가시오갈피나무의 정유성분 GC/MS 분석 (GC/MS Analysis of Volatile Constituents from Acanthopanax senticosus)

  • The chemical composition of the volatile constituents from the leaves, stems, and roots of Acanthopanax senticosus growing wild in Mt. Deok-Yu was determined by GC and GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were ${\delta}-elemene\;(23.14%),\;{\beta}-elemene\;(5.59%),\;{\gamma}-cadinene\;(5.43%),\;and\;{\alpha}-bergamotene\;(28.54%)$ in the leaves, ${\alpha}-pinene\;(11.50%),\;{\delta}-elemene\;(17.62%),\;{\alpha}-bergamotene\;(27.21%)$, and bicycloheptane derivative (6.38%) in the stems, and ${\alpha}-pinene\;(21.90%)$, and (+)-aromadendrene (3.77%) in the roots.

Cytotoxic and Mutagenic Effects of Cinnamomum cassia Bark-Derived Materials

  • The cytotoxic activities of Cinnamomum cassia (Blume) bark-derived materials toward six human HeLa epithelioid cervix, A549 lung, SK-OV-3 ovarian, SK-MEL-2 melanoma, XF-498 central nerve system, and HCT-15 colon tumor cell lines were evaluated by using sulforhodamine B assay and compared to those of the anticancer agents, cisplatin and mitomycin C. The biologically active constituent of the Cinnamomum bark was characterized as trans­cinnamaldehyde by spectroscopic analysis. The cytotoxic activity of cinnamaldehyde against HeLa, SK-MEL-2, and HCT -15 cell lines was comparable to that of cisplatin and mitomycin C. The compound showed lower activity against A549, SK-OV-3, and XF-498 cell lines than the anticancer agents. Eugenol exhibited moderate activity against SK-OV­3, XF-498, and HCT-15 tumor cells, and trans-cinnamic acid, cinnamyl alcohol, $\alpha-pinene,\;and\;\beta-pinene$ showed little or no activity against model tumor cells. Cinnamaldehyde was not mutagenic against four strains (TA 98, TA 100, TA 1535, and TA 1537) of Salmonella typhimurium (Castel and Chalm). These results indicate at least one pharmacological action of C. cassia.

ULTRA-FINE PARTICLES AND GASEOUS VOLATILE ORGANIC COMPOUND EXPOSURES FROM THE REACTION OF OZONE AND CAR-AIR FRESHENER DURING METROPOLIS TRAVEL

  • Experiments were conducted to identify the emissions from the car air freshener and to identify the formation of ultra-fine particles and secondary gaseous compounds during the ozone-initiated oxidations with emitted VOCs. The identified primary constituents emitted from the car air freshener in this study were $\alpha$-pinene, $\beta$-pinene, $\rho$-cymene and limonene. Formation of ultra-fine particles (4.4-160 nm) was observed when ozone was injected into the chamber containing emitted monoterpenes from the air freshener. Particle number concentrations, particle mass concentrations, and surface concentrations were measured in time dependent experiments to describe the particle formation and growth within the chamber. The irritating secondary gaseous products formed during the ozone-initiated reactions include formaldehyde, acetaldehyde, acrolein, acetone, and propionaldehyde. Ozone concentration (50 and 100 ppb) and temperature (30 and $40^{\circ}C$) significantly affect the formation of particles and gaseous products during the ozone-initiated reactions. The results obtained in this study provided an insight on the potential exposure of particles and irritating secondary products formed during the ozone-initiated reaction to passengers in confined spaces.

