Cloning and Characterization of ${\alpha}-Glucosidase$ Gene from Thermophilic Bacillus sp. DG0303

  • An ${\alpha}-glucosidase$ gene (aglA) from thermophilic Bacillus sp. DG0303 was cloned, sequenced, and expressed in Escherichia coli. The aglA was localized to the 2.1-kb PvuI-XmnI region within the 5.9-kb DNA insert of the gybrid plasmid pAG1. The gene consisted of an open reading frame of 1,686 bp with an unusual GTG initiation codon and TGA termination codon. The amino acid sequence deduced from the nucleotide sequence predicted a protein of 562 amino acid residues with a M, of 66,551 dalton. A comparative amino acid sequence analysis revealed that DG0303 ${\alpha}-glucosidase$ is related to bacillary oligo-1, 6-glucosidases. The Bacillus sp. DG0303 ${\alpha}-glucosidase$ showed a high sequence identity (36-59%) to the B. flavocaldarius, B. cereus, and B. thermoglucosidasius oligo-1, 6-glucosidases. The number of prolines in theses four ${\alpha}-glucosidases. was observed to increase with increasing thermostability of these enzymes. The cloned ${\alpha}-glucosidase was purified from E. coli $DH5{\alpha}$ bearing pAG1 and characterized. The recombinant enzyme was identical with the native enzyme in its optimum pH and in its molecular mass, estimated by sodium dodecy1 sulfate-polyacrylamide gel electrophoresis. The temperature optimum of the cloned ${\alpha}-glucosidase$ was lower than that of the native enzyme.

Effect of Artificially Cultured Phellinus linteus on Harmful Intestinal Bacterial Enzymes and Rat Intestinal ${\alpha}-glucosidases$ (재배상황버섯의 장내 세균 유해효소 및 알파글루코시다제 저해효과)

  • The objective of this study was to evaluate the bifidogenic effect and a-glucosidase inhibitory effect of artificially cultured Phellinus linteus. The water extract of P. linteus promoted the growth of Bifidobacterium breve as well as the decrease of final pH in the media culturing intestinal bacteria. The growth of lactic acid bacteria inhibited effectively the bacterial enzymes, ${\beta}-glucosidase,\;{\beta}-glucuronidase$ and tryptophanase, of intestinal bactetria. The water extract of P. linteus inhibited maltase, sucrase and ${\alpha}-amylase$ of rat intestine.

  • $\alpha$-Glucosidase contributing $\alpha$-glucan metabolism in microorganisms is characterized by the variety in substrate recognition. Recent studies on microbial enzymes show that $\alpha$-glucosidases are divided into two groups, family I and family n, in which family I enzymes have four conserved catalytic-regions of $\alpha$-amylase family. The presentation focusing on the difference of the $\alpha$-glucosidase families reviews i) the catalytic amino-acid residues of nucleophile and acid/base catalyst, and ii) the molecular evolution of two families.

Inhibitory Effect of GE974 isolated from Gyrophora esculenta on ${\alpha}-Glucosidase$ (석이에서 분리한 GE974의 ${\alpha}-Glucosidase$ 저해효과)

  • This study was conducted to search for the ${\alpha}-glucosidase$ inhibitor from the natural products. In the previous study, the water extract of Gyrophora esculenta exhibited a potent inhibitory effect on ${\alpha}-glucosidase$ activities. Then, by bioassay-guided fractionation followed by chromatographic separation of the water extract of Gyrophora esculenta, ${\alpha}-glucosidase$ inhibitor was isolated as GE974. GE974 showed significant inhibitory activities on some kinds of ${\alpha}-glucosidase$ in vitro. Its inhibitory mechanism seemed to be competitive for disaccharides. Also, it markedly inhibited ${\alpha}-glucosidases$ of intestine separated from both nondiabetics and diabetics.

  • Cyclodextrin glucanotransferase from Bacillus stearothemophilus and Bacillus macerans synthesized maltol and ethyl maltol monoglucoside, with a series of its maltooligo-glucosides by transglycosylation with dextrin as a donor, and maltol or ethyl maltol as an acceptor. The monoglucoside formed from reaction mixture of maltol or ethyl maltol by the successive actions of Bacillus stearothemophilus cyclodextrin glucanotransferase and Rhizopus glucoamylase was isolated by Diaion HP-20 column and silica gel column chromatography. The structure of the isolated monoglucoside was identified as maltol-$\alpha$-D-glucoside and ethyl maltol-$\alpha$-D-glucoside, respectively, by FAB-MS, UV, $^1$H-NMR, $^{13}$ C-NMR spectra and products by hydrolysis with acid, $\alpha$ - and $\beta$ -glucosidases.

