밀배아 지방질의 산화 안정성과 카로티노이드 및 토코페롤의 변화 (Oxidative Stability of Wheat germ Lipid and Changes in the Concentration of Carotenoid and Tocopherol during Oxidation)

  • 밀배아를 저장온도 $30^{\circ}C$에서 산화반응을 시키면서 밀배아 지방질의 구성성분과 지방산의 함량변화를 관찰하였고, 이에 따른 카로티노이드와 토코페롤의 산화양상을 살펴보았다. 밀배아 지방질 구성성분의 변화를 보면 초기에는 triglyceride 함량이 66%, 유리지방산 함량이 7%였는데 30일 저장 후에는 각각 49%, 24%로 변하였다. 유리지방산의 조성은 초기에 lauric acid(29%), palmitic acid(21%), linoleic acid(20%)이던 것이 30일 후에는 linoleic acid가 30%로 크게 증가하였다. 밀배아 중에 있는 카로티노이드로는 베타-, 알파-카로틴, lutein, taraxanthin 등이 있었으며 이들의 함량은 저장 전에는 밀배아 1g당 각각 306, 59, 383, 356ng이던 것이 20일 저장 후에는 36, 4, 203, 149 ng으로 현저하게 감소하였는데 특히 베타-카로틴의 산화가 심하여 22.5ng/day의 최대 산화속도를 나타냈다. 밀배아 중의 토코페롤 동족체로는 알파-, 베타-, 감마-토코페롤 등이 있었는데 이들의 함량은 밀배아 1g당 각각 $55,\;48,\;38\;{\mu}g$이었고, 특히 가장 산화가 심하였던 알파-토코페롤의 최대 산화속도는 $1.26\;{\mu}g/day$를 나타내었다.

천연색소 소재의 항산화 및 항비만 활성 (Antioxidant and Antiobesity Activity of Natural Color Resources)

  • 천연색소로 사용되는 식물류 11종(쪽, 아카시아, 코치닐, 아선약, 포도과피, 태수, 헤나, 국화, 붉은백단향, 대황, 서양꼭두서니)의 생리활성 규명을 위해 열수 추출 분말을 제조하여 항산화 및 항비만 활성을 비교 분석하였다. 총 페놀 함량은 아선약 추출물이 348.23 mg/g으로 가장 높았다. DPPH, ABTS 라디칼 소거능 및 FRAP법에 의한 환원력도 아선약 추출물의 활성이 가장 높았다. ${\beta}$-carotene linoleic acid system을 이용한 항산화 활성은 아카시아 추출물이 가장 높았다. 소화효소 저해 효과를 측정한 결과, ${\alpha}$-glucosidase 저해 활성은 아카시아 추출물이 93.93%로 가장 높았고, ${\alpha}$-amylase 저해 활성은 아선약 추출물이 64.53%로 가장 높았다. Trypsin 저해 활성은 아카시아 추출물이 60% 이상으로 가장 높았고, ${\alpha}$-chymotrypsin 저해 활성은 아선약 추출물이 40% 이상으로 가장 높았다. ipase 저해 활성은 아카시아 추출물이 52.73%로 가장 높았다. 3T3-L1 세포에 대한 지방 축적률은 아선약 추출물이 55.8%로 가장 낮아 지방세포 분화 억제 효과가 큰 것으로 나타났다. 결론적으로 식물류 색소원료의 중 아선약 추출물과 아카시아 추출물은 높은 항산화 활성을 나타내었으며, 탄수화물, 단백질 및 지방 분해효소 저해에도 상당한 효과를 가지는 것으로 나타났는데, 이러한 결과는 페놀 함량과 상관관계가 높은 것으로 판단된다. 특히 아선약 추출물의 경우 지방세포 분화 억제를 통해 항비만 활성을 나타내는 것으로 판단이 되며 그와 관련된 메커니즘 연구는 좀 더 진행되어야 할 것으로 사료된다.

