Free amino acid in ethanol extracts and total amino acids in hydrolysates of eleven species of edible mushrooms were analyzed and determinated the contents five kind of new amino acid by means of amino acid autoanalyzer and gas liquid chromatography. The result obtained from this study are as follows. 1) Five kind of new amino acid turned out to be ${\alpha}$-aminobutyric acid, allo-isoleucine, ethanolamine, $\gamma$-aminobutyric acid and ornithine. 2) By means of amino acid autoanalyzer, the monoethanolamine was identified on the chromatogram ahead of alanine, ${\alpha}$-aminobutyric acid between peak of threonine and glycine, allo-isoeleucine between peak of valine and leucine, isoleucine, ${\gamma}$-aminobutyric acid followed by proline between peak of leucine, isoleucine and methionine and ornithine between peak of phenylalanine and tyrosine 3) By means of Gas liquid chromatography, the ${\alpha}$-aminobutyric acid was identified on the chromatogram between peaks of alanine and valine, allo-isoleucine between peaks of methionine and isoleucine, monoethanolamine followed by ${\gamma}$-aminobutyric acid between peaks of phenylalanine and ammonia, ornithine between the peaks of ammonia and lysine. 4) Of five amino acids which were identified, ornithine was the highest of its content in the mushroom extracts, and allo-isoleucine, ethanolamine, and ${\gamma}$-aminobutyric acid came next in decreasing order. 5) Also which were identified, ornithine was the highest of its content in the hydrolysates, and ${\alpha}$-aminobutyric acid, ${\gamma}$-aminobutyric acid, allo-isoleucine came next in decreasing order, ethanol extracts and hydrolysates of Auriculariaauricula-Judae(Fr.) $Qu\acute{e}l$ species didn't contain any of five kind of new amino acid. Ornithine also was the highest in the hydrolysates of ll mushrooms.
The effect of germination on hydration and germination properties, and on the changes of fatty acids and amino acids profiles of a brown rice 'Keunnun' (KN) with a large embryo was compared to 'Ilpumbyeo' (IP) with a normal embryo. A rapid germination up to 24 hr was observed in both brown rice cultivars, afterward decreased with germination time. At 60 hr, the KN ($86.0{\pm}4.24%$) showed slightly lower germination capability than the IP ($97.0{\pm}1.41%$). Lower water uptake during germination was also found in the KN ($1.22{\pm}0.02\;g$) compared to the IP ($1.59{\pm}0.05\;g$). Major fatty acids were palmitic acid, oleic acid, and linoleic acid accounting for more than 95% of total fatty acids. The most abundant amino acid in both types was oleic acid, which was decreased during germination, whereas palmitic and linoleic acids were increased. Eight amino acids were detected, and a remarkable increase in ${\gamma}-amino$ butyric acid (GABA) during germination was observed. The KN was characterized with higher tasty amino acids of aspartic acid, glutamic acid, glycine, and alanine.
To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.
The environmental effects on radio-sensitivities of Lactobacillus and Saccharomyces cerevisiae were studied; Liquid suspensions of Lactobacillus and yeast were gamma-irradiated under various conditions; temperatures, hydrogen ion concentrations, amino acids and vitamins were treated seperately with variations of concentrations. (shown in figures) It is found that simultaneous heat treatment is effective to sterilize microorganisms than pre after treatment, and concentration of hydrogen ion does not affect the lethalty of yeast but or Lactobacilli was affect at the range of pH. 5.0 to 7.0. Ascorbic acid, thiamin and pyridoxine were protective dependently against lethal action of gamma-ray and its protective effects increase with the increasings of concentrations. Glutamic acid, aspartic acid, tyrosine and phenylalanine were proved to be protective for both strains at 0.1 between 1.0 percent. It can be suggested that industrial sterilizing doses of irradiation by gamma-ray for food should be applied more than those dose of saline or buffer suspension, because natural food stuffs are rich of vitamins and amino acids.
To elucidate the composition of extractive nitrogenous components in the fresh Capsosiphons fulvescens cultured off the southern coast of Korea, and to determine the monthly variation of these nitrogenous components, extract samples collected monthly from December to March at Jangheung-gun, Jeonnam Province were analyzed for total nitrogen, free and combined amino acids, ATP and related compounds, betaines, trimethylamine oxide (TMAO) and trimethylamine (TMA). The content of extractive nitrogen was 1,090~1,233 mg/100 g on dry basis. The number of 21~25 ninhydrin-positive substances was detected in the analysis of free amino acids, and their total amount was 3,710~4,788 mg/100 g on dry basis. Among them, free proline, asparagine, glutamic acid, alanine, taurine and glutamine were found to be abundant. The combined amino acids amounted to 1,573~2,121 mg/100 g in total and the total amount of ATP and related compound was 33.8~84.0 mg/100 g ($1.06{\sim}2.46\;{\mu}mol/g$) on dry basis. Betaine, glycinebetaine, $\beta$-alaninebetaine, $\gamma$-butyrobetaine, homarine and trigonelline were detected in most of samples. Levels of free and combined amino acids, ATP and related compounds fluctuated from sample to sample, with their contents higher in December and January and lower in March.
