Mg-BCP(Mg Substituted BCP)를 제조하기 위하여 $Ca(NO_3)_2{\cdot}4H_2O$(Katayama chemical, Japan)과 $(NH_4)_2{\cdot}HPO_4$ (Junsei chemical, Japan), $Mg(NO_3)_2{\cdot}6H_2O$(Junsei chemical, Japan)를 출발물질로 공침법(co-precipitation process)을 이용하여 합성하였다. 제조된 분말의 마그네슘의 첨가에 따른 분말의 분광학적 특성은 FT-IR(MAGNA-IR 560, Nicolet)을 통하여 분석하였으며, SEM(S-4200, Hitachi)을 통하여 미세구조를 분석하였다. XRD 회절피크의 면적적분강도를 Integral Analysis (Rigaku, Japan)를 이용하여 HAp와 ${\beta}$-TCP의 비율을 확인하였다. 1.0 wt% 마그네슘이 첨가된 BCP 샘플의 경우 Hank's solution에서의 2주 침적 후 표면에 침상의 HAp로 추정되는 결정이 성장하였다. 이러한 미세구조의 변화는 생활성을 가지는 마그네슘의 첨가가 BCP 조직 내에서 표면의 활성을 증가시켜 결정의 성장을 촉진시킨 것으로 판단된다.
Various calcium phosphate bioceramics are distinguished by their excellent biocompatibility and osteoconductivity. Especially, the exceptional biodegradability of $\beta$-TCP makes it a bone graft substitute of choice in many clinical applications. The activation of osteoclasts, differentiated from macrophage precursor cells, trigger a cell-mediated resorption mechanism that renders $\beta$-TCP biodegradable. Based on this evidence, we studied the biodegradation process of granular-type $\beta$-TCP bone graft substitute through in vitro and in vivo studies. Raw 264.7 cells treated with RANKL and M-CSF differentiated into osteoclasts with macrophage-like properties, as observed with TRAP stain. These osteoclasts were cultured with $\beta$-TCP nano powders synthesized by microwave-assisted process. We confirmed the phagocytosis of osteoclasts by observing $\beta$-TCP particles in their phagosomes via electron microscopy. No damage to the osteoclasts during phagocytosis was observed, nor did the $\beta$-TCP powders show any sign of cytotoxicity. We also observed the histological changes in subcutaneous tissues of rats implanted with granule-type $\beta$-TCP synthesized by fibrous monolithic process. The $\beta$-TCP bone graft substitute was well surrounded with fibrous tissue, and 4 months after implantation, 60% of its mass had been biodegraded. Also, histological findings via H&E stain showed a higher level of infiltration of lymphocytes as well as macrophages around the granule-type $\beta$-TCP. From the results, we have concluded that macrophages play an important role in the biodegradation process of $\beta$-TCP bone graft substitutes.
수용액의 pH, 소성온도, 반응시간 등을 변화한 조건하에서 $Ca(OH)_2$와 $H_{3}PO_4$ 수용액을 이용하여 인산칼슘계 화합물을 제조하였다. 산성분위기(pH=5.21)에서는 ${\beta}-TCP({\beta}-tricalcium phosphate)$가, pH>7.62에서는 HAp(hydroxyapatite)가 주 결정상으로 관찰되었다. XRD 분석결과, $Ca(OH)_2$와 $H_{3}PO_4$는 빠르게 반응(5분이내)하여 HAp를 생성하였고, 반응 16시간까지 안정적으로 구조를 유지하였다. 우수한 결정성을 나타내는 소성온도는 $1200^{\circ}C$로 관찰되었다. 주사전자현미경(SEM)에 의한 미세구조 관찰 결과, pH가 5.21일 경우 침상 형태의 ${\beta}-TCP$ 입자가, pH 7.62에서는 침상 형태의 HAp가 합성되었다. 그러나 pH 9.16에서는 매우 작고 균일한 구형의 HAp 입자들이 생성되었고, 다시 작은 입자들이 침상으로 재배열된 형상을 관찰할 수 있었다. 합성된 HAp(pH 9.16)를 본차이나 소지에 일정량 혼합하여 본차이나 물성, 즉 결정성, 흡수율, 색도, 파단면의 형상을 측정${\cdot}$분석하였다.