비타민 A 섭취가 에탄올을 급여한 흰쥐의 체내 항산화 영양소 상태에 미치는 영향 (Effect of Dietary Vitamin A on the Status of Antioxidants in Ethanol-Treated Rats)

  • 본 연구는 만성적인 에탄올 급여에 의한 조직내 항산화 영양소 함량의 변화에 대하여 비타민 A의 형태별 급여효과를 조사하고자 실시되었다. 실험동물은 Sprague-Dawley종 숫컷 흰쥐를 사용하였으며 에탄올 공급과 동시에 비타민 A 결핍군(FE), ${\beta}-carotene$ 섭취군(${\beta}E$), retinyl acetate 섭취군(RE) 그리고 13-cis-retinoic acid 성취군(RAE)과 에탄올 대신 동일 열량을 sucrose으로 공급 시킨 각각의 pair-fed군 등 모두 8군으로 나누어 7주 동안실섬 식이를 공급하였다. 항산화 영양소로 비타민 E, C, glutathione 그리고 무기질 함량의 분석을 통해서 에탄올에 의해 변화되는 생체내 항산화 영양소 상태와 이에 비타민 A의 형태별 급여 효과를 조사하여 다음과 같은 결과를 얻었다. 혈장내 ${\alpha}-tocopherol$ 함량은 에탄올­ 급여로 감소되었으나 간 조직내에서는 ${\beta}-carotene$ 섭취군만이 낮은 함량을 보였다. 그러나 간 조직내 tocopheryl acetate 함량은 pair-fed군에 비해서 에탄올 급여로 현저히 저하되었다. 혈장과 간 조직내에서 에탄올 급여에 의한 비타민 E 함량 감소는 retinyl acetate 공급으로 그 감소 정도가 가장 저하되었다. 혈장내 총 비타민 C 함량은 에탄올 급여와 동시에 ${\beta}-carotene$이나 retinyl acetate 섭 취군이 pair-fed군에 비해서 낮았으며 환원형과 산화형 비타민 C 함량비는 에탄올 급여로 낮았으나 ${\beta}-carotene$ 섭취시킨 군만이 높게 나타났다. 간 조직내 총 비타민 C 함량은 13-cis-retinoic acid 섭취군을 제외한 전 군에서 에탄올 급여로 증가하였으여 환원형과 산화형 비타민 C 비는 에탄올 급여군 중 ${\beta}-carotene$을 섭취시켰을 때 가장 높았다. 간 조직내 총 glutathion 함량은 에탄올 공급군과 pair-fed군 사이에 유의적인 차이를 보이지 않았으나 환원형과 산화형의 함량비에서는 에탄올과동시에 retinyl acetate나 13-cis-retinoic acid 섭취군에서 pair-fed군에 비해 높았다. 혈장내 Zn, Cu, Se, Fe 함량은 에탄올 급여군과 pair-fed군 사이에 일정한 경향을 보이지 않았으나 간 조직내 Zn, Cu 함량은 에탄올 급여로 감소된 반면 Fe 함량은 증가되었다. 이러한 결과들로 보아 에탄올은 체내 항산화 영양소 상태의 변화를 초래하며 비타민 A의 공급여부 뿐만 아니라 형태별 섭취에 의해서 항산화 영양소 상태가 변화될 수 있는 것으로 나타났다. 그러나 생체내 항산화 영양소들은 서로 상쇄, 상승 혹은 길항작용을 통해서 상호영향을 미치므로 각각의 항산화 영양소 상태 변화에 대한 정확한 기전 규명을 위해서는 다각적인 연구가 요구된다고 볼 수 있다. 그러나 지질과산화물의 함량과 체내 항산화 영양소 상태는 상호관련성이 있었으며 본 실험에서 에탄올 공급에 의한 항산화 영양소 함량의 감소는 대체로 비타민 A를 retinyl acetate 형태로 섭취시켰을 때 다소 경감되는 것으로 나타났다.

