR). Lipid accumulation in zebrafish was assessed by LipidGreen2 staining. Results: Tanshinone I at 5 μM largely blocked lipid accumulation and reduced TG content in differentiating 3T3-L1 cells. Furthermore, tanshinone I decreased the expression of CCAAT/enhancer-binding protein-α (C/EBP-α), peroxisome proliferator-activated receptor-γ (PPAR-γ), fatty acid synthase (FAS), acetyl CoA carboxylase (ACC), and perilipin A but also the phosphorylation of signal transducer and activator of transcription-3 (STAT-3) in differentiating 3T3-L1 cells. In addition, tanshinone I increased the phosphorylation of adenosine 3',5'-cyclic monophosphate (cAMP)-activated protein kinase (AMPK) while decreased the intracellular adenosine triphosphate (ATP) content with no change in the phosphorylation and expression of liver kinase-B1 in differentiating 3T3-L1 cells. Importantly, tanshinone I also reduced the extent of lipid deposit formation in developing zebrafish. Conclusions: These findings demonstrate that tanshinone I has strong anti-adipogenic effects on 3T3-L1 cells and reduces adiposity in zebrafish, and these anti-adipogenic effect in 3T3-L1 cells are mediated through control of C/EBP-α, PPAR-γ, STAT-3, FAS, ACC, perilipin A, and AMPK.

This study examined the effect of pretreated sandfish Arctoscopus japonicus meat as a surimi complex for preparing sandfish flavored fish paste. To prepare the sandfish-flavored paste, fine chopped sandfish meat including backbone was added in a ratio of 0 to 50 wt.% to thawed Alaska pollock Theragra chalcogramma surimi to make a mixed surimi gel. To prepare the sandfish-flavored paste, the mixed surimi was ground with salt using a silent cutter, mixed with starch and stabilizers 0.2% transglutaminase and gluconolactone 0.3%, stuffed in a rectangular container, left for 3 h at $25^{\circ}C$, cooked in hot water for 30 min at $90^{\circ}C$, and finally chilled for 20 min at $4^{\circ}C$. The effects of the pretreatment of sandfish meat were investigated by analyzing the quality of the paste produced. The proximate composition of FP (fish paste containing 40% steam-cooked sandfish meat and 0.3% gluconolactone) was moisture 76.1%, crude protein 12.0%, crude fat 3.8%, carbohydrate 6.1%, and ash 2.0%. The major minerals in FP were Na (23.77 mg/L), Mg (1.46 mg/L), Zn (1.04 mg/L), and Fe (0.41 mg/L), and the major free amino acids were taurine, anserine, alanine, and glutamic acid. The monounsaturated fatty acid content of FP was 566.22 mg%, and the polyunsaturated fatty acid content was 498.43 mg%. The n-3 fatty acid content was 398.01 mg%, and C20:5n-3 (218.85 mg %) was a major component.

The purpose of this study was to investigate the effects of garlic diet on blood coagulation in cholesterol-fed rats. 36 male rats were divided into six groups and fed experimental diet for three weeks. Two-levels (2% and 4%) of fresh and boiled garlic were supplemented to the stock diet with 2% cholesterol and 0.25% bile salt for the experimental diet. After the rats fed with experimental diets during 3 weeks, the whole blood coagulation time, the content of plasma fibrinogen, the fibrinolytic activity and the prothrombin time were measured. The results are summarized as follows; 1. Garlic diet have no influence on body weight and food consumption in rats. 2. In the case of cholesterol-fed group, the whole blood coagulation time was decreased 24.3% compared with the normal group(A). But garlic diet group(C) was similar to the normal group(A) and groupD-F was increased 4.9-11.5%. 3. The content of plasma fibrinogen of cholesterol-fed group was higher 114.5% than that of normal gropu(A). In the garlic diets(C-F) the contents of plasma fibrinogen were increased 33.1-63.8%. 4. The fibrinolytic activity of cholesterol diet was decreased 16.2% compared with the normal group(A) but was increased 10.9-44.5% compared to the garlic diets(C-F). 5. The prothrombin time of cholesterol-fed group was decreased 8.9% compared with normal group(A) but was increased 8.0-46.3% in garlic diets(C-F). As mentioned above, the garlic diets increased whole blood coagulation time, prothrombin time and fibrinolytic activity and decreased the content of fibrinogen. For that reason it is thought that the garlic diets have a anticoagulation effect regulating the fibrinogen synthesis in liver.