충북지역 자연휴양림의 피톤치드 발생 특성 (Characteristics of phytoncide production at the recreation forest in the Chungbuk area)

  • Phytoncide, which is emitted from plant against the insect species and bacterium, has been known as a health builder for human. Monoterpenes are major components of phytoncide. This study was conducted to investigate the distribution characteristics of phytoncide(monoterpene) depending on main species of tree, season, and meteorological factors in the 5 recreation forests, Chungbuk area. From the result of the study. it was shown that the annual concentrations of monoterpene are ranged in 236.0pptv - 698.3pptv depending on main species of tree. The variation of seasonal concentration was summer > spring > fall. The monoterpene concentration of coniferous forest was founded to be higher than broad leaved forest. In general, concentration of major components is ${\alpha}$-pinene > ${\beta}$-pinene > camphene > d-limonene. The concentration of terpene was founded to be greatly influenced by the meteological factors such as insolation and temperature.

Poly(dimethylsiloxane) Mini-disk Extraction

  • A novel sampling method of the headspace poly(dimethylsiloxane) (PDMS) mini-disk extraction (HS-PDE) was developed, optimized, validated and applied for the GC/MS analysis of spices flavors. A prototype PDMS mini-disk (8 mm outer diameter, 0.157 mm thickness, 9.4 mg weight) has been designed and fabricated as a sorption device. The technique uses a small PDMS mini-disk and very small volume of organic solvent and less sample size than the solvent extraction. This new HS-PDE method is very simple to use, inexpensive, rapid, requires less labor. Linearities of calibration curves for ${\alpha}$-pinene, ${\beta}$-pinene, limonene and ${\gamma}$-terpinene by HS-PDE combined with GC/MS were excellent having $r^2$ values greater than 0.99 at the dynamic range of 6.06~3500 ng/mL. The limit of detection (LOD) and the limit of quantitation (LOQ) showed very low values. This method exhibited good precision and accuracy. The overall extraction efficiency of this method was evaluated by using partition coefficients ($K_p$) and concentration factors (CF) for several characteristic components from nutmeg and mace. Partition coefficients were in the range from $2.04{\times}10^4$ to $4.42{\times}10^5$, while CF values were 0.88-15.03. HS-PDE was applied successfully for the analysis of flavors compositions from nutmeg, mace and cumin. The HS-PDE method is a very promising sampling technique for the characterization of volatile flavors.

GC/FID와 GC/MS 분석법에 의한 목질제품에서의 VOC 방출특성 비교 (The Comparison of VOC Characteristics Emitted from Wood-based Panels Using GC/FID and GC/MS)

  • In this study, the Total Volatile Organic Compounds (TVOC) emitted from wood-based panels were compared by two analytical methods using the GC/FID and the GC/MS. Japanese Larch, Yellow Poplar, Particle Board (PB) and Medium Density Fiberboard (MDF) were selected as target materials. Major compounds emitted from the panels were Toluene, ${\alpha}$-pinene, ${\beta}$-pinene and limonene. In case of TVOC using GC/FID method, MDF E2 (1,497 ${\mu}g/m^3$) revealed the highest concentration among all wood-based panels, while Japanese Larch (1,772 ${\mu}g/m^3$) showed the highest value with respect to GC/MS method. Furthermore, it was found that the amount of VOC emitted from panels was different depending upon analytical methods. This significant difference was attributed to analytical sensitivities of GC/FID and GC/MS for various VOC. Besides, it was found that the composition ratios of main VOC compounds were not significantly different.

산림치유 효능물질 함량에 관한 연구 (Research of Monoterpenes Content in the Atmosphere of Forest)

  • The biogenic emission of the atmospheric volatile organic compounds (VOCs) from the forests is dominated by monoterpenes. This study investigated the changes in the concentration of monoterpenes distributed in various types of forest near the Gangwon Nature Environment Research Park, Hongcheon, Gangwon Province, Korea. Samples were collected from the three sites of different types of forest, including coniferous, broad-leaved and mixed forests. Additionally, the seasonal and daily changes of monoterpene compounds were monitored. Our results found the several types of monoterpene such as ${\alpha}$-pinene, ${\beta}$-pinene, camphene, d-limonene, p-cymene and ${\alpha}$-terpinene. The highest total concentration of terpene compounds was observed in the coniferous forest. For the summer season, the total concentration of terpene compounds was highest in coniferous and mixed forests, and that was also highest in broad-leaved forest for the autumn.

