Biological Activity of Viola mandshurica Fermented with Bacillus methylotrophicus CBMB205 and Leuconostoc pseudomesenteroides NRIC1777 (미생물 분리주를 이용한 제비꽃 발효액의 생리활성 변화)

  • The aim of this study was to improve the efficacy and functionality of Viola mandshurica (VM). A water suspension of VM power was fermented for 72 hr with Bacillus methylotrophicus CBMB205 (BM) and Leuconostoc pseudomesenteroides NRIC1777 (LP) isolated from kimchi. The antioxidant activity and reducing power of fermented VM, its total phenolic and flavonoid compounds, as well its inhibitory activity on ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase were determined and compared to those of non-fermented VM (NVM), a negative control. The total phenolic and flavonoid compounds of VM fermented with BM and LP were higher than those of NVM by 1.4, 1.17, and about 3 times. There was no difference in 2, 2'-diphenyl-1-picryl hydrazyl (DPPH) radical scavenging activity between fermented VM and NVM. However, there was a 2.1- and 1.6-fold increase in 2, 2'-azino-bis-(3-ethylbenzothzoline-6-sulfonic acid) (ABTS) radical scavenging activity in VM fermented with BM and LP, respectively. The reducing power of BM was 1.6 times as high as NVM, but no significant difference was found between LP and NVM. Fermented VM's inhibitory activity on ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase was much higher than that of NVM. Fermenting VM with BM was superior to fermenting it with LP, except flavonoid content. Taken together, VM fermented with BM could be used as a functional food and as an additive to cosmetics.

  • The herbicidal activity of thiocarbamate herbicides and its effect on cell division and elongation were determined. The herbicides studied were molinate(S-ethyl-N,N-hexamethylenethiocarbamate), dimepiperate [S-(${\alpha}$,${\alpha}$-di methylbenzyle)piperidine-1-carbothioate], esprocarb [S-benzyl-N-ethyl-N-(1,2-dimethylpropyl) thiocarbamate], and thiobencarb [S-(4-chlorobenzyl)-N, N-diethylthiocarbamate]. The herbicides applied at the rates ranged from $10^{-6}$ to $10^{-5}M$ did not affect germination and post-germination root growth of rice(Oryza sativa L.) and barnyardgrass [Echinochloa crus-galli(L.) P. Beauv.], but inhibited the post-germination shoot growth. There was no inhibition of gibberellin-induced ${\alpha}$-amylase biosynthesis in de-embryonated rice seeds by the herbicides at $10^{-5}M$, but about 50-60% inhibition occurred at $10^{-4}M$. When the herbicides were applied 7 days after seeding, the rates required to 50% growth inhibition of barnyardgrass were 146g, 91g, 96g, and 102g ai/10a for molinate, dimepiperate, esprocarb, and thiobencarb, respectively. No effect of the herbicides on cell division was found at $10^{-4}M$, but about 31 to 47% inhibition as compared with the untreated check was obtained by treatment of the herbicides at $10^{-3}M$. However, about 33 to 38% inhibition of cell elongation occurred at $10^{-5}M$ of the herbicides. They also inhibited IAA-induced cell elongation.

Nutritional Characteristics and Screening of Biological Activity of Crataegi fructus (산사 영양성분 분석 및 생리활성 탐색)

  • The purpose of this study is to determine the possibility of using Crataegi fructus as a natural food source. To accomplish this purpose, the contents of general and biological activities were measured. The contents of carbohydrate, crude protein, crude lipid and ash are 85.6%, 2.4%, 1.9% and 0.4%, respeectively. Further, the calories of Crataegi fructus was 369.1 kcal. The contents of essential and non-essential amino acids were 852.26 mg and 1,178.29 mg, respeectively. The K was the largest mineral followed by Ca, P, Mg, which means Crataegi fructus is an alkali material. Crataegi fructus extracts slightly(17.6~32.8) inhibited ${\alpha}$-glucosidase activity. However, there is no inhibitory activity against ${\alpha}$-amylase. In terms of proteslytic activity, Crataegi fructus extracts showed a strong activity than pancreatin(used as a positive control). These results indicate that Crataegi fructus can be used as a natural resource for material aiding digestion.