The Korean rice of Paldal variety was milled, in March, 1969, into brown rice and 92% polished one and each 1.5kg sample was packed in the kraft paper bags. With Co-60 gamma-irradiation, the samples were treated in the various ranges of 500, 800 and 1,000 Krad, and then they were stored at the room temperature. The changes of the contents of free amino acids in the samples were determined by the amino acid autoanalyzer (Yanagimoto, Japan, LC-type). The results obtained in June and October of the year were as follows. 1) In the disinfecting irradiation of rice by the higher doses of 500, 800 and 1,000 Krad, there wasn't any particular distinction between the control lot and irradiated lot in respect to the free amino acid contents. 2) The contents of free amino acids in rice during storage had the decreasing tendencies, and especially prominent in case of glutamic acid.
In order to investigate the composition of dried layer, Porphyra dentata cultured at the south coast of Korea, the dried layer was analyzed for extractive nitrogen, free amino acids, combined amino acids, ATP and its related compounds and quaternary ammonium basis, The extractive nitrogen contents of dried laver extracts were 670-1,304 mg/100 g (on dry basis). From twenty-eight to twenty-nine kinds of free amino acids were found in the dried laver extracts and their total amounts were $2,796\~6,277\;mg/100\;g$ (on dry basis). The extracts were rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and phosphoserine, From eighteen to twenty-one kinds of combined amino acids were found in the extracts and their total amounts were $1,406\~2,142\;mg/100\;g$ (on dry basis). The amounts of ATP and its related compounds were $65.7\~124,7\;mg/100\;g(2.13\sim3.68{\mu}mol/g$ on dry basis), Homarine was detected in all samples but $\beta$-alaninebetaine, $\gamma$-bufobetaine and trigonelline disappeared during processing. TMAO and TMA were detected in all samples. During processing of dried layer, free amino acids, TMAO and TMA were increased but the other constituents such as combined amino acids, ATP and its related compounds and betaines were decreased in all specimens.

In order to investigate the composition and monthly variation of extractive nitrogenous components in the laver Porphyra dentata cultured at the south coast of Korea, the fresh laver was analyzed separately for the amounts of free amino acids, combined amino acids, ATP and its related compounds, and quaternary ammonium basis in fresh laver were measured. The extractive nitrogen contents of fresh laver extracts were 760~870 mg/100g (dry basis). Twenty-seven to thirty-one kinds of free amino acids were detected in the laver extracts and their total amounts were 2,404~3,966 mg/100g (on dry basis). The laver extracts showed rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and aspartic acid. Sixteen to twenty-three kinds of combined amino acids were detected in the extracts and their total amounts were 1,429~2,692 mg/100g (on dry basis). Proline, glutamic acid, glycine, phosphoserine, serine were the amin combined amino acids in the extracts. The amounts of ATP and related compounds were 73.3~94.4 mg/100 g (2.04~4.43 $\mu$mol/g, on dry basis). Homarine and trigonelline were detected in all specimens but $\beta$-alaninebetaine, ${\gamma}$-butyrobetaine were found in some. Small amounts of trimethylamine were detected in all samples. Free and combined amino acids were occupying almost 90% of extractive nitrogen. Most of free and combined amino acids showed a marked monthly variation with a maximum in January and March, and a minimum in February and April. The fresh laver P. dentata did not differ much from the fresh laver P. yezoensis in qualitative com-position of extractive components, but their contents were generally low level.
$\gamma$-Glutamyl transpeptidase ($\gamma$-GTP; EC 2.3.2.2) present in the culture filtrate of Bacillus sp. KUN-17 was purified 400-fold through a consecutive procedure including organic precipitation and column chromatography. The enzyme has an estimated molecular weight of 70, 000 and consists of hetero-subunits with molecular weights of 42, 000 and 22, 000. In vitro optimal conditions for those transfer and hydrolysis reactions appeared to be pH 7.0 at $50^{\circ}C$ and pH 8.4 at $40^{\circ}C$, respectively. The denatured enzyme recovered most of its $\gamma$-GTP activity by removing detergents such as sodium dodecyl sulfate (SDS) or urea with dialysis. The enzyme showed higher affinities against a number of amino acids as $\gamma$-glutamyl acceptors than glycylglycine in the following order: L-valine, L-methionine, L-glutamic acid or L-as-paragine, L-alanine. Also, it was shown that L-glutamine was the most suitable $\gamma$-glutamyl donor for the transfer reaction among those tested. Amino acids generally inhibited the enzyme activity for the transfer reaction, but not for the hydrolysis reaction.
Some chemical constituents were monitored to evaluate the biochemical and nutritional aspects of ${\gamma}-irradiated$ mushrooms associated with shelf-life extension. Volatile components identified by GC and GC-MS were composed primarily of 1-octen-3-ol(68%)). benzaldehyde(13%), 3-octanone(8%), benzyl alcohol(5%), 3-octanol(2%)). 1-octen-3one(1%). etc. Treatment with 2kGy-irradiation and subsequent storage for 17 days at $(9{\pm}1^{\circ}C\;and\;80{\pm}7%$ RH resulted in appreciable changes In their contents. even though negligible changes were observed in GC patterns between the nonirradiated and 2 kGy-irradiated samples. Most of the amino acids were resistant to ionizing energy of 2 kGy, while sulfur-containing free amino acids were affected significantly by ${\gamma}-irradiation$.
