Expression of CD40, CD86, and HLA-DR in CD1c+ Myeloid Dendritic Cells Isolated from Peripheral Blood in Primary Adenocarcinoma of Lung (원발성 폐선암환자의 말초혈액에서 분리한 CD1c+ 골수성 수지상 세포에서의 CD40, CD86 및 HLA-DR의 발현)

  • Background: There have been several reports using animal experiments that CD1-restricted T-cells have a key role in tumor immunity. To address this issue, we studied the expression of markers for CD1c+ myeloid dendritic cells (DCs) isolated from peripheral blood in the clinical setting. Material and Method: A total of 24 patients with radiologically suspected or histologically confirmed lung cancer who underwent pulmonary resection were enrolled in this study. The patients were divided according to histology findings into three groups: primary adenocarcinoma of lung (PACL), primary squamous cell carcinoma of lung (PSqCL) and benign lung disease (BLD). We obtained 20 mL of peripheral venous blood from patients using heparin-coated syringes. Using flow-cytometry after labeling with monoclonal antibodies, data acquisition and analysis were done. Result: The ratio of CD1c+CD19- dendritic cells to CD1c+ dendritic cells were not significantly different between the three groups. CD40 (p=0.171), CD86 (p=0.037) and HLA-DR (p=0.036) were less expressed in the PACL than the BLD group. Expression of CD40 (p=0.319), CD86 (p=0.036) and HLA-DR (p=0.085) were less expressed in the PACL than the PSqCL group, but the differences were only significant for CD86. Expression of co-stimulatory markers was not different between the PSqCL and BLD groups. Expression of markers for activated DCs were dramatically lower in the PACL group than in groups with other histology (CD40 (p=0.005), CD86 (p=0.013) HLA-DR (p=0.004). Conclusion: These results suggest the possibility that CD1c+ myeloid DCs participate in control of the tumor immunity system and that low expression of markers results in lack of an immune response triggered by dendritic cells in adenocarcinoma of the lung.

Deuterium oxide stabilizes conformation of tubulin: a biophysical and biochemical study

  • The present study was aimed to elucidate the mechanism of stabilization of tubulin by deuterium oxide ($D_2O$). Rate of decrease of tryptophan fluorescence during aging of tubulin at 4$^{\circ}C$ and 37$^{\circ}C$ was significantly lower in $D_2O$ than in $H_2O$. Circular dichroism spectra of tubulin after incubation at 4$^{\circ}C$, suggested that complete stabilization of the secondary structure in D2O during the first 24 hours of incubation. The number of available cysteine measured by DTNB reaction was decreased to a lesser extent in $D_2O$ than in $H_2O$. . During the increase in temperature of tubulin, the rate of decrease of fluorescence at 335 nm and change of CD value at 222 nm was lesser in $D_2O$. Differential Scanning calorimetric experiments showed that the $T_m$ values for tubulin unfolding in $D_2O$ were 58.6$^{\circ}C$ and 62.17$^{\circ}C$, and in $H_2O$. those values were 55.4$^{\circ}C$ and 59.35$^{\circ}C$.

Effects of Settings in Dynamic Ranges and Frequency Modes on Ultrasonic Images (초음파 영상에서 동적영역과 주파수 방식의 설정에 따른 효과)

  • It is important to get clinical ultrasonic images of good quality for accurate diagnosis. In this study, it observed the change of ultrasonic images against setting frequency, dynamic range(DR) and type of probes on ultrasonic scanner. In the experiment it evaluated image of LCS (Low Contrast Sensitivity) targets(-15, -6, -3, +3, +6, +15 dB) of a standard ultrasonic test phantoms(539,551, ATS, USA) similar to solid and cystic lesions. Its imaged from convex (C3-7IM) and linear probe (L5-12IM) on SA-9900 (Medison Ltd, Korea) scanner. The images obtained altering the setting parameters which are frequency(gen, pen, res, harmonic) mode and DR($40{\sim}100\;dB$). The quality of images evaluated compare with the nominal LCS value of target and measured LCS value. The results show that there was no significant changing of quality images altering DR 40, 60, 80, 100 dB against frequency in Convex probe but the image being the highest in LCS target at DR 60 dB, harmonic of frequency mode in the -15 dB target close to cystic lesion. In Linear probe, DR 40 dB, harmonic mode at -15 dB LCS target close to nominal value. It discussed necessity of evaluation about ROC(Receiver Operating Characteristic) from the psychological viewpoint and limit of evaluation from quantified images.