Purpose: Biphasic calcium phosphates have been of great interest recently. Mixing adequate ratios of hydroxyapatite(HA) and beta-tricalcium phosphate($\beta$-TCP) allowed to control the resorption rate without distorting its osteoconductive property. This study evaluated the bone formation effect of newly developed biphasic calcium phosphate(BCP) in calvarial defect of rabbits. Materials and Methods: 6 male New Zealand rabbits were used. Four defects with 8mm in diameter were created on each animal. BCP with HA/$\beta$-TCP ratio of 7:3 and particle size of $0.5{\sim}1.0\;mm$ was used as the test group and bovine bone with $0.25{\sim}1.0\;mm$ particle size, as the control group. Both test and control group materials were randomly implanted in the calvarial defects and were covered witha polymer membrane. The animals were sacrificed after 12, 24, and 48 weeks of implantation under general euthanasia. Resin blocks were obtained and were stained by masson's trichrome for histological observation. Results: Overall results were uneventful without any defect exposure or inflammation. The amount of new bone formation and bone maturity increased with increase in healing period at both groups. New bone in test group was mostly formed along the material particle surrounded by osteoblasts, and observation of osteoblastic stream was also present. Bone maturity increased as it was closer to thedefect margins. Under the same healing period, the test group showed more bone formation than the control group with more stable bovine bone particles remaining even after 48 weeks, whereas considerable resorption took place in BCP. Almost total defect closure was observed in test group with new bone formation in the central part of the defect. However, limited new bone formation was observed in the control group. Conclusion: Within the limits of the study, the present study reveals the newly developed BCP to be a good osteoconductive material. However, further studies are needed to be conducted in a different study model with a larger sample size.

Background: Autogenous bone grafts have several limitations including donor-site problems and insufficient bone volume. To address these limitations, research on bone regeneration is being conducted actively. In this study, we investigate the effects of a three-dimensionally (3D) printed polycaprolactone (PCL)/tricalcium phosphate (TCP) scaffold on the osteogenic differentiation potential of adipose tissue-derived stem cells (ADSCs) and bone marrow-derived stem cells (BMSCs). Methods: We investigated the extent of osteogenic differentiation on the first and tenth day and fourth week after cell culture. Cytotoxicity of the 3D printed $PCL/{\beta}-TCP$ scaffold was evaluated by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay, prior to osteogenic differentiation analysis. ADSCs and BMSCs were divided into three groups: C, only cultured cells; M, cells cultured in the 3D printed $PCL/{\beta}-TCP$ scaffold; D, cells cultured in the 3D printed $PCL/{\beta}-TCP$ scaffold with a bone differentiation medium. Alkaline phosphatase (ALP) activity assay, von Kossa staining, reverse transcription-polymerase chain reaction (RT-PCR), and Western blotting were performed for comparative analysis. Results: ALP assay and von Kossa staining revealed that group M had higher levels of osteogenic differentiation compared to group C. RT-PCR showed that gene expression was higher in group M than in group C, indicating that, compared to group C, osteogenic differentiation was more extensive in group M. Expression levels of proteins involved in ossification were higher in group M, as per the Western blotting results. Conclusion: Osteogenic differentiation was increased in mesenchymal stromal cells (MSCs) cultured in the 3D printed PCL/TCP scaffold compared to the control group. Osteogenic differentiation activity of MSCs cultured in the 3D printed PCL/TCP scaffold was lower than that of cells cultured on the scaffold in bone differentiation medium. Collectively, these results indicate that the 3D printed PCL/TCP scaffold promoted osteogenic differentiation of MSCs and may be widely used for bone tissue engineering.

Background: This study is performed to evaluate anchor-related outcomes and complications after arthroscopic rotator cuff repair using 30% ${\beta}$-tricalcium phosphate (${\beta}$-TCP) with 70% poly lactic-co-glycolic acid (PLGA) biocomposite suture anchors. Methods: A total of 78 patients (mean age, $61.3{\pm}6.9years$) who underwent arthroscopic medium-to-large full-thickness rotator cuff tear repair were enrolled. The technique employed 30% ${\beta}$-TCP with 70% PLGA biocomposite suture anchors at the medial row (38 patients, Healix $BR^{TM}$ anchor [Healix group]; 40 patients, Fixone anchor B [Fixone group]). The radiologic outcomes (including perianchor cyst formation or bone substitution) and anatomical outcomes of the healing failure rate were evaluated using magnetic resonance imaging at least 6 months after surgery, the pain visual analogue scale at 3, 6 months, and final follow-up visit, and American Shoulder and Elbow Surgeons scores at least 1 year postoperatively. Anchor-related complications were also evaluated. Results: The perianchor cyst formation incidence was similar for both groups (60.5%, Healix group; 60.0%, Fixone group; p=0.967), although severe perianchor cyst incidence was slightly lower in the Fixone group (15.0%) than in the Healix group (21.1%). There was no occurrence of anchor absorption and bone substitution. No differences were observed in the healing failure rate (13.2%, Healix group; 15.0%, Fixone group; p=0.815) and functional outcome between groups (all p>0.05). Anchor breakage occurred in 5 patients (2 Healix anchors and 3 Fixone anchors); however, there were no major anchor-related complications in either group. Conclusions: No differences were observed in the clinical outcomes of the Healix and Fixone groups, neither were there any accompanying major anchor-related complications.
