In order to broaden the spectrum of substrate utilization of a Gram negative bacterium Zymomonas mobilis which has a great potential as an industrial ethanol producing microorganism, cloning of $\alpha$-amylase gene into Z. mobilis ZM4 was tried. The $\alpha$-amylase gene was isolated from Bacillus stearothermophilus. By Southern blot analysis, it was proven that the $\alpha$-amylase gene fragment was originated from a naturally occuring plasmid of B. stearothermophilus ATCC 31195. To place $\alpha$-amylase gene under the control of Z. mobilis promoter, two different Z. mobilis expression vectors, pZA26 and pLOI204, were used. The truncated $\alpha$-amylase gene was then introduced into these vectors. Both qualitative and quantitative activities of $\alpha$-amylase were observed in Z. mobilis cells harboring these plasmids with the $\alpha$-amylase gene inserted. Gas chromatographic analysis of ethanol showed that one of the Z. mobilis transconjugants was capable of producing 67 mM ethanol from rich medium(RM) containing 5% soluble starch as a sole carbon source.

E. coli-S. cerevisiae shuttle vector인 plasmid YRp7을 이용하여 B. amyloliquefaciens의 $\alpha$-amylase gene을 E. coli 내에 cloning하였다. 이때 제한 효소 Sau 3 AI에 의해 얻어진 $\alpha$-amylase gene의 크기는 약 1.95kb정도였으며 E. coli내에서 비교적 안정하게 유지되고 발현되었다. 재조합 plasmid p-EA24를 함유한 E. coli는 B. amyloliquefaciens의 약 65% 정도의 $\alpha$-amylase를 생성하였으며, 최적온도, pH, CaCl$_2$의 영향등 $\alpha$-amylase의 효소학적인 성질을 비교 조사해 본 결과 B. amyloliquefaciens의 $\alpha$-amylase와 동일하였다. 또한 E. coli에서 생성된 $\alpha$-amylase로 70% 정도가 periplasmic space에 존재하였으며 나머지는 세포 내부에 존재함을 알았다.

Schwanniomyces castellii의 제놈 DNA로 제조된 유전자 은행으로부터 cloning된 ${\alpha}$-amylase 유전자가 Sacchromyces cerevisiae에서 발현되었다. Cloning된 삽입 DNA 절편의 크기는 약 5.0 kb이었고, Southern 및 immunoblot 분석 결과 cloning된 ${\alpha}$-amylase 유전자가 Sch. Castellii로부터 유래되었음이 확인되었다. S. cerevisiae SHY3 형질전환체에서 Sch. Castellii ${\alpha}$-amylase 유전자발현은 모균주에 비해 낮았으나, 단백질의 분자량 및 효소의 성질은 Sch. Castellii에서 분리한 ${\alpha}$-amylase의 그것과 차이가 없었다.

The expression of the mouse $\alpha-amylase$ gene in the methylotrophic yeast, P pastoris was investigated. The mouse $\alpha-amylase$ gene was inserted into the multi-cloning site of a Pichi a expression vector, pPIC9, yielding a new expression vector pME624. The plasmid pME624 was digested with SalI or BglII, and was introduced into P. pastoris strain GSl15 by the PEG1000 method. Fifty-three transformants were obtained by the transplacement of pME624 digested with SaiII or BglII into the HIS4locus $(38\;of\;Mut^+\;clone)$ or into the AOX1 locus $(15\;of\;Mut^s\;clone)$. Southern blot was carried out in 11 transformants, which showed that the mouse $\alpha-amylase$ gene was integrated into the Pichia chromosome. When the second screening was performed in shaker culture, transformant G2 showed the highest $\alpha-amylase$ activity, 290 units/ml after 3-day culture, among 53 transformants. When this expression level of the mouse $\alpha-amylase$ gene is compared with that in recombinant Saccharomyces cerevisiae harboring a plasmid encoding the same mouse $\alpha-amylase$ gene, the specific enzyme activity is eight fold higher than that of the recombinant S. cerevisiae.

E. coli-Yeast shuttle vector YEp 13에 B. amyloliquefaciens의 $\alpha$-amylase gene을 cloning하여 얻은 hybrid plasnidlasmid를 E. coli를 숙주세포로 하여 형질을 발현시켰다. 형질전환주의 $\alpha$-amylase 활성 측정 결과, B. amyloliquefaciens에 대해 20-30%의 상대 활성도를 나타내었다. 형질전환주의 경우 생성된 $\alpha$-amylase의 60-65%가 periplasm에 축적되었으며 세포 외부로의 분비는 없었다. Hybrid DNA를 agarose-gel 전기영동으로 조사한 결과 그 크기가 다른 다수의 hybrid DNA가 확인되었으며 pHA28의 plasmid (a)(Fig.3)에서만 $\alpha$-amylase 활성을 나타내는 것으로 밝혀졌다 pHA28의 plasmid(a)의 크기가 YEp 13 plasmid DNA보다 작은 것은 YEp13 plasmid에서 yeast gene부분이 deletion된 결과로 추측되었다.

The relationship between Schwanniomyces occidentalis CBS 2863 (formerly castellii) and CBS 1153 (formerly alluvius), and their variety persoonii was examined at alpha-amylase gene level. Using Sch. occidentalis alpha-amylase gene as probe, Sch. occidentalis alpha-amylase gene homologues were obtained from Sch. occidentalis CBS 1153 and Sch. occidentalis var. persoonii. The restriction analysis of these homologues showed that the restriction enzyme sites between Sch. occidentalis CBS 2863 and CBS 1153 was identical but different between these strains and Sch. occidentalis var. persoonii.