Vitamin A status of 20- to 59-year-old adults living in Seoul and the metropolitan area, Korea

  • Dietary intakes and plasma concentrations of retinol and carotenoids were estimated in assessing the vitamin A status of Korean adults living in Seoul and the metropolitan area. Three consecutive 24-h food recalls were collected from 106 healthy subjects (33 males and 73 females) aged 20-59 years. Fasting blood samples of the subjects were obtained and plasma retinol and carotenoids were analyzed. The daily vitamin A intakes ($mean{\pm}SD$) were $887.77{\pm}401.35{\mu}g $ retinol equivalents or $531.84{\pm}226.42{\mu}g$ retinol activity equivalents. There were no significant differences in vitamin A intakes among age groups. The retinol intake of subjects was $175.92{\pm}129.87{\mu}g/day$. The retinol intake of the subjects in their 50's was significantly lower than those in their 20's and 30's (P<0.05). Provitamin A carotenoid intakes were $3,828.37{\pm}2,196.29{\mu}g/day$ ${\beta}$-carotene, $472.57{\pm}316.68{\mu}g/day$ ${\alpha}$-carotene, and $412.83{\pm}306.46{\mu}g/day$ ${\beta}$-cryptoxanthin. Approximately 17% of the subjects consumed vitamin A less than the Korean Estimated Average Requirements for vitamin A. The plasma retinol concentration was $1.22{\pm}0.34{\mu}mol/L$. There was no significant difference in plasma retinol concentrations among age groups. However, the concentrations of ${\beta}$-carotene, lycopene, and lutein of subjects in their 50's were significantly higher than those of in their 20's. Only one subject had a plasma retinol concentration < $0.70{\mu}mol/L$ indicating marginal vitamin A status. Plasma retinol concentration in 30% of the subjects was 0.70- < $1.05{\mu}mol/L$, which is interpreted as the concentration possibly responsive to greater intake of vitamin A. In conclusion, dietary intakes and status of vitamin A were generally adequate in Korean adults examined in this study.

감귤 과피 Carotenoid 색소의 분리 및 이화학적 성질 (Isolation and Physicochemical Properties of Carotenoid Pigments from Orange Peels)

  • Carotenoids isolated from orange peels were determined physicocohemical properties with TLC, UV-spectrophotometer and HPLC etc., and the results were as follows . Maximum absorption wavelength of the isolated carotenoids was 415nm when the result was similar to $\beta$-carotene as 423nm. Eight spots were obtianed from TLC, and identified lutein, lycopene, $\alpha$-carotene and $\beta$-carotene with HPLC. The effect pH during the storage period of isolate carotenoids, the period when the amount of pigment retention was over 50% in pH 5, 6 and 7 , after 10 days . The amount of pigment retention was lower in control than in treatment of sugars such as fructose, glucose and sorbitol , but sucrose was similar to the control, Isolated carotenoids were stable to ascorbic acid, and the amount of pigment retention was over 70% after 10 days. The amount of pigment retention in the effect oforganic acid was higher in treated citric acid , lactic acid and tartaric acid than in control, but lower in treated maleic acid and succinic acid. Isolated carotenoids were stable at 50 $^{\circ}C$, and the amount of pigment retention was over 50% at 10$0^{\circ}C$.

Beta-carotene prevents the spermatogenic disorders induced by exogenous scrotal hyperthermia through modulations of oxidative stress, apoptosis, and androgen biosynthesis in mice

