Tolfenamic Acid Suppresses Inflammatory Stimuli-Mediated Activation of NF-κB Signaling

  • Tolfenamic acid (TA) is a traditional non-steroid anti-inflammatory drug (NSAID) and has been broadly used for the treatment of migraines. Nuclear factor kappa B (NF-${\kappa}B$) is a sequence-specific transcription factor and plays a key role in the development and progression of inflammation and cancer. We performed the current study to investigate the underlying mechanisms by which TA suppresses inflammation focusing on NF-${\kappa}B$ pathway in TNF-${\alpha}$ stimulated human normal and cancer cell lines and lipopolysaccharide (LPS)-stimulated mouse macrophages. Different types of human cells (HCT116, HT-29 and HEK293) and mouse macrophages (RAW264.7) were pre-treated with different concentrations of TA and then exposed to inflammatory stimuli such as TNF-${\alpha}$ and LPS. Transcriptional activity of NF-${\kappa}B$, $l{\kappa}B-{\alpha}$-degradation, p65 translocation and mitogen-activated protein kinase (MAPK) activations were measured using luciferase assay and Western blots. Pre-treatment of TA repressed TNF-${\alpha}$- or LPS-stimulated NF-${\kappa}B$ transactivation in a dose-dependent manner. TA treatment reduced degradation of $l{\kappa}B-{\alpha}$ and subsequent translocation of p65 into nucleus. TA significantly down-regulated the phosphorylation of c-Jun N-terminal kinase (JNK). However, TA had no effect on NF-${\kappa}B$ signaling and JNK phosphorylation in HT-29 human colorectal cancer cells. TA possesses anti-inflammatory activities through suppression of JNK/NF-${\kappa}B$ pathway in different types of cells.

Kojic Acid, a Potential Inhibitor of NF-$textsc{k}$B Activation in Transfectant Human HaCaT and SCC-13 Cells

  • The activation of NF-$\kappa$B induced by kojic Acid, an inhibitor of tyrosinase for biosynthesis of melanin in melanocytes, was investigated in human transfectant HaCaT and SCC-13 cells. These two keratinocyte cell lines transfected with pNF-$\kappa$B-SEAP-NPT plasmid were used to determine the activation of NF-$\kappa$B. Transfectant cells release the secretory alkaline phosphatase (SEAP) as a transcription reporter in response to the NF-$\kappa$B activity and contain the neomycin phosphotransferase (NPT) gene for the dominant selective marker of geneticin resistance. NF-$\kappa$B activation was measured in the SEAP reporter gene assay using a fluorescence detection method. Kojic Acid showed the inhibition of cellular NF-$\kappa$B activity in both human keratinocyte transfectants. It could also downregulate the ultraviolet ray (UVR)-induced activation of NF-$\kappa$B expression in transfectant HaCaT cells. Moreover, the inhibitory activity of kojic Acid in transfectant HaCaT cells was found to be more potent than known antioxidants, e.g., vitamin C and N~acetyl-L-cysteine. These results indicate that kojic Acid is a potential inhibitor of NF-$\kappa$B activation in human keratinocytes, and suggest the hypothesis that NF-$\kappa$B activation may be involved in kojic Acid induced anti-melanogenic effect.

인체 전립선 상피세포에서 HDAC 저해제 trichostatin A의 caspase 및 NF-κB의 활성화를 통한 apoptosis 유도 (Induction of Apoptosis by HDAC Inhibitor Trichostatin A through Activation of Caspases and NF-κB in Human Prostate Epithelial Cells.)