Studies on Thermophilic Flat-sour Bacteria in Soymilk: Isolation, Indentification and Determination of Heat Resistance (두유내의 고온성 Flat-sour 변질균의 분리.동정 및 열저항성에 관한 연구)

  • For the optimization of thermal processing conditions in soymilk process, 4 strains of thermoresistant flat-sour bacteria were isolated from soymilk. The isolates were aerobic spore-forming rods, and grew at $-65^{\circ}C$. Based on the morphological and physiological properties, all of the isolated strains were identified as Bacillus stearothermophilus. The heat resistance of spores of 3 isolates and Bacillus stearothermophillus ATCC 12980 as a reference was determined in soymilk(pH 7.0) and pH 7.0 buffer solution. For each of the spores studied, linear regression equations with standard error were presented for the thermal destruction at 110, 115, 121, and $125^{\circ}C$. It was not obvious that the components of soy milk increased the heat resistance of spores. Between the strains studied, variability was noted in the D values at the different temperature, and no one strain was consistently the most heat resistant at all the given temperatures. The average D value for the 4 strains was 77.27, 20.20, 2.76 and 1.39 min at 110, 115, 121 and $125^{\circ}C$, respectively, and the average z value was $8.36^{\circ}C$.

Inhibition of Cyclooxygenase and Prostaglandin E2 Synthesis by Crude Methanolic Extract from Euonymus Alatus (Thunb.) Sieb in SKBR3 Human Breast Cancer Cell Line

  • In the present study, we examined the effect of crude methanolic extract (CME) from Euonymus alatus (Thunb.) Sieb on arachidonic acid (AA) cascade in SKBR3 human breast cancer cell line. CME had a potent inhibitory activity of prostaglandin E2 (PGE2) release induced by A23187, a $Ca^{2+}$ ionophore. The inhibition was concentration-dependent, with the 50 value of about 5 M. CME had no inhibitory effect on A23187-induced phosphorylation of p42/p44 extracellular signal regulated kinase/mitogen-activated protein kinase or on the liberation of [14C]-AA from the cells labeled with [14C]-AA. However, CME concentration-dependently inhibited the conversion of AA to $PGE_2$ in microsomal preparations, showing its possible inhibition of cyclooxygenase (COX). In enzyme assay in vitro, CME inhibited the activities of both constitutive COX (COX­I) and inducible COX (COX-2) in a concentration-dependent manner, with the 50 values of about 0.8 and 2M, respectively. Lineweaver-Burk plot analysis indicated that CME competitively inhibited the activities of both COX-l and -2. This study is a first demonstration that CME directly inhibits COX activity.

  • Using KUCCS, which enables real-time monitoring and controlling, the various boundary condition and in-situ field stress condition was simulated, to derive the correlation among ${{\sigma}_v}'-Dr-N$in domestic sandy soils. Soil specimens, having various relative density and confined stress, were formulated to evaluate N-value from the SPT. and Pile Driving Analyzer, PDA, was employed as a measuring device for the energy transfer efficiency in the rod. From the quantitative analysis of N-value, the correlating equation, $N_{60}/{D_r}^2=16.35+14.45{{\sigma}_v}'$ was obtained on the basis of Skempton's method(1986). More reliable soil parameters can be obtained from the N-value by using this study which considered regional characters and the correlation among ${{\sigma}_v}'-Dr-N$.

Physicochemical Composition of Buckwheat Microgreens Grown under Different Light Conditions (다른 광조건 하에서 재배된 메밀 새싹채소의 이화학적 특성)

  • As consumers interest in microgreens is increasing worldwide, the production of leafy microgreens uisng different LED lights was investigated in this study. The experiment was carried out to evaluate the effects of different LED lights on the composition and vitamin C contents of buckwheat microgreens. Physicochemical properties of buckwheat microgreens grown under different lights (red, blue, and white) and control exposed to a dark room were investigated. Moisture contents of buckwheat microgreens were 95.65% under white light (WL), 95.75% under blue light (BL), 90.77% under red light (RL), and 89.71% under dark room (DR). Crude ash contents of buckwheat microgreens grown under WL, DR, RL, and BL were 0.39%, 0.39%, 0.31%, and 0.37%, respectively. Crude protein contents of buckwheat microgreens grown under DR, RL, WL, and BL were 7.12%, 7.81%, 1.60%, and 2.40%, respectively. Crude fat contents of buckwheat microgreens grown under DR, BL, RL, and WL were 1.12%, 0.54%, 0.35%, and 0.22%, respectively. $^{\circ}Brix$ was the highest in microgreens grown under BL and RL and the lowest in buds grown under DR. Vitamin C content was the highest in buds grown under WL and the lowest in buds grown under BL. Total chlorophyll content was the highest in microgreens grown under RL and the lowest in buds grown under WL. For mineral content measurement of buckwheat microgreens, Ca, K, Mg, and P contents were high whereas B, Cu, and Zn contents were not detected. The mineral contents of buckwheat microgreens according to each color of light showed significant differences. These results demonstrated that treatment of different colored LED lights during cultivation was able to increase vitamin C content up to affecting the nutritional value of buckwheat microgreens.

