Mutagenic Activity of Smoke Flavoring Processed from Oak and Apple Wood on Manufacturing Temperature (굴참나무와 사과나무로부터 제조한 훈연액의 제조온도에 따른 돌연변이원성에 관한 연구)

  • The study was carried out to screen mutagenicity of smoking materials for the determination of optimum smoking temperature for meat products. Wood materials employed for smoking were oak and apple trees. Temperatures of the generator for manufacturing of smoke flavoring were set to 250$^{\circ}C$, 400$^{\circ}C$ and 500$^{\circ}C$, respectively. Mutagenic activities of smoke flavoring were assayed according to Ames test using Salmonella typhimurium TA98 and TA 100. In oak wood smoke flavoring, Salmonella typhimurium TA98 without S-9 mix showed strong mutagenic activities at the concentration of 6$\mu\textrm{g}$/plate(250$^{\circ}C$), 4$\mu\textrm{g}$/plate(400$^{circ}C$) and 6$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA100 with S-9 mix showed strong mutagenic activities at the concentration of 10$\mu\textrm{g}$/plate(250$^{\circ}C$), 20$\mu\textrm{g}$/plate(400$^{\circ}C$) and 10$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA98 with S-9 mix showed strong mutagenic activities at the concentration of 30$\mu\textrm{g}$/plate(250$^{\circ}C$), 40$\mu\textrm{g}$/plate(400$^{\circ}C$) and 20$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA100 with S-9 mix showed strong mutagenic activities at the concentration of 30$\mu\textrm{g}$/plate(250$^{\circ}C$), 50$\mu\textrm{g}$/plate(400$^{\circ}C$) and 20$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA100 without S-9 mix showed strong mutagenic activities at the concentration of 10$\mu\textrm{g}$/plate(250$^{\circ}C$), 20$\mu\textrm{g}$/plate(400$^{\circ}C$) and 20$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA98 with S-9 mix showed strong mutagenic activities at the concentration of 30$\mu\textrm{g}$/plate(250$^{\circ}C$), 40$\mu\textrm{g}$/plate(400$^{\circ}C$) and30$\mu\textrm{g}$/plate(500$^{\circ}C$). Salmonella typhimurium TA100 with S-9 mix showed strong mutagenic activities at the concentration of 30$\mu\textrm{g}$/plate(250$^{\circ}C$), 20$\mu\textrm{g}$/plate(400$^{\circ}C$) and 30$\mu\textrm{g}$/plate(500$^{\circ}C$). From these results, it could be concluded that optimum smoking temperature for meat products should be set below 400$^{\circ}C$, that the compounds like benzo[a]pyrene etc. contain a variety of mutagenic potentials, which could be generated at the higher smoking temperature.

  • Hydroxyapatite films were prepared on Si(100) substrates by using a sol-gel method with calcium nitrate and phosphoric acid as starting materials. Precursor sols were spin-coated onto the substrates and prefired at $500^{\circ}C$ for 10 min in air. Formation of the hydroxypatite structure was confirmed in the sample annealed at $500^{\circ}C$ by the X-ray diffraction $\theta$-2$\theta$ scans and a tricalcium phosphate phase was observed in the samples annealed at both temperature regions of $500^{\circ}C$~$700^{\circ}C$ and $900^{\circ}C$. From the results of Fourier transform infrared spectroscopy, the change of a carbon content and improvement of crystallinity have been discussed as a function of increase of annealing temperature.

  • Tin oxide was prepared by microwave heating for anode material of lithium ion battery. The samples were heated at 300, 500 and $700^{\circ}C$ for 3h under flowing oxygen after microwave heating. The effect of microwave heating on the electrochemical performance of the manufactured tin oxide and the reversible capacity performance were investigated. Tin oxide heated at $500^{\circ}C$ showed higher capacity than those at $300^{\circ}C$ and $700^{\circ}C$ under microwave heating condition. Comparing microwave and furnace heating, microwave heating condition showed higher capacity. The discharge capacity after microwave heating and $500^{\circ}C$ heating showed 1,500 mAh/g.