  • 본 연구에서는 인체전립선 상피세포인 267B1 세포에서 HDAC 저해제인 TSA에 의한 증식억제가 apoptosis 유도에 의한 것임을 제시하였다. 이러한 TSA에 의한 267B1 세포의 apoptosis에는 c-IAP-1 및 c-IAP-2와 같은 IAP family의 발현감소가 동반되었으나 Bax 및 Bcl-2와 같은 Bcl-2 family의 발현에는 큰 변화가 없었다. 그리고 TSA에 의한 267Bl 세포의 apoptosis는 caspase의 활성에 의한 표적 단백질들의 분해와 연관성이 있었다. 또한 TSA에 의한 apoptosis 유도에서 $NF-{\kappa}B$의 활성이 증가된다는 것을 세포질에서 $NF-{\kappa}B$의 핵 내로의 이동에 따른 전사활성의 증가 현상에 의한 것임을 다양한 방법으로 제시하였다. 본 연구의 결과는 TSA와 같은 HDAC 저해제에 의한 apoptosis 유도에는 $NF-{\kappa}B$의 활성 증가가 동반될 수 있음을 보여주는 결과로서 HDAC 저해제의 항암활성에 대한 $NF-{\kappa}B$의 새로운 역할 가능성을 제시하여 주는 것으로서 이에 관한 추가적인 연구의 필요성을 제시하였다.

봉독의 세포자멸사를 통한 LNCaP 세포증식 억제 (Bee Venom Inhibits LNCaP Cell Proliferation Through Induction of Apoptosis Via Inactivation of $NF-{\kappa}B$)

  • 목적 : 이 연구는 봉약침의 봉독과 그 주요성분인 멜리틴이 $NF-{\kappa}B$의 활성억제와 세포자멸사 관련 단백질의 발현 조절을 통하여 세포자멸사를 유도하고 전립선 암세포주인 LNCaP 세포의 성장을 억제하는지를 확인하고 해당 기전을 살펴보고자 하였다. 방법 : 봉독이나 멜리틴을 처리한 후 LNCaP의 성장억제를 관찰하기 위해 WST-1 assay, CCK-8 assay를 시행하였고, 세포자멸사의 관찰에는 DAPI, TUNEL staining assay를 시행하였으며, 세포자멸사 조절단백질의 변동 관찰에는 western blot analysis를 시행하였고, 세포자멸사와 연관된 $NF-{\kappa}B$의 활성 변화를 관찰하기 위해 EMSA를 시행하였으며, LNCaP에서 봉독이나 멜리틴과 $NF-{\kappa}B$의 상호작용을 관찰하기 위해 transient transfection assay를 시행 시 세포생존율과 $NF-{\kappa}B$의 활성 변동을 측정하였다. 결과 : LNCaP 세포에 봉독이나 멜리틴을 처리한 후, 전립선암세포의 성장, 세포자멸사의 유발, 세포자멸사 관련 단백질의 발현, $NF-{\kappa}B$의 활성, $NF-{\kappa}B$의 p50 치환 후 $NF-{\kappa}B$의 활성과 LNCaP 세포 증식에 미치는 영향을 관찰하여 다음과 같은 결과를 얻었다. 1. LNCaP 세포에서 봉독이나 멜리틴을 처리한 후 세포자멸사가 유도되어 세포성장이 억제되었고, 세포자멸사 관련 단백질 중 분리된 PARP, caspase-9, Bax는 유의한 증가를, Bcl-2, p-Akt, MMP 13, XIAP, cXIAP는 유의한 감소를 나타내었다. 2. LNCaP 세포에서 봉독이나 멜리틴을 처리한 후 $NF-{\kappa}B$의 활성의 유의한 감소를 나타내었다. 3. LNCaP 세포에서 $NF-{\kappa}B$ p50를 치환하여 작용기를 없애고 봉독이나 멜리틴을 처리하였을 경우에도 $NF-{\kappa}B$의 활성의 유의한 감소를 나타내었다. 결론 : 이상의 결과는 봉독이나 멜리틴이 $NF-{\kappa}B$의 활성 억제를 통하여 인간 전립선암세포주인 LNCaP의 세포자멸사를 유발함으로써 증식억제 효과가 있음을 입증한 것으로 전립선암의 예방과 치료에 대한 효과적인 치료제 개발에 도움이 될 것으로 기대된다. 다만 그 기전에서 봉독이나 멜리틴은 기존연구와 달리 $NF-{\kappa}B$ p50의 작용기와 직접적으로 상호작용을 하지는 않는 것으로 보이므로 심화 연구를 요한다.