YEp 13 plasmid에 B. amyloliquefaciens의 $\alpha$-amylase gene을 cloning시켜서 얻은 hybrid plasmid를 E. coli C 600으로 형질전환시켜서 amylase 활성을 나타내는 균주를 선별하였다. 선별된 E. coli C 600균주를 plasmid추출하여 전기영동해 본 결과 plasmid가 매우 불안정하였으며, 그중 가장 단순한 plasmid band를 지니고 있으며 amylase활성이 강한 E. coli균주를 선별하였다. 선별된 균주의 균체내에 있는 2개의 plasmid DNA를 분리하여 각각의 plasmid를 pTG 17-1, pTG 17-2로 명명하였으며 S. cerevisiae MC 16에서 형질전환이 가능한 pTG 17-2 DNA를 제한효소 EcoRI과 Pst I으로 restriction한 결과 EcoRI으로 처리한 경우는 7.3, 4.8, 2.4 kb인 3개의 분획으로 나타났으며 Pst I으로 처리한 경우는 linear로 14.5kb임을 알았으며 이로써 pTG 17-2 plasmid의 size가 약 14kb임을 알았다. 또한 E.coli균체내에서의 ampicillin sensitive로써 이 plasmid의 ampicillin resistance site가 결실되었음을 알았고 효모의 형진전환체로 부터의 $\alpha$-amylase는 균체외로 분비되지 않았고 효모균체내의 $\alpha$-amylase는 Somogyi-Nelson방법과 Agar diffusion 방법으로 확인하였다.

국내 사찰에서 제조된 된장으로부터 내열성 ${\alpha}$-amylase 생산균으로 분리된 YB-1234는 형태적 특성, 생화학적 성질 및 16S rRNA 유전자 염기서열에 근거하여 Bacillus licheniformis로 동정되었다. B. licheniformis YB-1234의 ${\alpha}$-amylase 유전자를 클로닝하여 그 염기서열을 결정하였으며 그로부터 유추된 ${\alpha}$-amylase의 아미노산 서열은 glycosyl hydrolase family 13에 속하는 B. licheniformis의 내열성 ${\alpha}$-amylases와 매우 높은 상동성을 보였다. ${\alpha}$-Aamylase 유전자를 함유한 재조합 대장균과 B. licheniformis에 의해 각각 생산된 ${\alpha}$-amylase는 pH 6.0에서 최대활성을 보였으나, 최적 반응온도는 약간의 차이가 있었다. 또한 B. licheniformis로부터 ${\alpha}$-amylase는 재조합 대장균에서 생산된 효소보다 열안정성이 매우 높았다. 이들 효소에 의한 maltotetraose와 maltohexaose의 주된 가수분해산물로는 glucose, maltose 및 maltotriose가 관찰되었다.

Plasmid pBR322와 runaway Plasmid pSY343을 vector로 사용하여 E. stearothermophilus IAM 11062내의 $\alpha$-amylase 유전자를 E. coli내에 클로닝 하였다. 이때 얻어진 $\alpha$-amylase유전자는 제한효소 Hind III의 말단을 갖고 있는 4.7kb의 크기였으며 E. coli내에서 이들 유전자는 비교적 안정적 있게 유지되고 발현되었다. 재조합 $\alpha$-amylase유전자가 클로닝된 E. coli는 B. stearothermophilus IAM 11062보다 3배의 $\alpha$-amylase를 더 많이 생성하였다. EDTA를 사용한 osmotic shock 방법에 의하여 E. coli내에서 생성된 $\alpha$-amylase는 그 효소 생성량의 75%정도가 periplasm에 존재함이 밝혀졌다. 재조합된 $\alpha$-amylase 유전자에 의해서 E. coli에서 생성된 $\alpha$-amylase는 최적 작용온도가 55$^{\circ}C$로서 이들의 열안정성과 분자량(61,000)도 B. stearothermophilus IAM 11062의 $\alpha$-amylase와 거의 동일하게 나타나 E. coli와 B. stearothermophilus IAM 11062에서 생성된 $\alpha$-amylase는 효소학적 성질이 같음을 보여주었다.

Bacillus amyloliquefaciens가 생성하는 액화형 $\alpha$-amylase의 구조유전자를 클로닝하기 위하여 이 균주 염색체의 2 - 5kb 획분을 분리하여 Mbo I으로 부분분해한 후 pUB110플라스미드의 BamHI 부위에 삽입하여 hybrid vector pSKS3 를 제작하였다. 이 재조합플라스미드를 세한수식결손 세포인 Bacillus subtilis ED 107에서 subcloning한 후 당화형 $\alpha$-amylase를 생성하는 Bacillus subtilis NA 64의 $\alpha$-amylase결실변이주 Bacills subtilis KM 107 주에서 형질 발현시켰다. 이때 형질전환빈도는 $10^{-5}$ - $10^{-7}$정도였으며 그중 약 50%가 $\alpha$-amylase 분비능이 있었으나 거의 대부분이 쉽게 실활되었다. 최종선별과정에서 $\alpha$-amylase를 가장 강하게 생성하는 형질전환주 Bacillus subtilis KM213으로부터 재 추출한 재조합플라스미드 pSKS3는 5.7kb 삽입된 단편이 BamH I/Mbo I 부위에 결합되어 있었다. pSKS3에 클로닝된 유전자에 의해 발현되는 $\alpha$-amylase 활성은 B. amyloliquefaciens 활성의 약 25%였으며, 이 pSKS3에 의해 발현되는 $\alpha$-amylase 단백은 B.amyloliquefaciens의 $\alpha$-amylase와 동일한 전기영동성을 나타내었음을 볼 때 pSKS3에 클로닝된 유전자는 B. amyloliquefaciens에서 유래함을 알 수 있었다.