  • Closed field unbalanced magnetron sputtering 방법을 이용하여 CrZr-Si-N 박막을 증착하였다. Si Target power의 변화에 따라 박막을 증착하여 XRD, SEM, XPS, GDOES, AFM, XPS, Nanoindentation을 이용하여 박막의 미세구조, 성분분석, 표면 조도, 경도를 측정하였다. $500^{\circ}C$에서 annealing한 후 상온에서의 박막의 경도와 비교하였고, 상온과 $500^{\circ}C$에서 마모 실험을 행한 후 마찰 계수를 측정하여 비교하였다. $Cr_{39.4}Zr_{12.3}N_{48.3}$ 박막은 $500^{\circ}C$ annealing 후 경도는 30 GPa에서 24 GPa로 감소하였고 마찰계수는 0.23에서 0.81로 약 4배 증가하였다. $500^{\circ}C$ annealing 후 $Cr_{34.6}Zr_{10.6}-Si_{6.4}-N_{48.4}$ 박막의 경도는 30 GPa로 상온에서의 경도 32 GPa과 비슷하였고 $500^{\circ}C$와 상온에서 수행된 마모시험 결과는 $500^{\circ}C$에서 마찰계수 0.43으로 상온 마모시험 결과와 거의 비슷한 결과를 보였다. 상온의 경우 Si 함량에 따른 기계적 특성 및 마모특성의 변화는 거의 없었다. 그러나 $500^{\circ}C$ annealing 후 CrZi-Si-N 박막의 기계적 특성 및 마모특성은 Si 함량에 따라 큰 차이를 나타내었다. 이러한 결과들을 통해 Si 첨가가 CrZrN 박막의 고온 안정성 향상에 기여함을 확인할 수 있었다.

THE EFFECTS OF HEAT TREATMENT OF ORTHODONTIC WIRES (교정용 강선재의 열처리 효과에 관한 실험적 연구)

  • The purpose of this study was to evaluate the effect of heat treatment on physical properties of 0.016' and 0.016' x 0.022' stainless steel wires. Temperature of heat treatment had intervals of $50^{\circ}C$ from $400^{\circ}C$ to $700^{\circ}C$, and time of heat treatment were 3, 6 and 9 minutes. Tensile tests were measured by ultimate tensile strength and yield strength. Bending tests were assessed by maximum bending force, recovery force, and stiffness. Torsion test was evaluated by torsion cycle until wires were fractured. The results were as follows: 1. In round wires, the highest value of ultimate tensile strength and yield strength were recorded of heat treatment at $500^{\circ}C$. In rectangular wires, the highest value of ultimate tensile strength were after 9 minutes at $400^{\circ}C,\;450^{\circ}C$ and 3, 6 minutes of heat treatment at $50^{\circ}C$, yield strength were the highest value after 3, 6 minutes of heat treatment at $500^{\circ}C$. 2. In both round and rectangular wires, maximum bending force and recovery force were the highest values after 6 minutes of heat treatment at $500^{\circ}C$. In round wires, highest value of stiffness were formed after 9 minutes at heat treatment at $500^{\circ}C$. In rectangular wires, the highest value of stiffness were for 6 minutes in $500^{\circ}C$. 3. In rectangular wires, torsion cycle was minimum after 6 minutes of heat treatment at $500^{\circ}C$. 4. In all of tension, bending, and torsion tests, the heat treated wires were softened over at $700^{\circ}C$. 5. In all of tension, bending, and torsion tests, physical properties of the wires were more influenced by the temperatures than the duration of the heat treatment.

  • Phase transition kinetics of nonstoichiometric amorphous titanium oxide thin films deposited by reactive sputtering was investigated after cooling down with various rate followed by l0min.-3hrs. annealing at $500^{\circ}C$~$600^{\circ}C$ After short duration and fast cooling. Magneli was the only crystalline phase because the oxidation rates of $TiO_{2-x}$, could be relatively slower than that of crystallization. When the films were cooled slowly between $500^{\circ}C$~$300{\circ}C$, Magneli was transformed into an anatase and stabilized, but directly into a rutile under fast cooling. Because the rutile also prevailed after cooling from $600^{\circ}C$, it was concluded that the rutile phase could be formed directly from Magneli as well as converted from the anatase. Changes in volume and surface morphology were observed related to crystallization and oxidation processduring heat treatment.

Changes in Properties and Surface FT-IR Spectra of Wood Charcoal at Different Carbonization Temperatures (탄화온도에 따른 목탄 특성 및 표면의 FT-IR 변화)

  • To analyze the changes in pH and elemental content ratio of wood charcoal and in FT-IR spectra of their surfaces, wood charcoals carbonized from Pinus koraiensis were used. pHs of wood charcoals carbonized from Pinus koraiensis at 300 and $400^{\circ}C$ were 5-27 and 6.80, respectively, whereas they were between 9.25~10.35 for the wood charcoals manufactured between $500{\sim}900^{\circ}C$ From the changes in the elemental ratios of Pinus koraiensis wood charcoal by increasing carbonization temperature, carbon (C) contents increased by elevating the carbonization temperature with the decreasing in content ratios of O and H. The largest changes in the ratio was found between the carbonization temperature 400 and $500^{\circ}C$. Ratios of C, O, H of the wood charcoal manufactured at 300 and $400^{\circ}C$ were 67.7, 28.9, 3.0% and 72.2, 24.9, 2.5%, respectively, while those at $500{\sim}900^{\circ}C$ were between 83.3~90.5, 13.6~9.0, 2.7~0.3%. The surface functional groups of Pinus koraiensis wood charcoals were determined by comparison of FT-IR spectra of the wood powder and the wood charcoal carbonized with the wood powder. The functional groups on the surface of wood charcoals carbonized at 300 and $400^{\circ}C$ were considered to be acidic groups like Lactone, Lactol, Carboxylic acid, Carboxylic anhydride, whereas Pyrone types could be the major functional groups for the wood charcoals carbonized between 500 and $900^{\circ}C$.