  • Cobrotoxin의 항염증 치료 기전에 대해 연구하기 위하여 성상세포에 LPS 및 SNP로 염증을 유도한 후 NF-${\kappa}B$와 DNA의 결합 능력, NF-kB Dependent Luciferase 발현, Astrocyte의 세포활성, NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}BB$ 및 염증(炎症)관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2의 발현과 GSH와 DTT로 sulf-hydryl기를 환원시 NF-${\kappa}B$와 DNA의 결합 능력, NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향과 Cobrotoxin의 성상세포 내 유입 등을 관찰하여 다음과 같은 결론을 얻었다. 1. LPS 로 염증을 유발한 후 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 Cobrotoxin 0.1${\mu}g/m{\ell}$ 처리군, Astrocyte 내에서의 Cobrotoxin 0.1, 0.5${\mu}g/m{\ell}$ 처리군에서 모두 대조군에 비하여 유의한 억제를 나타내었다. 2. LPS로 염증을 유발한 후 Astrocyte 내에서 NF-${\kappa}B$ Dependent Luciferase 발현을 살펴본 결과 Cobrotoxin 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 3. SNP로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었고, P-$1{\kappa}B$는 Cobrotoxin $0.1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 억제를, Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 4. LPS로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 5. SNP로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2, iNOS, cPLA2 모두 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 6. LPS로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 견전자(遣傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2와 cPLA2의 경우 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서, iNOS의 경우 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 7. Astrocyte내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 BNA의 결합 능력을 관찰한 결과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군은 각각 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 8. Astrocyte 내에서 LPS로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 5mM의 동시처리군 모두에서 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 9. Astrocyte 내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기 환원시 NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향을 관찰한 결과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 DTT 1 또는 5mM 동시처리군과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 GSH 1 또는 5mM 동시처리군 모두에서 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에 비하여 발현의 유의한 증가를 나타내었다. 10. Cobrotoxin의 세포 내 유입 확인을 살펴본 결과 Astrocyte 내에서 Cobrotoxin이 세포내로 유입되는 것으로 나타났다. 이상의 결과로 보아 Cobrotoxin 처리는 성상 세포 들을 대상을 LPS 및 SNP로 유도된 NF-${\kappa}B$ 관련 염증 기전과 iNOS, COX-2, cPLA2와 같은 염증관련 유전자의 발현 및 NO, PGE2,에 유의한 변동을 나타내었고, 이들 결과는 Cobrotoxin의 항염증 효과 및 그 치료기전에 대하여 입증한 것이며, 향후 안전성 연구를 바탕으로 신경계 및 심혈관계 염증치료에 약침개발과 같은 적극적인 활용이 기대된다.

Effect of Minocycline on Activation of Glia and Nuclear Factor kappa B in an Animal Nerve Injury Model

  • Glial cells are activated in neuropathy and play a key role in hyperalgesia and allodynia. This study was performed to determine whether minocycline could attenuate heat hyperalgesia and mechanical allodynia, and how glial cell activation and nuclear factor kappa B (NF-kappaB) were regulated by minocycline in a model of chronic constriction of sciatic nerve (CCl). When minocycline (50 mg/kg, oral) was daily administered from 1 day before to 9 days after ligation, heat hyperalgesia and mechanical allodynia were attenuated. Furthermore, when minocycline treatment was initiated 1 or 3 days after ligation, attenuation of the hypersensitive behavior was still robust. However, the effect of attenuation was less when minocycline was started from day 5. In order to elucidate the mechanism of pain attenuation by minocycline, we examined the changes of glia and NF-kappaB, and found that attenuated hyperalgesia and allodynia by minocycline was accompanied by reduced microglial activation. Furthermore, the number of NF-kappaB immunoreactive cells increased after CCI treatment and this increase was attenuated by minocycline. We also observed translocation of NF-kappaB into the nuclei of activated glial cells. These results suggest that minocycline inhibits activation of glial cells and NF-kappaB, thereby attenuating the development of behavioral hypersensitivity to stimuli.

