Changes in Seed Vigour of Sweet and Super Sweet Corn Hybrids as Affected by Storage Conditions (단옥수수와 초당옥수수의 저장조건에 따른 종자 활력변화)

  • An experiment was conducted to characterize the seed vigour of sweet (su) and super sweet (sh2) corn seeds stored at different temperatures and relative humidities (RH). Hybrid seeds of Early Sunglow ${\times}$ Golden Cross Bantam 70 (su) and Xtrasweet 82 ${\times}$ Fortune (sh2) were stored at different temperatures ($5\;and\;15^{\circ}C$) and RH(70 and 85%) for 10 months. Results of the experiment show that seed deterioration of super sweet corn was much faster than that of sweet corn under all storage conditions. Germination rate of sweet corn seeds at $25^{\circ}C$ and emergence rate in cold test showed similar patterns. Emergence rate of super sweet corn in cold test was significantly lower than the germination rate at $25^{\circ}C$. Germination rate of both sweet and super sweet corns was positively correlated to the emergence rate in cold test, but the correlation coefficient of super sweet corn was much lower compared to the sweet corn. This implies that the viability of super sweet corn seeds should be tested in the cold test to estimate field emergence rate. Seeds of sweet corn could be stored for 5 months under all storage conditions without significant seed deterioration, while those of super sweet corn should be stored at low temperature and RH. The emergence rate of sweet corn in cold test was not correlated to the leakage of total sugars, electrolytes or ${\alpha}-amylase$ activity, while that of super sweet com was positively correlated to the ${\alpha}-amylase$ activity, negatively correlated to the leakage of electrolytes, and was not correlated to the leakage of total sugars.

Inhibition of Listeria monocytogenes by Bacteriocin(s) from Lactic Acid Bacteria Isolated from Kimchi (김치에서 분리한 젖산균 bacteriocin에 의한 Listeria monocytogenes의 억제)

  • Four strains of lactic acid bacteria which produced bacteriocins inhibitory to Listeria species were isolated from Kimchi, and were identified as Leuconostoc mesenteroides subsp. mesenteroides (2 strains), Leuconostoc paramesenteroides and Pediococcus pentosaceus. The bacteriocins produced by the isolates inhibited all of the Listeria monocytogenes strains tested, but L. denigrificans 28 and L. welchimeri 89 were not inhibited by the bacteriocin produced by the Leu. paramesenteroides isolate. The bacteriocin produced by the P. pentosaceus isolate was more inhibitory against sensitive strains and showed broader spectrum of antimicrobial activity than those produced by other isolates. The bacteriocins produced by Leuconostoc isolates were sensitive to pronase E treatment, but that produced by the P. pentosaceus isolate was not completely inactivated. The bacteriocins produced by all of the isolates were not sensitive to catalase, ${\alpha}$-amylase and lysozyme and heat (30 min at $100^{\circ}C$) treatments.

Effect of Sodium Sulfate on Ramie Fabrics Treated with Pectinase (저마직물의 펙티나제 정련 시 황산나트륨의 영향)

  • This study examines the scouring effect of pectinase on ramie fabric and influence of sodium sulfate as an activator for pectinase. The scouring effects were measured by the weight loss and pectin contents. SEM, weight loss, stiffness, moisture regain and dye ability of ramie fabric teated with pectinase/sodium sulfate were also measured. When ramie fabrics were desized with $\alpha$-amylase, the optimum conditions were pH 6.5 at $60^{\circ}C$ for 80 min with 1%(o.w.f) $\alpha$-amylase concentration. When ramie fabrics were scoured with pectinase, the optimum conditions were pH 8.5 at $55^{\circ}C$ for 30 min with 10%(o.w.f) pectinase concentration. Addition of sodium sulfate improved enzyme activity significantly, which increased proportionally with increasing sodium sulfate concentration. When 50 g/l of sodium sulfate was added, the surface became cleaner compared to the enzyme treatment without salt: weight and tensile loss, moisture regain and dyeability of the treated fabrics increased, while pectin contents and stiffness decreased. Therfore, sodium sulfate was effective activator for the pectinase treatment of flax fiber.

Changes in Enzyme Activities of Rice with Respect to the Viviparity and the Duration of Lodging Time

  • The Japonica cultivar 'Janganbyeo' was cultivated and about half of paddy field was totally lodged at 30 days after heading. Both lodged paddy and unlodged paddy were harvested at intervals. The lodged paddy and brown rice did not show any signs of viviparity until the $4^{th}$ day of lodging. The activities of $\alpha$-amylase, diastase, and lipase of lodged paddy and brown rice began to increase on the $2^{nd}$ day of lodging and then increased slowly afterwards. The activities of protease and phytase of lodged paddy and brown rice reached the maximum values on the $2^{nd}$ day and then decreased until the final day of lodging. In contrast, unlodged paddy and brown rice showed relatively small increase in viviparity and in activities of $\alpha$-amylase, diastase, protease, lipase, and phytase. Especially, activities of diastase and lipase were observed even in unlodged paddy and brown rice.