  • Austenitic stainless steel type 304L has been nitrided under the low pressure of high nitrogen environment for 5 hours by the square-wave-pulse-d.c. plasma as a function of temperature 400~$600^{\circ}C$ and of pulsation. At the temperature range lower than $500^{\circ}C$ and at the relatively high ratio of pulse duration to pulse period, nonstoichiometric stainless steel nitride has been developed in the form of a thin layer which has many cracks. At the temperature range higher than $500^{\circ}C$, with the increasing temperature or with the increasing ratio of the pulse duration to pulse period up to 50s/100s, the nitrided layer was composed mainly of CrN and Fe4N phases and became thick, uniform, columnar and nearly crack-free. The nitrided layer at $500^{\circ}C$ was mixed with the low-temperature layer and the high temperature layer and was very brittle.

Optical, Structural and Photo-catalytic properties of TiO2 thin films prepared by using Ti-naphthenate (Ti-naphthenate를 이용하여 제조한 광감응성 TiO2 박막의 광학적 및 구조적 특성)

  • Photo-reactive $TiO_2$ thin films on soda-lime-silica slide glass were prepared by spin coating technique with a Ti-naphthenate precursor. Optical, structural and photo-catalytic properties of the films after annealing at $500^{\circ}C{\sim}600^{\circ}C$ were evaluated. As increase with annealing temperature, absorption bands and total transmittance of the films showing an average transmittance (about 80%) at visible spectra range were shifted to UV spectra range and slightly decreased. Refractive index and thickness of the films were increased from 2.16 to 2.63 and decreased from 484 nm to 439 nm, respectively, with increase of annealing temperature. Anatase phase was visible at all annealing temperature. More rougher surface structure was obtained at $600^{\circ}C$ than those of films annealed at $500^{\circ}C$ and $550^{\circ}C$. The hydrophilic conversion was found within 45 min by UV stimulation and optical activation was UVC>UVA>UVB at the case of $500^{\circ}C{\sim}550^{\circ}C$ and UVA>UVC>UVB at the annealing temperature of $600^{\circ}C$. The lowest initial contact angle was obtained at $600^{\circ}C$.

Effects of Temperature and Water Pressure on the Material Properties of Granite & Limestone from Gagok Mine (온도와 수압이 가곡광산 화강암과 석회암의 물성에 미치는 영향)

  • This study focuses on having a temperature and water pressure effects on the change of material properties of rocks. Granite and limestone specimens from Gagok Mine were thermally treated with predetermined temperatures of 200, 300, 400, 500, 600 and $700^{\circ}C$ (excepting $700^{\circ}C$ for limestone) to estimate the reduction of material properties of rocks caused by heat. Specific gravity, effective porosity, elastic wave velocity, uniaxial compressive strength, Young's modulus and Poisson's ratio for pre-heated specimens were measured. With increasing temperature, material properties of both rock specimens change sequentially. Significant changes of specific gravity, effective porosity and elastic wave porosity occur above $400^{\circ}C$ for granite and $300^{\circ}C$ for limestone. Changes of uniaxial compressive strength, Young's modulus and Poisson's ratio seem to be similar to those of physical properties. GSI of 500, 600 and $700^{\circ}C$ specimens inferred by using uniaxial compressive strength and Young's modulus of preheated granite specimens is found to be 81, 66 and 58 each. In case of pre-heated limestone specimens of 400, 500 and $600^{\circ}C$, the corresponding GSI is 76, 71 and 65 each. 500, 600 and $700^{\circ}C$ granite specimens and 400, 500 and $600^{\circ}C$ limestone specimens were pressurized to 7.5 MPa and their effective porosity, elastic wave velocity, uniaxial compressive strength and Young's modulus were measured. The average value of material properties (mentioned above) of 500, 600 and $700^{\circ}C$ granite specimens under water pressure compared with material properties of non-pressurized pre-heated specimens exhibits the reduction of 7.6, 11.3 and 14.9%, respectively. In case of 400, 500 and $600^{\circ}C$ limestone specimens under water pressure, the average value of material properties decreases by 8.2, 13.8 and 21.9%, respectively.