  • Cyclodextrin glucanotransferase (CGTase) from Bacillus stearothermophilus synthesized vanillin and ethyl vanillin monoglucoside, with a series of its maltooligoglucosides by transglycosylation with dextrin as a donor, and vanillin or ethyl vanillin as an acceptor. The monoglucoside formed from reaction mixture of vanillin or ethyl vanillin by the successive actions of CGTase and Rhizopus glucoamylase was isolated by extraction with n-butanol saturated with water and silica gel column chromatography. The structure of the isolated monoglucoside was identified as vanillin- $\alpha$ -D-glucoside and ethyl vanillin- $\alpha$ -D-glucoside, respectively, by FAB-MS, UV, IR, 1H-NMR, 13C-NMR spectra and products by hydrolysis with acid, $\alpha$ - and $\beta$ -glucosidases.

Comparative Analysis of $\alpha$-glucosidase Activity in Bombyx mori and Antheraea yamamai

  • [ $\alpha$ ]Glucosidase (EC 3.2.1.20) is a glycosidase that hydrolyzes disaccharides, oligosaccharides, and polysaccharides resulting in the release of α-D-glucose. In this study, $\alpha$-glucosidase activity in the hemolymph and midgut of the mulberry silkworm Bombyx mori and Japanese oak silkmoth Antheraea yamamai was measured using maltose, sucrose, trehalose, and p-nitrophenyl $\alpha$-D-glucopyranoside as substrates. In general, hemolymph $\alpha$-glucosidase activity was higher in B. mori than in A. yamamai. In contrast, midgut $\alpha$-glucosidase activity was higher in A. yamamai than in B. mori for all of the substrates tested. $\alpha$-Glucosidase activity in the midgut of both B. mori and A. yamamai showed similar responses to changes in pH and temperature for all of the substrates tested. Native (7.5%) PAGE of hemolymph and midgut proteins from B. mori and A. yamamai followed by staining with 4-methylumbelliferyl $\alpha$-D-glucoside (MUG) indicated that the $\alpha$-glucosidases of these related lepidopterans are functionally similar but structurally different. In comparison to $\alpha$-glucosidase activity from A. yamamai, $\alpha$-glucosidase activity from B. mori was generally less sensitive to the $\alpha$-glucosidase inhibitors, 1-deoxynojirimycin (DNJ), acarbose, and voglibose when the activity was determined using maltose, sucrose, and trehalose.

Inhibitory Effects of Crude Drugs on $\alpha$-Glucosidase

  • The inhibitory activity of several crude drugs on $\alpha$-glucosidases, which are the key enzymes for carbohydrate digestion and the prevention of diabetic complications, was investigated. Several crude drugs including Terminaliae Fructus, Mori Cortex Radicis, Caesalpiniae Lignum and Cyrophora esculenta potently inhibited maltase and sucrase isolated from rat intestine, while Arecae Semen and Corni Fructus remarkably inhibited $\alpha$-amylase. Caesalpiniae Lignum and Gyrophora esculenta exhibited significant reductions of blood glucose elevation in mice loaded with maltose and sucrose.

  • $\beta$ -Glucosidase-catalysed synthesis of glucosides with aromatic alcohols and monoterpene alcohols as accepters and cellobiose as a donor in the presence of various commercial $\beta$ -glucosidases were described. $\beta$ -Glucosidases from Aspergillus niger spp,. Trichoderma spp., Penicillium sup. and bitter almond have been shown to catalyze synthesis of $\beta$ -glucosides of benzyl alcohol, 2-hydroxybenzyl alcohol, 4-hydroxybenzyl alcohol, 2-phenylethyl alcohol, geraniol and citronellol in the presence of cellobiose as sugar donor. Among enzyme preparations tested, each $\beta$ -glucosides prepared from Aspergillus niger were isolated in the pure state by Diaion HP-20 and silica gel column chromatography. The products were identified as $\beta$ -glucosyl products of benzyl alcohol, 2-hydroxyhenzyl alcohol, 4-hydroxybenzyl alcohol, 2-phenyl ethyl alcohol, geraniol and citronellol by spectrometry (UV, IR, $^1$H-NMR, $^{13}$ C-NMR) and enzymatic hydrolysis with $\beta$ - glucosidase. Monoterpene alcohols with a sterically hindered hydroxyl group, such as linalool, $\ell$-menthol and $\alpha$-terpineol were not used as acceptors in transglycosylation reaction.

Hypoglycemic Effect of GE974 isolated from Gyrophora esculenta in Normal and Diabetic mice (석이에서 분리한 GE974의 혈당상승억제효과)

  • GE974, isolated from Gyrophora esculenta, showed significant inhibitory effect on several ${\alpha}-glucosidases$ in vitro in previous study. In the present study, GE974 showed significant inhibitory effect on blood glucose elevation in mice loaded with maltose, sucrose, starch and lactose. Also, it exhibited similar effect in alloxan and streptozotocin induced diabetic mice, and genetic diabetic mice(db/db mice) loaded with maltose in dose dependent manner. These results suggest that GE974 may possess hypoglycemic effect that inhibit competitively ${\alpha}-glucosidase$ activities.