We investigated the effect of paclobutrazol (PB) and prohexadione-Ca (PC) on seed yield, and the content of oils and endogenous gibberellin in flax grown in a greenhouse. Flax seedlings were cultivated in a greenhouse with average ambient temperature ranging from $35^{\circ}C$ to $38^{\circ}C$ during the growing period, and then PB and PC were applied on the leaves. The height of flax plants was reduced significantly by the foliar application of PB and PC. The highest ripening seed rate recorded was 94.5% with 2000 ppm PC; a 10.3% increase compared with that of the control. Both PB and PC markedly increased the fructification of flax seed. The application of two plant growth retardants resulted in higher seed production in flax seedlings grown in open field than in those grown in a greenhouse. The highest oil content was observed in flax treated with PC, whereas, the lowest oil content was observed in flax treated with PB. The oil content and ratio of unsaturated fatty acids were higher in flax seedlings grown in greenhouse than in those grown in open field. The level of two endogenous bioactive gibberellins ($GA_1$ and $GA_4$) in flax seeds decreased with the application of PB and PC. The flax plants treated with PB and PC presented higher secoisolariciresinol diglucoside content than that of the control. The optimal concentration of PC was found to be 2000 ppm. In conclusion, the foliar application of 2000 ppm PC might be useful in increasing oil content and seed production in flax plants.

Although classical metabolic engineering strategies have succeeded in developing microbial strains capable of producing desired bioproducts, metabolic imbalance resulting from extensive genetic manipulation often leads to decreased productivity. Thus, abiotic strategies for improving microbial production performance can be an alternative to overcome drawbacks arising from intensive metabolic engineering. Herein, we report a promising abiotic method for enhancing lycopene production by UV-C irradiation using a radiation-resistant ΔcrtLm/crtB+dxs+ Deinococcus radiodurans R1 strain. First, the onset of UV irradiation was determined through analysis of the expression of 11 genes mainly involved in the carotenoid biosynthetic pathway in the ΔcrtLm/crtB+dxs+ D. radiodurans R1 strain. Second, the effects of different UV wavelengths (UV-A, UV-B, and UV-C) on lycopene production were investigated. UV-C irradiation induced the highest production, resulting in a 69.9% increase in lycopene content [64.2 ± 3.2 mg/g dry cell weight (DCW)]. Extended UV-C irradiation further enhanced lycopene content up to 73.9 ± 2.3 mg/g DCW, a 95.5% increase compared to production without UV-C irradiation (37.8 ± 0.7 mg/g DCW).
In this study, the differences in the contents of vitamin C and minerals in perilla leaves were compared according to the age of leaf and storage conditions. The content of vitamin C in perilla leaves the younger the perilla leaves, the higher the content of vitamin C: Vitamin C content of mature leaf at the lowest part of the stem was 63.7mg/100g, and that of young leaf at the top of the stem was 129.0mg/100g. However, the contents of Ca and Fe by the leaf age were in an opposite tendency with vitamin C: Ca content of the leaves at the lowest and the top part of the stem were 449.4 mg and 311.8mg/100g, respectively. But, the contents of Na and Mg were poor in mature leaf, and the content of K showed no particular difference according to leaf age. When stored at 5$^{\circ}C$, residual rate of vitamin C content was 92.56, 81.52 and 77.05%, after stored 1, 5 and 10 days, respectively. In the case of 20$^{\circ}C$, residual rate of vitamin C was 85.80, 79.90 and 72.53%, respectively. When stored at 5$^{\circ}C$, Ca content in perilla leaves was in the range of 348.93∼408.81mg/100g, and at 20$^{\circ}C$, the content of Ca was 360.26∼392.25mg/100g. Storage time and temperature did not make a significant difference in the mineral content of perilla leaves.
The effects of growth temperature and nutritional components on the synthesis of poly-3-hydroxybutyric acid, PHB, by filamentation-suppressed recombinant Escherichia coli XL1-Blue (pSYL107) were studied. After culturing XL1-Blue(pSYL107) for 48 hours in complex medium at 30$\circ$C, 7Al g/l of PHB could be obtained with the PHB content and PHB yield of 82% and 0.371 g PHB/g glucose, respectively. Lower concentration of PHB(3.2 g/l) was obtained when cultu- red at 37$\circ$C, which seemed to be due to the instability of this strain having amplified FtsZ activity. The PHB concentration of 3.75 g/l was obtained after culturing 60 hours in R medium supplemen- ted with 20 g/l glucose at 30$\circ$C, which was more than twice higher than that obtained with XL1-Blue(pSYL105). This suggested that the enhancement of PHB synthesis by suppressing filamenta- tion was more significant in a defined medium than complex medium. PHB synthesis could be further enhanced by supplementing a small amount of various complex nitrogen sources. When 5 g/l of beef extract was added to a defined medium, PHB concentration, PHB content, and PHB yield obtained after 60 hours of cultivation at 30$\circ$C were 7.46 g/l, 86%, and 0.375 g PHB/g glucose,respectively.
Cation exchange polymer electrolyte membrane for the application of direct methanol fuel cell (DMFC) was studied. Poly(vinyl alcohol)(PVA) well known as a methanol barrier in pervaporation separation was used fur the base materials and poly(acrylic acid)(PAA) was used for the crosslinking agent with various concentration. Methanol permeability, ion conductivity, ion exchange capacity, water contents and fixed ion concentration of the membranes were investigated to evaluate the performance of the fuel cell electrolyte membrane. Methanol permeability and ion conductivity of the membranes were decreased with increasing PAA content and were increased over 15% of PAA content. These phenomena would be explained with the introduction of hydrophilic crosslinking agent. The membranes with 15% content of PAA showed methanol permeability of $6.49{\times}10^{-8}/cm^2/s,\; 2.85{\times}10^{-7}CM^2/s$ at $25^{\circ}C,\; 50^{\circ}C$ of operating temperatures, respectively. ion conductivities of the membrane were $2.66{\times}10^{-3}\;S/cm,$$9.16{\times}10^{-3}\;S/cm$ at $25^{\circ}C,\; 50^{\circ}C$ of operating temperatures, respectively. ion exchange capacity, water content and fixed ion concentration of the membrane were revealed 1.32 meq/g membrane,0.25 g $H_2$O/g membrane and 5.25 meq/g $H_2O$, respectively.