  • We investigated whether ${\beta}$-carotene (${\beta}-CA$) or ellagic acid (EA), originating from various fruits and vegetables, has a preventive effect against male infertility induced by exogenous scrotal hyperthermia. ICR adult mice were intraperitoneally treated with 10 mg/kg of ${\beta}-CA$ or EA daily for 13 days consecutively. During this time, mice were subjected to transient scrotal heat stress in a water bath at $43^{\circ}C$ for 20 min on day 7, and their testes and blood were obtained on day 14 for histopathologic and biochemical analyses. Heat stress induced significant testicular weight reduction, germ cell loss and degeneration, as well as abnormal localization of phospholipid hydroperoxide glutathione peroxidase (PHGPx) and manganese superoxide dismutase (MnSOD) in spermatogenic and Leydig cells. Heat stress also altered the levels of oxidative stress (lipid peroxidation, SOD activity, and PHGPx, MnSOD, and $HIF-1{\alpha}$ mRNAs), apoptosis (Bax, Bcl-xL, caspase 3, $NF-{\kappa}B$, and $TGF-{\beta}1$ mRNAs), and androgen biosynthesis (serological testosterone concentration and $3{\beta}$-hydroxysteroid dehydrogenase mRNA) in testes. These changes were all improved significantly by ${\beta}-CA$ treatment, but only slightly improved by EA treatment. These findings indicate that ${\beta}-CA$, through modulations of oxidative stress, apoptosis, and androgen biosynthesis, is a potent preventive agent against testicular injuries induced by scrotal hyperthermia.

Evaluation of Antioxidant Status and Correlation among Antioxidant Indices in Female College Students

  • This study was done to evaluate the antioxidant status of female college students by determining their intakes and plasma levels of antioxidnt vitamins (vitamin C, A and E) and total antioxidant status (TAS). Subjects were 46 healthy female college students aged 20 - 29 years. Body composition was determined by a multifrequency bioelectrical impedance analysis. Dietary intakes were examined by 24hr record method and nutrients intakes were analyzed by the Computer Aided Nutritional analysis program for professional (CAN-pro). Plasma vitamin C level were measured by spectrophotometric method and retinol, ${\beta}$-carotene, ${\alpha}$-tocopherol were measured by HPLC. Plasma TAS was measured with a Randox kit using the trolox equivalent antioxidant capacity (TEAC) method. Daily energy and protein intakes of the female college students were 1670.5㎉ (83% of RDA) and 63.3g (115.1% of RDA), respectively. However their intakes of Ca and Fe were below 75% of RDA. Their intakes of vitamin A and C were 596.6 ${\mu}$ gRE (85.2% of RDA) and 71.0mg (101.4% of RDA), respectively. Plasma levels of vitamin C, retinol, ${\beta}$-carotene and ${\alpha}$-tocopherol were 14.7mg/L, 0.7mg/L, 0.2mg/L and 9.1mg/L, respectively which were within normal range. There was no subject with deficiency or marginal level in plasma vitamin A and C. However 1.6% of the subjects had below adequate level in vitamin E. Plasma TAS level was 1.2mmol/L. Correlation data showed that all plasma antioxidant vitamins were positively correlated with plasma TAS. Overall data indicate that the antioxidant status of female college students were pretty good. However it might be necessary to educate them to eat more fruits and vegetables for preventing many chronic diseases in a later life. (J Community Nutrition 5(1) : 13∼20, 2003)

The Effects of Bioactive Compounds and Fatty Acid Compositions on the Oxidative Stability of Extra Virgin Olive Oil Varieties

  • The aim of this study was to determine the various bioactive components of five olive oil varieties, as well as to assess their contribution to the oxidative stability of the oils. Fatty acids, ${\alpha}$-tocopherol, ${\beta}$-carotene, total flavonoids, total phenols, and certain phenolic compounds of extra virgin olive oils (EVOO; blended, arbequina, hojiblanca, and picual) and pure olive oil (POO) were examined. Oxidation stability was evaluated by the peroxide value (POV). The total content of all the studied antioxidant compounds was significantly higher in the EVOOs than the POO (p<0.05). Among the EVOOs, picual had the highest levels of ${\alpha}$-tocopherol ($10.18{\pm}0.40\;mg/100\;g$), ${\beta}$-carotene ($557{\pm}8\;{\mu}g/100\;g$), and total phenols ($110.7{\pm}1.3\;mg/g$), which correlated strongly with antioxidative capacity. Furthermore, the lowest POV occurred in picual EVOO and correlated with the highest monounsaturated fatty acid (MUFA, C16:1 and C18:1) and lowest polyunsaturated fatty acid (PUFA, C18:2 and C18:3) compositions, suggesting the ratio of MUFA to PUFA is a critical parameter for the oxidative stability of olive oil. Our results indicate that the oxidative stability and antioxidant potential of EVOO depends not only on the antioxidant vitamins, but also on the amount of phenolic compounds and fatty acid profile of the oil.

