Fatty Acid constituents and Relative Compositions of Reproductive Tract Fluids in Korean Native Cows (한우에 있어서 생식기관액 중 지방산 조성과 조성율)

  • To investigate fatty acid constituents and relative compositions in the fluid of the follicles, oviducts, uterine body and uterine body in Korean native cow, the fluids of the reproductive tract were analyzed using gas chromatography. The samples were taken from various reproductive tract of 23 Korean native cows. q. Caprylic acid (C8:0), myristic acid(C14:0), palmitic acid(C16:0), palmitoleic acid(C16:1), stearic acid(C18:0), oleic acid(C18:1), linoleic acid(C18:2), arachidonic acid(C20:4) were found in the reproductive tracts of the cows, which made 8 kinds of fatty acid in total. 2. Palmitic acid, oleic acid, stearic acid were predominant with 35.67%, 24.98% and 17.52%, respectively. while low levels of fatty acids(<5%) were myristic acid, palmitoleic acid and caprylic acid with 1.75%, 1.28% and 2.69%, respectively. 3. Two kinds of polyunsaturated fatty acids, linoleic acid arachidonic acid were found in the reproductive tracts of cows. 4. Palmitic acid among saturated fatty acids and oleic acid among unsaturated fatty acids were the highest level in all of the reproductive tracts. 5. The Highest level of arachidonic acid was found in the uterine horn. 6. The sum of the palmitic acid and oleic acid were 61.72%, 63.72%, 57.66% and 57.65% for the fluid of follicle, oviduct, uterine horn and uterine body of the cows, respectively. 7. The relative compositions of palmitic acid, palmitoleic acid and caprylic acid were higher during the luteal phase than during the follicular phase. 8. The relative compositions of arachidonic acid was higher during the follicular phase in the fluid of uterine horn and uterine body of the cows. 9. The long chain fatty acid, the palmitic acid, stearic acid, oleic acid and linoleic acidshowed higher relative compositions during the follicular phase(86.49%∼95.51%) than during the luteal phase(85.64%∼88.93%).

  • The different developmental stage embryos and oviductal and uterine fluids of rabbit were analyzed by gas chromatography. Myristic (C 14:0), palmitic (C 16:0), palmitoleic (C 16:1), stearic (C 18:0), oleic (C 18:1), linoleic (C 18:2), linolenic (C 18:3), arachidic (C 20:0), arachidonic (C 20:4), docosahexaenoic (C 22:6) and lignoceric (C 24:0) acids were the common fatty acid constituents with little exception. In most of the samples palmitic, oleic, linoleic and arachidonic acids were observed in high concentration. Moreover, linoleic, linolenic and arachidonic acids were the three poly-unsaturated fatty acids in both type sample except day-1 oviductal fluids. Similarly, in both day-1 and day-2 oviductal and uterine fluids myristic, palmitoleic, stearic, linolenic, arachidic and docosahexaenoic acids were in less composition or undetected.

  • The fatty acid compositions of oocytes, follicular, oviductal and uterine fluids of pig and cow were analyzed using gas chromatography. Myristic (C 14: 0), palmitic (C 16: 0), palmitoleic (C 16: 1), stearic (C 18 : 0), oleic (C 18: 1), linoleic (C 18: 2), linolenic (C 18: 3) and arachidonic (C 20: 4) acids were identified as the common fatty acid constituents with little exception. Oleic acid composition was the highest (21.90 to 36.24%) in both pig and cow followed by palmitic (18.61 to 31.90%) and stearic (10.34 to 20.39%) acid. The three polyunsaturated fatty acids like linoleic, linolenic and arachidonic acids were detected in both pig and cow reproductive fluid samples. Myristic acid was not detected in pig oviductal fluid. Similarly, in cow oocytes myristic, palmitoleic and linolenic acids were not detected. Moreover, palmitic, stearic, oleic and linoleic acid comprised about 80% (73.74 to 88.00%) of the total fatty acids in the different samples analyzed in both animals.