Transfer of Orally Administered Terpenes in Goat Milk and Cheese

  • The objective of the present study was to investigate the relationships between terpenes… intake and their presence in animal tissues (blood and milk) as well as in the final product (cheese). Eight dairy goats were divided in two balanced groups, representing control (C) and treatment (T) group. In T group oral administration of a mixture of terpenes (${\alpha}$-pinene, limonene and ${\beta}$-caryophyllene) was applied over a period of 18 d. Cheese was produced, from C and T groups separately, on three time points, twice during the period of terpenes… oral administration and once after the end of experiment. Terpenes were identified in blood by extraction using petroleum ether and in milk and cheese by the use of solid phase micro-extraction (SPME) method, followed by GC-MS analysis. Chemical properties of the milk and the produced cheeses were analyzed and found not differing between the two groups. Limonene and ${\alpha}$-pinene were found in all blood and milk samples of the T group after a lag-phase of 3 d, while ${\beta}$-caryophyllene was determined only in few milk samples. Moreover, none of the terpenes were traced in blood and milk of C animals. In cheese, terpenes' concentrations presented a more complicated pattern implying that terpenes may not be reliable feed tracers. We concluded that monoterpenes can be regarded as potential feed tracers for authentification of goat milk, but further research is required on factors affecting their transfer.

빨간집모기(Culex pipiens pallens) 유충에 대한 식물오일의 살충효과 (Insecticidal Activity of Essential Oils against Larvae of Culex pipiens pallens)

  • 빨간집모기(Culex pipiens pallens) 유충에 대한 34종 식물오일의 살충력을 조사하였다. 80% 이상의 살충효과를 나타낸 오일은 6종(basil, fennel, caraway seed, lime, thyme red, thyme white)이었다. 각 오일의 구성성분을 GC와 GC-MS로 분석하고 각 성분의 살충력을 검정한 결과, basil 및 fennel 오일의 주요성분인 anethole과 caraway seed 오일 및 lime 오일의 주요성분인 (+)-limonene 이 높은 살충력을 나타내었다. 또한 ${\alpha}$-phellandrene, p-cymene, $\gamma$-terpinene, ${\beta}$-pinene, thymol 도 대조약제인 Bt제 보다 높은 살충효과를 나타내었다.

Antibacterial Activity of Essential Oil from Abies holophylla against Respiratory Tract Bacteria

  • We extracted essential oils from four species (Pinus densiflora, Larix kaempferi, Pinus koraiensis, and Abies holophylla) in the family Pinaceae to investigate their antibacterial activities against respiratory tract bacteria (Klebsiella pneumoniae, Haemophillus influenzae, Streptococcus pyogenes, Streptococcus pneumonia, and Neisseria meningitides). Among the tested oils, that from A. holophylla was showed strong activity based on disc diffusion and broth medium dilution (minimum inhibitory concentration, MIC) assays. Qualitative analysis of A. holophylla oil was carried out by GC-MS; ${\alpha}$-pinene, camphene, ${\beta}$-pinene, 3-carene, limonene, bornyl acetate, borneol, ${\beta}$-caryophyllene, ${\alpha}$-caryophyllene, caryophyllene oxide, and ${\alpha}$-bisabolol were identified as its major constituents. Fractionation by silica gel chromatography was performed to analyze the active constituents of the crude oil. In particular, one fraction containing caryophyllene oxide as the major constituent showed stronger antibacterial activity than the crude oil of A. holophylla. Growth rates of bacterial strains exposed to fraction D were explored by optical density (OD600) measurements while morphology was examined by optical microscopy observations ( ${\times}1000$). OD600 of K. pneumoniae decreased from 0.2582 to 0.005 in response to treatment with fraction D at a MIC value of $0.31{\mu}{\ell}/m{\ell}$.