This study investigated the effect of heat treatment conditions on the quality and antioxidant activities of Korean traditional actinidia (Actinidia arguta) cultivars puree. Heat treatment on actinidia puree was conducted at $70{\sim}90^{\circ}C$ for 1 min to 5 mins, while a control sample of the actinidia puree was prepared without heat treatment. In all the samples, except for the sample treated at $90^{\circ}C$ for 5 mins, pH and Brix degree was not dramatically changed. Pulp content decreased with increasing temperature and time. L and the a value of color increased with increasing temperature and time. However, the b value showed a reverse tendency with L and the a value. Viscosity and gumminess increased as heat temperature and time increased, though cohesiveness was not significantly changed depending on temperature and time. Coliform and yeast were not detected on all samples and the number of aerobic bacteria and mold decreased as temperature and time increased. Vitamin C and total phenolic content of the puree was not changed for 1, 3, and 5 mins at $70^{\circ}C$ and for 1 and 3 mins at $80^{\circ}C$, but significantly decreased with increasing time at $90^{\circ}C$. Furthermore, the antioxidant activities of the puree, such as DPPH radical scavenging activity, ABTS radical scavenging activity, and FRAP (ferric reducing antioxidant power) showed a similar tendency with total phenolic content.

In this study, we investigated the amino acids content of shiitake mushrooms grown in eight different media and under three different temperature conditions. The sawdust media were supplemented with 4 kinds of nutrients - rice bran, barley, bean curd refuse, and red ginseng refuse in an 8:2 ratio with or without 0.5% calcium chloride, and the fruiting temperature was set to low ($13-15^{\circ}C$), medium ($18-20^{\circ}C$), or high temperatures ($24-26^{\circ}C$). Seventeen amino acids were detected, and the content varied depending on the compositions of the media. The highest total amino acid content (209.7 mg/g) was measured in mushrooms from the media with red ginseng refuse, and the glutamic acid content was the highest (51.3 mg/g) in mushrooms from the media with barley. In terms of fruiting temperature, the amino acid content was the highest (38.2%) at medium temperature followed by the content at low (32.5%) and high temperature (29.3%). The amino acid composition of shiitake mushrooms was similar between the medium and low temperature conditions.