Component Analysis and Digestive Enzyme Activities of Fermented Crataegi Fructus Extracts (산사 발효액의 함유 성분 분석 및 소화 활성)

  • Currently many studies aimed at enhancing efficacy of medicinal food on biological activity using bioconversion technology including fermentation process. In this study, the quality characteristics and antioxidative activity of fermented Crataegi fructus was investigated. The antioxidant activity of fermented Crataegi Fructus was assessed by various radical scavenging assays using DPPH (2,2-Diphenyl-1-picrylhydrazyl), FRAP (Ferric ion reducing antioxidant power), Reducing power and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture content of fermented Crataegi Fructus was $39.3{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.20{\pm}0.01$, $1.77{\pm}0.04$, and $1.40{\pm}0.59%$, respectively. Moreover, the hunter's color values of fermented Crataegi Fructus were 79.24 (lightnees), 1.58 (redness), and 31.25 (yellowness), respectively. Total phenolic contents of fermented Crataegi Fructus were $3,015{\pm}250$ GAE ${\mu}g/g$. The antioxidative activities of fermented Crataegi Fructus significantly increased in a dose dependent manner. In addition, fermented Crataegi Fructus slightly (10.4%) inhibited ${\alpha}$-glucosidase activity; however, there was no inhibitory activity against ${\alpha}$-amylase. In terms of proteolytic activity, fermented Crataegi Fructus showed a strong activity than pancreatin (used as a positive control). These results indicate that fermented Crataegi Fructus can be used as a natural resource for material aiding digestion.

Enzymatic Properties of Barley $\alpha$-Amylase Chimeric Enzymes Produced by Staggered Extension Process (Staggered Extension Process를 통해 제조한 보리 알파아밀라제 Chimera 효소의 특성)

  • Barley malt produces two different $\alpha$-amylase isozymes (AMY1 and AMY2), which share up to 80% of amino acid sequence identity with each other. However, their enzymatic properties differ remarkably. In this study, five chimeric enzymes between AMY1 and 2 were constructed by staggered extension process (StEP) technique, and their enzymatic properties were characterized. According to the results, chimeric AMY-D2, D8, and E12 showed the mixed or intermediate types of calcium-dependent activity between AMY1 and 2. Meanwhile, only AMY-E10 chimera could be significantly inhibited by barley $\alpha$-amylase/subtilisin inhibitor (BASI) protein. Chimera AMY-C6 showed the same calcium-dependency as AMY1, while AMY-E10 was closely similar to AMY2. As a result, it can be proposed that some amino acid residues in the region II, III, and IV of barley $\alpha$-amylases can play very important roles in the interaction with BASI, and those in III, V, VI, and VII may partly affect on the calcium-dependent activity.

  • This study examined the inhibitory activity of Ecklonia cava (EC) against ${\alpha}$-amylase to evaluate the availability of EC extract as a functional food agent. To verify the inhibitory activity of EC against porcine pancreatic ${\alpha}$-amylase, potato starch was used as a substrate. This analysis revealed that EC ethanol extract exhibited high ${\alpha}$-amylase inhibitory activity. For potential application within the food industry, the stability of the activity of EC ethanol extract under various heat and pH conditions was examined. The ${\alpha}$-amylase inhibitory activity of EC ethanol extract was not affected by the heat and pH treatment conditions used in this study. These results suggest that EC has the potential for development as a functional food agent.

  • The objective of this study was to evaluate the bifidogenic effect and a-glucosidase inhibitory effect of artificially cultured Phellinus linteus. The water extract of P. linteus promoted the growth of Bifidobacterium breve as well as the decrease of final pH in the media culturing intestinal bacteria. The growth of lactic acid bacteria inhibited effectively the bacterial enzymes, ${\beta}-glucosidase,\;{\beta}-glucuronidase$ and tryptophanase, of intestinal bactetria. The water extract of P. linteus inhibited maltase, sucrase and ${\alpha}-amylase$ of rat intestine.

Inhibitory Effects of Various Mulberry Fruits (Morus alba L.) on Related Enzymes to Adult Disease (품종이 다른 오디(Morus alba L.)의 성인병 관련 효소 억제효과)

