The Constraint for Oxidation of Ascorbic Acid (Ascorbic Acid의 산화억제)

  • Ascorbic acid which has various physiological benefits as the functional substance is easily oxidized and destroyed by the structural instability. Liposome encapsulated pure ascorbic acid was prepared for the sake of the constraint of oxidation. The influence of cholestrol or $\beta$-sitosterol on the stabilization of liposome was investigated. Butylated hydroxytoluene(BHT), tertiary butylhydroquinone(TBHQ), $\alpha$ -glycosyl rutin and natural concentrated tocopherol were used for constraint of oxidation of ascorbic acid. The presence of cholesterol or $\beta$-sitosterol decreased oxidation of ascorbic acid. That results were thought that cholesterol or $\beta$-sitosterol so increased rigidity of bilayer that the leakage of vitamin C decreased. As a result the oxidation and degradation of vitamin C were constrained. At 0.3w/w% cholesterol content the most stable liposome was formulated. The whole antioxidant that used at the research constrained oxidation of ascorbic acid. The antioxidation for ascorbic acid increased in order of tertiary butylhydroquinone, $\alpha$-glycosyl rutin, butylated hydroxytoluene and natural concentrated tocopherol. But u -glycosyl rutin is preferable to tertiary butylhydroquinone which was the most effective in antioxidation as the antioxidant of ascorbic acid which was utilized in cosmetics and pharmacy.

Analysis of Vitamin E in Some Commonly Consumed Foods in Korea (국내에서 소비되는 일부 상용 식품의 비타민 E함량 분석)

  • Tocopherol and tocotrienol contents of commonly consumed foods in Korea were determined by saponification or direct solvent extraction followed by normal phase liquid chromatography. All samples were locally obtained in the year of 2002 and 2003. The study included 13 meats, 15 fishes and shellfishes, 4 seaweeds, 7 mushrooms, 19 milk and milk products, 6 legumes, 12 nuts, 17 processed foods and 17 Korean traditional foods. All of the vitamin E isomers were quantitated and the results were expressed as $\alpha-tocopherol$ equivalent $(\alpha-TE)$. The relatively higher amount of vitamin E was found in beef boiled in soy, dried squid, toasted seaweed, milk powder, soritae, sunflower seeds, ramyon (instant noodle), and kochujang from meats, fishes, seaweeds, milk products, legumes, processed foods, and traditional foods, respectively. This study provided reliable vitamin E data in commonly consumed foods in Korea for the nutritional information and food composition database.

Protective Effects of $\alpha$-Tocopherol and Ischemic Preconditioning on Hepatic Reperfusion Injury

  • This study evaluated the effect of $\alpha$-tocopherol ($\alpha$-TC), ischemic preconditioning (IPC) or a combination on the extent of mitochondrial injury caused by hepatic ischemia/reperfusion (I/R). Rats were pretreated with $\alpha$-TC (20 mg/kg per day, i.p.) for 3 days before sustained ischemia. A rat liver was preconditioned with 10 min of ischemia and 10 min of reperfusion, and was then subjected to 90 min of ischemia followed by 5 h or 24 h of reperfusion. I/R increased the aminotransferase activity and mitochondrial lipid peroxidation, whereas it decreased the mitochondrial glutamate dehydrogenase activity. $\alpha$-TC and IPC individually attenuated these changes. $\alpha$-TC combined with IPC ($\alpha$-TC+IPC) did not further attenuate the changes. The mitochondrial glutathione content decreased after 5 h reperfusion. This decrease was attenuated by $\alpha$-TC, IPC, and $\alpha$-TC+IPC. The significant production of peroxides observed after 10 min reperfusion subsequent to sustained ischemia was attenuated by $\alpha$-TC, IPC, and $\alpha$-TC+IPC. The mitochondria isolated after I/R were rapidly swollen. However, this swelling rate was reduced by $\alpha$­TC, IPC, and $\alpha$-TC+IPC. These results suggest that either $\alpha$-TC or IPC reduces the level of mitochondrial damage associated with oxidative stress caused by hepatic I/R, but $\alpha$- TC combined with IPC offers no significant additional protection.

Molecular Strategy for Development of Value-Added Sesame Variety

  • There are two groups of significant functional constituents in sesame seeds on the whole; one is the vegetable oils and another is the anti-oxidative compounds. However, although high amounts of major fatty acids are synthesized in sesame seeds, their composition is unfavorable because the contents of alpha- and gamma-linolenic acid, the essential fatty acids, are very low or do not produced in sesame seeds. So, to increase these fatty acids in sesame seeds, one strategy is to overexpress their genes, ${\omega}$-3 fatty acid desaturase for alpha-linolenic acid and delta-6 fatty acid desaturase for gamma-linolenid acid, in them. Another molecular target is to enhance alpha-tocopherol, vitamin E, because its content is very low in sesame seeds. The enzyme, gamma-tocopherol methyltransferase, catalyzes the conversion of gamma-tocophero to alpha-tocopherol. Overexpression of this enzyme in sesame seeds could be also a good molecular breeding target. Reduction of phytic acid is also another molecular target in sesame seeds because phosphorus pollution may be caused by its high content in sesame seeds. Accordingly, to do so, one of target enzymes could be myo-inositol 1-phosphate synthase which is a key regulatory enzyme in the pathway of phytic aicd biosyntheses. In this lecture, a molecular strategy for development of value-added sesame crop is described in association with some results of our experiments involved in the molecular characterizations of the genes mentioned above.