Physicochemical Compositions of Raw and Dried Wolha Persimmons (월하시 생감 및 곶감의 이화학적 성분 분석)

  • This study analyzed the major chemical components of raw and dried persimmons (Diospyros kaki Thunb.). Raw and dried persimmons contained (respectively) 85.52% and 47.36% moisture, 0.10% and 0.13% crude fat, 0.30% and 1.89% crude protein, and 0.56% and 2.0% crude fiber. The main free sugar components in both raw and dried persimmons were glucose and fructose. Seventeen amino acids were identified in the dried persimmons, amongst which the dominant ones were aspartic acid, glutamic acid, histidine and arginine. The total amino acid content of raw and dried persimmons was 3,130.76 ppm and 12,849.33 ppm, respectively. The major fatty acids in total lipids were palmitic acid, palmitoleic acid and linolenic acid in both raw and dried persimmons. The raw persimmons had 23.22% palmitoleic acid and 32.70% linolenic acid, suggesting that they have a high ratio of unsaturated fatty acids. The mineral content of both raw and dried persimmons was Na < Fe < Ca < P < K.

  • The total amount of lipid content in the mullet roe is 20.5%, and iodine value and unsaponifiable matters content are 118 and 38.7%, respectively. The lipid composition of the muscle, roe and liver of the mullet, Mugil cephalus, shows differences. Triglyceride, wax esters, and free fatty acids are mainly contained in the lipids of the muscle, roe and liver, respectively. The mullet roe lipids are mainly composed of 59.1% of wax esters with a trace of sterol esters, 26.9% of polar lipids with pigments, 9.0% of triglyceride plus a trace of free fatty alcohols and fatty acids, and 3.0% of sterol contaminated with a trace of fatty alcohols. The major fatty acids of wax esters are C16 : 0, 47.5%, C18 : 1, 23.0%, C16 : 3, 6.5%, C20 : 5, 4.0%, those of triglyceride are C16 : 1, 25.1%, C18: 1, 16.7%, C16 : 0, 16.3%, C22 : 1, 7.9%, C18 : 0, 5.5%, C22 : 6, 4.4%, and those of polar lipids are C16 : 0, 35.0%, C18 : 1, 24.7%, C16 : 1, 6.1%, C20 : 5, 5.3%, C22 : 6, 4.2%. The major alcohols of wax esters are 51.0% of cetyl alcohol, 18.2% of palmitoleyl alcohol, and 10.7% of oleyl alcohol, and considerable amounts of odd-numbered alcohols such as C15 : 0, C15 : 1, C17 : 0, C17 : 1 and C19 : 1 are also found.

Seasonal Variation in Body Composition in Boleophthalmus pectinirostris Collected from Gangjin, Jeonnam, Korea (전남 강진산 짱뚱어(Boleophthalmus pectinirostris) 체성분의 계절변화)

  • This study was carried out to investigate seasonal changes in body composition in Boleophthalmus pectinirostris in Korea. B. pectinirostris collected in Gangjin, Jeonnam, consisted of 58.7-75.8% moisture content, 16.1-17.6% protein, 0.8-3.4% crude lipids, and 1.3-3.6% crude ash. Livers consisted of 15-17% moisture content, 14.0-16.8% crude protein, 34.4-71.2% crude lipids, and 0.5-1.2% crude ash. Calcium, iron, potassium, etc., content in B. pectinirostris muscle averaged 267.5-599.8 mg/100 g, 1.98-28.3 mg, 160.7-327.9 mg, 20.6-60.2 mg, 0.4-2.2 mg, 0.23-0.46 mg, 66.5-192.9 mg, and 1.32-3.8 mg, respectively. (Please clarify: the list of measured values must correspond directly with what was measured, not just "etc.") The major amino acids in B. pectinirostris muscle, in proportional order, were glutamic acid, isoleucine, and leucine; whereas the major amino acids in the liver were glutamic acid, lysine, aspartic acid, and leucine. The major saturated fatty acids in the muscle were palmitic acid (15-19%) and stearic acid (8-11%). The major monoenes and polyenes were palmitoleic acid (C16:1) and oleic acid (C18:1), and EPA and DHA, respectively. The major saturated fatty acids and polyenes in the liver were the same as in muscle, but the monoenes were palmitoleic acid (C16:1) and cis-10-hepta-decenoic acid (C17:1). Seasonal changes in B. pectinirostris body composition may be attributable to differential accumulation of fat and nutrients in comparison to the spawning and hibernation period.