표고버섯의 아미노산(酸) 및 비타민분석(分析) (Analysis of Amino Acid and Vitamin in Oak Mushroom (Lentinus edodes Sing))

  • 추용(椎茸)버섯의 영양학적(營養學的) 기초자료(基礎資料)의 보강(補强) 및 미분석자료(未分析資料)를 제공(提供)하기 위하여 일반분석(一般分析), amino 산(酸) 분석(分析), vitamin 분석(分析)을 행한 결과(結果)를 요약(要約)하면 다음과 같다. 1) 일반분석(一般分析)은 이미 발표(發表)된 영양분석표(榮養分析表)의 기본(基本) 분석치(分析値)와 유사(類似)하였으며, 2) Amino 산(酸)은 모두 17종 즉, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, valine, methionine, isoleucine, lecucine, tyrosine, phenylalanine, tryptophan, lysine, bistidine, arginine 등이 검색(檢索)되었는데, 가장 많이 함유(含有)된 성분(成分)은 glutamic 산(酸)이었다. 3) 필수(必須) 아미노산 8종(種) 즉, threonine, valine, methionine, isoleucine, lecucine, phenylalanine, tryptophan, lysine 등이 모두 함유(含有)되어 있어서 매우 중요(重要)한 식품(食品)이라는 것을 알수 있었다. 4) Vitamin은 총(總) 7종(種) 즉, A(${\beta}$-carotene), $B_1$ (thiamin), $B_2$(riboflavin), C (ascorbic acid), E (${\alpha}$-tocopherol), Niacin, Cholinechloride이 분석(分析)되었으며 가장 많은 양(量)은 Cholinechloride 이었다.

Microplate-Based Oxygen Radical Absorbance Capacity (ORAC) Assay of Hydrophilic and Lipophilic Compartments in Plasma

  • Methods have been developed to evaluate the total antioxidant capacity of foods and plasma but limitations are associated with their ability to determine precisely the contribution of lipophilic antioxidants in a lipid milieu as well as interactions among them Thus, we modified the Oxygen Radical Absorbance Capacity (ORAC) assay to determine the peroxyradical scavenging ability of both hydrophilic and lipophilic compartments in plasma The hydrophilic ORAC assay was performed in a phosphate buffer system utilizing 2,2'-azobis (2-amidinopropane) dihydrochloride as a peroxyradical generator and fluorescein as the target The lipophilic ORAC assay was carried out in a dimethylsulfoxide :butyronitrile (DMSO/BN, 9:1 v/v) system using 2,2'-azobis (2,4-dimethyl valeronitrile) as a peroxyradical generator and BODIPY C11 581/591 as the target Analyses were conducted in bovine serum supplemented with water - and lipid - soluble antioxidants and in human plasma. Albumin (0.5$\sim$5 g/dL) and uric acid (0.1$\sim$0.5 $\mu$mol/L) increased hydrophilic ORAC values in a dose-dependent fashion ($R^{2}$=0.97 and 0.98, respectively) but had no impact on lipophilic ORAC values. $\alpha$-Tocopherol (15$\sim$200 $\mu$mol/L) increased lipophilic ORAC values in a dose-dependent fashion ($R^{2}$=0.94); neither $\alpha$-tocopherol nor $\beta$-carotene had an impact on hydrophilic ORAC values. However, addition of $\beta$-carotene at physiological concentration (0.23$\sim$1.86 $\mu$mol/L), either alone or in combination with other carotenoids, had no significant impact on lipophilic ORAC values. Thus, while assays of 'total antioxidant capacity' in biological matrices would be a useful research and clinical tool, existing methods are limited by the lack of complete responsiveness to the full range of dietary antioxidants.