초오의 항알레르기 염증반응 및 기전탐색에 관한 연구 (Experimental Study about Pathway of Aconiti Ciliare Tuber on Allergic Reaction of Inflammation)

  • Objetives : The purpose of this study was to examine the pathway of anti-allergic effects of Aconiti Ciliare Tuber (ACT). Methods : We examined cell viability, ${\beta}$-hexosaminidase release, pro-inflammatory cytokines secretion and mRNA expressions, nuclear factor-kappa B (NF-${\kappa}B$) (p65) activation, inhibitor kappa B-alpha ($I{\kappa}B-{\alpha}$) degradation, and MAPKs activation from RBL-2H3 cells pre-treatment by ACT of 1.0 mg/ml, 2.0 mg/ml separately. Results : We observed that ACT reduced the secretion of ${\beta}$-hexosaminidase, TNF-${\alpha}$, IL-4 and the expression of COX-2 mRNA in RBL-2H3 cells. Futhermore, ACT inhibited the levels of activation of NF-${\kappa}B$ (p65) protein, ERK MAPK, and degradation of $I{\kappa}B-{\alpha}$ in RBL-2H3 cells. Conclusions : These results show that ACT has an anti-histamine effect and inhibitory effect of NF-${\kappa}B$ (p65) through regulation of $I{\kappa}B-{\alpha}$ degradation. This improves that ACT could be used as an anti-allergic medicine.

천맥탕(天麥湯)의 알레르기 염증반응 및 기전탐색에 관한 연구 (Experimental Study about the Pathway of Inflammatory Allergic Reaction of Cheonmaec-tang)

  • Objectives The purpose of this study was to examine the pathway of anti-allergic effects of Cheonmaec-tang (CMT).   • Objectives : The purpose of this study was to investigate the toll-like receptor (TLR)-4 mediated anti-inflammatory effects of extract from Shigyungbanha-tang (SBT) on the peritoneal macrophage. Methods : To evaluate of TLR-4 mediated inflammatory of SBT. we examined NO and cytokine production in TRL-4 ligand (LPS : lipopolysaccharide) induced macrophages. Furthermore, we examined its molecular mechanism using western blot. Results : Extract from SBT itself does not have any cytotoxic effect in the peritoneal macrophages. Extract from SBT reduced LPS-induced nitric oxide (NO). tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin (IL)-6 and IL-12 production in peritoneal macrophages. SBT inhibited degradation of inhibitor kappa B-alpha ($I{\kappa}B-{\alpha}$) in the TLR-4 mediated peritoneal macrophages. Conclusions : These results suggest that SBT inhibits NO and cytokines production through inhibiting nuclear factor-kappaB (NF-${\kappa}$B) activation in peritoneal macrophage and that SBT may be beneficial oriental medicine for inflammation.

Guggulsterone Suppresses the Activation of NF-${\kappa}B$ and Expression of COX-2 Induced by Toll-like Receptor 2, 3, and 4 Agonists

  • Toll-like receptors (TLRs) induce innate immune responses recognizing conserved microbial structural molecules. All TLR signaling pathways culminate in the activation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$). The activation of NF-${\kappa}B$ leads to the induction of inflammatory gene products such as cyclooxygenase-2 (COX-2). Guggul has been used for centuries to treat a variety of diseases. Guggulstreone, one of the active ingredients in guggul, has been used to treat many chronic diseases. However, the mechanism as to how guggulsterone mediate the health effects is largely unknown. Here, we report biochemical evidence that guggulsterone inhibits the NF-${\kappa}B$ activation and COX-2 expression induced by TLR2, TLR3, and TLR4 agonists. Guggulsterone also inhibits the NF-${\kappa}B$ activation induced by downstream signaling components of TLRs, myeloid differential factor 88 (MyD88), $I{\kappa}B$ kinase ${\beta}$ ($IKK{\beta}$), and p65. These results imply that guggulsterone can modulate the immune responses regulated by TLR signaling pathways.