  • The objective of this research was to develop the functional material of water and 60% ethanol extracts from nine kinds of mulberry fruits (Morus alba L.) that influence the inhibitory activity on angiotensin-converting enzyme, xanthine oxidase, ${\alpha}$-amylase, and ${\alpha}$-glucosidase. The total phenolic contents in the water extracts were over 2 mg/g in two species (Cheongilppong and Kangwon III) and five species (Daeyoupchosaeng, Cheongilppong, Kangwon III, Hihak, and Cataneo) of 60% ethanol extracts. The inhibitory activity against the angiotensin-converting enzyme was determined with them. Baekwoon III was $90.9{\pm}4.5%$ in the water extracts, and Hihak was $81.8{\pm}4.5%$ in the 60% ethanol extracts. The inhibitory activity of Kuksang 20 against xanthin oxidase was about 10% in the water extracts, and Cataneo was $21.4{\pm}2.3%$ in the 60% ethanol extracts. Six of the species (Daeyoupchosaeng, Suwonppong, Cheongilppong, Kangwon III, Hihak, and Kuksang 20) in the water extracts showed inhibitory activities against ${\alpha}$-amylase, as 100%, respectively. The inhibitory activity of ${\alpha}$-glucosidase was determined for these nine species. Four species (Baekwoon III, Daeyoupchosaeng, Cheongilppong, Kangwon III, Hihak, and Kuksang 20) in the water extracts and three species (Daechoukmyeun, Kangwon III, and Kuksang 20) in the 60% ethanol extracts showed inhibition of over 20%. The results revealed strong biological activity in spite of little total phenolic contents. These water and 60% ethanol extracts with high-quality biological activity from various mulberry fruits (Morus alba L.) are expected to represent good candidates for the development of antihypertentive and antidiabetes sources.

Screening of Biological Activities of Extracts from Rhododendron mucronulatum Turcz. Flowers (진달래꽃(Rhododendron mucronulatum Turcz. Flower) 추출물의 생리활성 탐색)

  • Extracts from Rododendron mucronulatum Turcz. flowers were tested for antioxidant and their inhibitory activities of ${\alpha}$-amylase, ${\alpha}$-glucosidase and angiotensin converting enzyme (ACE). Total contents of phenolics were found as $30.6{\pm}0.14mg/g$ (60% EtOH extract) and $23.2{\pm}0.21mg/g$ (water extract). Electron donation ability (EDA), ABTS [2,2azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical decolorization, Antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARs) were measured for the antioxidative activity of the extracts from Rododendron mucronulatum Turcz. flowers. The water extract were determined as 97.5% at ethanol extract showed 83.2% and 60% EtOH extract were 89.7% in EDA. The water extract showed higher antioxidant activity than 60% EtOH extract when evaluated by ABTS radical decolorization and antioxidant PF. The TBARS of water extracts and 60% EtOH extracts were shown as $0.29{\times}10^2{\mu}M\;and\;0.28{\times}10^2{\mu}M$, respectively, and were lower than control. ACE inhibitory activity in water extract (67.6% inhibition) was higher than that of 60% EtOH extract (46.7% inhibition) at $200{\mu}g/mL$. Water extracts had higher inhibitory activities on ${\alpha}$-amylase and ${\alpha}$-glucosidase than 60% EtOH extracts. The result suggests that the water extract from Rododendron mucronulatum Turcz. flowers will be useful as natural antioxidants and functional foods.

Biological Activity of Extracts from Cherry Sage (Salvia officinalis L.) (체리 세이지(Salvia officinalis L.) 추출물의 생리활성 탐색)

  • In this study, extracts from S. officinalis were tested for antioxidative effects and inhibitory activities against $\alpha$-amylase, angiotensin converting enzyme (ACE) and xanthine oxidase (XOase). The content of total phenolic compounds in water, 60% ethanol, 60% methanol and 60% acetone extracts were 36.2, 42.7, 40.3 and 39.6 mg/g, respectively. In 60% ethanol extracts, the EDA by DPPH free radical scavenging test of S. officinalis was $66.3{\pm}2.2%$ at $200{\mu}g/ml$. The inhibition rate of ABTS was $97.6{\pm}0.1%$, the antioxidant protection factor was $2.26{\pm}0.63$ PF, and TBARS was $0.62{\pm}0.05$ (${\times}100{\mu}M$ in the control and $0.29{\pm}0.02$ (${\times}100{\mu}M$). Also in 60% ethanol extracts of S. officinalis, the inhibitory activity against XOase was 78% and was not shown to be against ACE. According to the $12.6{\pm}0.14\;mm$ of clear zone formed, the inhibition rate against $\alpha$-Amylase was 7.6% at $200{\mu}g/ml$ of phenolics content. Antimicrobial activities of 60% ethanol extracts of S. officinalis against Helicobacter pylori exhibited an inhibition rate of $12.5{\sim}66.1%$ according to the $10{\sim}15\;mm$ of clear zone at $50{\sim}200{\mu}g/ml$. The results suggest that the 60% ethanol extracts from Salvia officinalis L. will be useful as natural antioxidants and functional food sources.