Antioxidant Activity and Contents of Bioactive Components in Polar Microalgae (극지미세조류의 유용성분 함량 및 항산화 활성)

  • In this study, bioactive components such as polyohenols, flavonoids and tocopherols were determined in cultured polar microalgae (Fragilariopsis pseudonana, Chaetoceros neogracile, Stellarima microtrias, Porosiara pseudodenticular). Antioxidant activity of methanol extracts of polar microalgae was also investigated. ${\alpha}-Tocopherol$ contents in Fragilariopsis pseudonana were almost two times higher than those of Chaetoceros neogracile in. The antioxidant activity of methanol extracts of Fragilariopsis pseudonana methanol extracts determined by ABTS assay was higher than other algae. Total polyphenol contents of methanol extracts also showed a similar trend as antioxidant activity. The protective activity against oxidative damages induced by glutamate in PC 12 cells was shown in only Chaetoceros neogracile.

Effect of Lipoxygenase on the Oxidation of Rainbow Trout Lipid in Model system (모델시스템에 있어서 무지개 송어 지방질의 산화에 대한 Lipoxygenase의 영향)

  • The effect of lipoxygenase (LOX) on the oxidation and co-oxidation of lipid fraction was studied in the model system of rainbow trout. For the reaction in model system 1 g of lipid fraction and 50mL of enzyme extract(LOX, 140 unit in 50mL phosphate buffer solution at pH 7,4)), which were obtained from rainbow trout, were homoginized in the presence of Tween 20 and kept at 23$\circ$C for 3 days. The activity of LOX was decreased to 43% of initial level during the reaction in the model system. The initial composition of rainbow trout lipid was showed to be consisted of trigliceride(TG;82%) and free fatty acid(FFA;0.1%), while this converted to 59% of TG and 20% of FIFA, respectively after reaction in model system. Change of fatty acid composition was also observed and the content of linoleic acid, one of the major fatte acids, was decreased to 13% from 54% in the content of total fatty acids after reaction. The carotenoids in rainbow trout were composed of 0.4% $\alpha$-carotene, 1.6% $\beta$ -carotene, 80% canthaxanthin, 7% lutein and 11% zeaxanthin, thus the canthaxanthin was the major component. This canthaxanthin was the most degraded carotenoid by lipoxygenase catalyzed co-oxidation during the reaction. On the other hand the tocopherol isomers found in the rainbow trout were $\alpha$ and $\beta$ -tocopherol, and $\alpha$-tocopherol had a higher degradation rate by the lipoxygenase catalyzed co-oxidation than of $\beta$-tocopherol in the reaction of model system.

Antioxidant and Synergistic Effect of Sesame Oil Cake Extract Treated from $\beta$-Glucosidase ($\beta$-Glucosidase 처리된 참깨박 추출물의 항산화 및 상승효과)

  • The antioxidant and synergistic effects of sesame oil cake extract treated with $\beta$ -glucosidase were examined. The sesamin and sesamolin were identified from the 80% ethanol extract of seame oil cake treated with $\beta$ -glucosidase, which suggested the presence of the active substances as their glycosides in sesame seed. The contents of sesamin and sesamolin in sesame oil cake extract were about 8.32% (8,315.4 mg/100g) and 0.28% (2,824.5mg/100g) , respectively. Sesame oil cake extract showed antioxidant activity at concentrations of 50ppm, 100ppm and 200ppm, and the effect was Increased with the addition of sesame oil cake extract. The antioxidant effect of sesame oil cake extract was stronger than that of $\alpha$-tocopherol or ascorbyl palmitate, but weaker than of BHT Also, when the sesame oil cake extract(50ppm) was used in combination with $\alpha$-tocopherol(50 ppm), the sesame oil cake showed very strong synergistic effect.

Effects of Dietary Conjugated Linoleic Acid (CLA) on Antioxidant System in the Liver of Chronically Ethanol-Treated Rats (식이에 첨가한 Conjugated Linoleic Acid (CLA)가 만성적으로 알코올을 섭취한 쥐에서 간조직의 항산화 체계에 미치는 영향)

  • The study was designed to observe antioxidant activities of conjugated linoleic acid (CLA) by determining antioxidant enzyme protein levels [cytochrome P4502 El (CYP2E1), Copper, Zinc-superoxide dismutase (CuZn-SOD), glutathione peroxidase (CSH-Px), glutathione S-transferase (GST)] by Western blot analysis and the levels of ${\alpha}$-tocopherol and 2-thiobarbituric acid reactive substances (TBARS) in the liver of chronically ethanol-treated rats. Sixty Sprague Dawley male rats were divided into 3 groups (Control, EtOH, EtOH+CLA). All rats were fed Lieber-DeCarli liquid diet for 4 weeks by pair-feeding against the EtOH group. The liquid diet was supplemented with 1.77g CLA mixture per kg diet in the EtOH+CLA group. Isocaloric maltose dextrin was added in replace of 50g ethanol (36%kcal) for the Control group. Ethanol ingestion significantly increased the levels of CYP2E1 protein and TBARS, but significantly reduced CuZn-SOD protein level and increased GST protein level. There was no significant effect on the level of GSH-Px protein and ${\alpha}$-tocopherol in the liver by ethanol. CLA supplementation with ethanol significantly increased the levels of CuZn-SOD, GSH-Px and GST and also significantly attenuated TBARS level, whereas there was no significant effect on the levels of CYP2E1 protein and ${\alpha}$-tocopherol by CLA. Overall, the CLA supplemented to ethanol could significantly increase the levels of CuZn-SOD, GSH-Px and GST proteins and reduce the level of TBARS in the liver of chronically ethanol-treated rats.