전복패각을 Ca원으로 이용하여 바이오 소재로 중요한 인산칼슘(calcium phosphates)들을 침전법을 통해 합성하였다. 전복패각에서 유래한 수산화칼슘(Ca(OH)2)을 칼슘 공급원(전구체)으로 사용하였다. 수용액상에서 수산화칼슘과 인산(H3PO4)을 반응시켜 침전반응을 유도하여 인산칼슘화합물로의 합성을 유도하였다. 초기 전구체 Ca/P 비율을 1.50, 1.59 및 1.67로 조절하였으며 이 조성변화와 침전물에 대한 열처리가 분말 및 소결체의 물성에 미치는 영향에 대해 조사하였다. 초기 전구체 Ca/P 비율을 조절함으로써 소결체 상합성의 조절이 가능하였고, 1150℃에서 소결한 소결체에서(hydroxyapatite(HAp), β-tricalcium phosphate(β-TCP) 및 HAp와 β-TCP가 혼합된 2상 인산칼슘(BCP, HA/β-TCP))들이 합성되었다. 이러한 결과는 저비용, 고가용성을 가지는 경제적인 출발물질로부터 고부가가치 인산칼슘을 합성할 수 있는 가능성을 보여주었다.
조직 공학에 있어 인공지지체는 손상된 조직 및 기관의 기능을 재생하기 위한 거푸집으로 제공되며 3 차원 구조물이다. 인공지지체의 재료 중에서 폴리카프로락톤(Polycaprolactone, PCL)과 삼인산칼슘(${\beta}$-tricalcium phosphate, ${\beta}$-TCP)은 생분해성과 생체적합성을 가지고 있다. 본 연구에서는 다축 인공지지체 제작 시스템을 이용하여 3 차원 PCL, blended PCL(60 wt %)/${\beta}$-TCP(40 wt %), 그리고 ${\beta}$-TCP 인공지지체를 제작하였다. 제작된 인공지지체는 주사전자현미경 분석을 통해 $600{\pm}20{\mu}m$의 공극 크기로 잘 제작되었다. 기계적 특성 평가를 통해 3 차원 PCL, blended PCL(60 wt %)/${\beta}$-TCP(40 wt %), 그리고 ${\beta}$-TCP 인공지지체의 효과는 분석되었다. 게다가 Saos-2 세포를 이용한 in vitro 연구를 수행하여 세포 증착 및 증식과 같은 세포 거동에 의한 3 차원 인공지지체의 효과를 확인하였다. 요컨대 3D blended PCL(60 wt %)/${\beta}$-TCP(40 wt %) 인공지지체가 압축 강도와 생체적합성 그리고 골전도성에 있어서 인체의 해면골에 더욱 적합하였다. 따라서 3D 인공지지체의 제작에 있어 PCL과 ${\beta}$-TCP를 혼합하는 것은 효과적인 골 재생을 위해 촉망되는 전략이 될 것이다.
In this study, the fracture strength for fracture mechanism porous calcium phosphate made from sintered with ${\beta}$-tricalcium phosphate obtained by wet precipitation procedure is analyzed using finite element method and experiment measurement. First, three $3{\times}3{\times}3mm^3$ and $5{\times}5{\times}5mm^3$ specimens are prepared and tomographic images of one $5{\times}5{\times}5mm^3$ specimen are obtained by micro focus X-ray CT. The compression tests using the specimens are carried out to measure the elastic modulus and fracture strength to analyze the fracture mechanism of porous calcium phosphate specimen. The tomographic images are reconstructed by 3D reconstruction program. The finite elements are directly built up in the reconstructed specimen. The numerical simulation for the compression tests is performed using the element. The mechanism of calcium phosphate of simulation are obtained by the compression tests using there cylindric specimen of height 19.5 mm and diameter 10 mm. From the results, the applicability of porous calcium phosphate is evaluated to care fracture and vacant bone of a patient as the reinforcement material.