Evaluation of Three Candidate Genes Affecting Fatty Acid Composition in Pigs

  • The association of three candidate genes, fatty acid synthase (FASN), microsomal triglyceride transfer protein (MTTP) and fatty acid binding protein 3 (FABP3), with fatty acid (FA) composition in Duroc pigs was investigated. Identified single nucleotide polymorphisms (SNPs) were used for polymerase chain reaction-restriction fragment length polymorphism genotyping. The c.265C>T SNP of FASN gene was significantly associated with high levels of palmitoleic acid (C16:1) (p<0.05), oleic acid (C18:1) (p<0.01), and mono-unsaturated fatty acid (MUFA) (p<0.01), but low levels of linoleic acid (C18:2) (p<0.01), alpha linolenic acid (C18:3) (p<0.05), and poly-unsaturated fatty acid (PUFA) (p<0.01) in animals having the CT genotype. The c.2573T>C SNP in the MTTP gene had a significant effect only in elevating the level of palmitoleic acid (C16:1) (p<0.05) in heterozygote animals. The polymorphism in FABP3 showed no significant effects on any fatty acid composition traits. These results suggest that the identified SNPs in the FASN and MTTP genes can be useful markers for selecting Duroc pigs having desirable healthy fatty acid composition.

Characteristics of a New Obligate Methanol-Oxidizing Bacterium (새로운 절대 메탄올 산화세균의 분리 및 특성)

  • A new methyltrophic bacterium which utilizes methanol as a sole source of carbon and energy was isolated from soil. It was Gram-negative, nonmotile, nonspore-forming rod, and strictly aerobic bacterium. Catalase and oxidase activities were present. Nitrate was reduced to nitrite. Vitamins and other growth factors were not required. Generation time was 1.6 hr under the optimal condition. The isolate assimilated methanol via the ribulose mono-phosphate pathway (Enter-Doudoroff varient) and did not have .alpha.-ketoglutarate dehydrogenase. It assimilated ammonia through glutamate dehydrogenase. The guanine plus cytosine content of the DNA was 61.0 mol%. The celular fatty acid composition was primarily straight-chain saturated $C^{16 : 0}$ acids (palmitic acids) and unsaturated $C_{16 :1}$ acid (palmitoleic acids), and the isolate also contained two unidentified $C_{17}$ branched fatty acids. The major ubiquinone was Q-8, and Q-6 and Q-7 were present as minor components. Phosphatidylethanolamine and phosphatidylglycerol were predominantly present, and diphosphatidyglycerol was also detected. Based on the physiological and biochemical properties, the isolate was assigned to a novel species of the genus Methylobacillus, Methylobacillus methanolovorus sp. nov.

  • Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.

  • To investigate fatty acid constituents and relative compositions in the fluid of the follicles, oviducts, uterine ho군 and uterine body in sows, the fluids of the reproductive tract were analyzed using Gas chromatography. The samples were taken from various reproductive tract of 21 sows slaughtered. 1. Caprylic acid(C8: 0), capric acid(C10:0), lauric acid(C12:0), myristic acid(C14:0), palmitic acid(C16:0), plamitolele acid(C16:1), stearic acid(C18:0), oleic acid(C18:1), linoleic acid(C18:2) and arachidonic acid(C20:4) were found in the reproductive tracts of the sows, which made 10 kinds of fatty acid intotal. 2. Two kinds of polyunsaturated fatty acids, linoleic acid and archidonic acid were found inthe reproductive tracts. 3. Palmitic acid among saturated fatty acids and oleic acid among unsaturated fatty acids were the hihgest level in all of the reproductive tracts. 4. Palmitic acid, oleic acid and stearic acid showed higher rate with 44.89%, 23.69% and 14.36%, respectively, and lauric acid, capric acid, palmitoleic acid, arachidonic acid ad myristic acid showed lower rate with 0.62%, 1.13%, 1.65%, 1.97% and 2.24%, respectively in the reproductive fluid. 5. The highest level of arachidonic acid was found in the uterine horn. 6. The sum of the palmitic acid and oleic acid were 66.91%, 70.41%, 66.14% and 73.36% in the fluid of follicle, oviduct, uterine horn and uterine body, respectively. 7. The relative composition of arachidonic acid was higher during the follicular stage than during the luteal phase in the fluid of oviduct and uterine. 8. The long chain fatty acids such as the palmitic acid, stearic acid, oleic acid and linoleic acid showed higher relative compositions during the follicular phase(93.18%∼96.83%) than during the luteal phase(82.56%∼88.37%). 9. Caprylic acid, luric acid and palmitoleic acid were undetected in the fluid of all of the reproductive tracts during the follicular phase. Low relative compositions of capric acid, myristic acid andarachidonic acid were found during the follicular phase, while the low relative compositions (<5%)of capric acid, lauric acid, myristic acid, plamitoleic acid and arachidonic acid were found during the luteal phase.