Dyeing Properties on Polylactic Acid (PLA) Fabrics by Disperse Dyes (분산염료에 의한 PLA 직물의 염색성)

  • This study optimizes a suitable dyeing method for polylactic acid (PLA) fabrics using disperse dyes. For this, disperse red 60 (DR 60), disperse blue 56 (DB 56), and disperse yellow 54 (DY 54) were used and dyed on PLA fabrics dependent of dyeing temperature and time. The fastness of PLA fabrics dyed with three disperse dyes were evaluated; in addition, dye exhaustion, color strength (K/S value), and colorimetric properties of PLA fabrics were compared with PET fabrics. The experiments indicated optimum dyeability of PLA fabrics with disperse dyes. The dyeing temperature was $90^{\circ}C$ for every dye and the dyeing time were 20 min, 60 min, and 40 min for DR 60, DB 56, DY 54, respectively. PLA fabrics had good color fastness to washing, dry cleaning fastness, hot pressing fastness, rub fastness, and perspiration fastness by DR 60, DB 56, and DY 54. The dye exhaustion of PLA fabrics were lower than PET fabrics; however, K/S values were higher than PET fabrics.

Expression and Characterization of a Novel Deoxyribose 5-Phosphate Aldolase from Paenibacillus sp. EA001

  • A novel deoC gene was identified from Paenibacillus sp. EA001 isolated from soil. The gene had an open reading frame (ORF) of 663 base pairs encoding a protein of 220 amino acids with a molecular mass of 24.5 kDa. The amino acid sequence was 79% identical to that of deoxyribose 5-phosphate aldolase (DERA) from Geobacillus sp. Y412MC10. The deoC gene encoding DERA was cloned into an expression vector and the protein was expressed in Escherichia coli. The recombinant DERA was purified using Ni-NTA affinity chromatography and then characterized. The optimum temperature and pH of the enzyme were $50^{\circ}C$ and 6.0, respectively. The specific activity for the substrate deoxyribose 5-phosphate (DR5P) was $62\;{\mu}mol/min/mg$. The $K_m$ value for DR5P was determined to be 145 mM with the $k_{cat}$ value of $3.2{\times}10^2/s$ from Lineweaver-Burk plots. The EA001 DERA showed stability toward a high concentration of acetaldehyde (100 mM).

Are Biomarkers Predictive of Anthracycline-Induced Cardiac Dysfunction?

  • Background: The early detection of anthracycline- induced cardiotoxicity is very important since it might be useful in prevention of cardiac decompensation. This study was designed with the intent of assessing the usefulness of cardiac troponin T (cTnT) and NT- Pro BNP estimation in early prediction of anthracycline induced cardiotoxicity. Materials and Methods: In this prospective study histologically proven breast cancer patients who were scheduled to receive anthracycline containing combination chemotherapy as a part of multimodality treatment were enrolled. Baseline cardiac evaluation was performed by echocardiography (ECHO) and biomarkers like cardiac troponin T (cTnT) and N terminal- pro brain natriuretic peptide (NT- Pro BNP). All patients underwent cTnT and NT- Pro BNP estimation within 24 hours of each cycle of chemotherapy and were followed up after 6 months of initiation of chemotherapy. Any changes in follow up ECHO were compared to ECHO at baseline and cTnT and NT- Pro BNP levels after each cycle of anthracycline-based chemotherapy. Results: Initial data were obtained for 33 patients. Mean change in left ventricular diastolic diameter (LVDD) within 6 months was $0.154{\pm}0.433cms$ (p value=0.049). Seven out of 33 patients had an increase in biomarker cTnT levels (p value=0.5). A significant change in baseline and follow up LVDD was observed in patients with raised cTnT levels (p value=0.026) whereas no change was seen in ejection fraction (EF) and left atrial diameters (LAD) within 6 months of chemotherapy. NT- Pro BNP levels increased in significant number of patients (p value ${\leq}0.0001$) but no statistically significant change was observed in the ECHO parameters within 6 months. Conclusions: Functional monitoring is a poorly effective method in early estimation of anthracycline induced cardiac dysfunction. Estimation of biomarkers after chemotherapy may allow stratification of patients in various risk groups, thereby opening window for interventional strategies in order to prevent permanent damage to the myocardium.