Hypoglycemic and Angiotension Converting Enzyme Inhibitory Effect of Water and Ethanol Extracts from Haesongi Mushroom (Hypsizigus marmoreus)

  • Water and ethanol extracts were prepared from the haesongi mushroom (Hypsizigus marmoreus) to measure functional components. The ability of the extracts to inhibit angiotensin-converting enzyme (ACE) and their hypoglycemic effects were also determined; the latter was measured by $\alpha$-amylase and glucosidase inhibition. Extraction yield, protein content, total phenol, and $\beta$-glucan in the water extracts were 55.86, 17.71, 1.89, and 21.93%, respectively. The respective values for the ethanol extracts were lower than those for water extracts. Both water and ethanol extracts showed dosedependent ACE inhibition, the effect of the former being greater. The water extract inhibited ACE activity by 95.34% at 40 mg/mL. The $IC_{50}$ values of the water extracts were 63.32 and 0.41 mg/mL for $\alpha$-amylase and glucosidase, respectively. Thus, the water extracts had a greater hypoglycemic effect than the ethanol extracts. From these results, water is a better solvent than ethanol to extract from the haesongi mushroom functional components that show ACE inhibition and have hypoglycemic effects.

In vitro Screening of Dietary Factors on Buckwheat(Fagopyrum esculentum Moench) Influencing the Regulation of Blood Pressure, Glucose and Cholesterol Level (혈압, 혈당 및 콜레스테롤농도 조절에 영향을 주는 식이 메밀인자의 in vitro 검정)

  • Generally, buckwheat has been regarded as a crop of secondary importance in many countries. In vitro functionalities of buckwheats as a food were evaluated in this study. Five of buckwheat cultivars were extracted with methanol, and the extractant were dried and lyophilized, separately. Or water soluble buckwheat components were digested with the commercial enzymes and the obtained protein hydrolysate was again fractionated by acid precipitation. The antioxidant capacity of the methanol extracts determined using Fe2+-ascorbic acid system was dependent ont the cultivars: The extract of Suwon 4 showed 3.3 times stronger activity than ascorbic acid in terms of IC50. Also, the extracts of buckwheats inhibited efficiently the activities of $\alpha$-amylase and lens aldose reductase. Buckwheat soluble protein or rutin suppressed the in vitro activities of angiotensin-converting enzyme, and the inhibitory degree depended largely on the cultivars. Buckwheat proteins exerted higher hydrophobicity being related to the sterol binding capacity than casein. The results suggested that buckwheat seeds may be desirable and functional food resources in human living in current society.

Physicochemical and Microbiological Properties of Korean Traditional Meju (한국 재래식 메주의 이화학적 및 미생물학적 특성)

  • The objective of this study was to obtain basic data on Korean traditional meju collected in 17 regions of Korea, to define and control meju quality. The moisture, crude fat, crude protein, and amino nitrogen contents of meju were 9.83-36.24%(w/w), 17.46-28.74%(w/w), 42.00-45.54%(w/w), and 223.65-1137.68 mg%, respectively. Meju was the enzyme source which made the soy sauce and doenjang. The $\alpha$-amylase, $\beta$-amylase, and protease levels were 130.32-1254.45, 30.07-167.88 and 72.53-340.04 units, respectively. Regional enzyme activities differed widely. Bacterial levels were $4.8{\times}10^7-2.6{\times}10^{10}cfu/g$, and molds and yeasts were at $4.3{\times}10^4-7.9{\times}10^6cfu/g$.

Establishment of optimal soybean Koji manufacturing conditions (콩 코오지의 최적 제조조건 설정)

  • To establish the optimal manufacturing conditions of soybean koji, soybean Koji prepared with Aspergillus oryzae 6-M-1 and Bacillus subtilis 3-B-1 isolated from traditional Korean meju. During 7 days of making Koji, the amount of amino-type nitrogen was getting more increase. The amount of amino-type nitrogen of Koji prepared with A. oryzae 6-M-1 was 686.16 mg% (w/w), that of Koji with B. subtilis 3-B-1 was 643.46 mg% (w/w) at seventh day of making Koji. The ${\alpha}$-amylase activity of Koji prepared with A. oryzae 6-M-1 was 1472.54 unit/g, that of Koji with B. subtilis 3-B-1 was 791.00 units/g on the seventh day of the making. The acidic protease activity of Koji prepared with A. oryzae 6-M-1 was 309.00 unit/g, that of Koji with B. subtilis 3-B-1 was 135.88 unit/g at 7th day of making. The amount of amino-type nitrogen and enzyme activities of soybean Koji prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 were produced more than those of wheat flour Koji made in factory. Sensory evaluation on a commercial doenjang and doenjangs prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 was not significantly different at p<0.05.

Novel $\alpha$-Glucosidase from Extreme Thermophile Thermus caldophilus GK24

  • $\alpha$-Glucosidase of an extreme thermophile, Thermus caldophilus GK24 (TcaAG), was purified 80-fold from cells to a homogeneous state and characterized. The enzyme exhibited optimum activity at pH 6.5 and $90^{\circ}C$, and was stable from pH 6.0 to 85 and up to $90^{\circ}C$. The enzyme had a half-life of 85 minutes at $90^{\circ}C$. An analysis of the substrate specificity showed that the enzyme hydrolyzed the non-reducing terminal unit of $\alpha$-1,6-glucosidic linkages of isomaltosaccharides and panose, $\alpha$-1,3-glycosidic bond of nigerose and turanose, and $\alpha$-1,2-glycosidic bond of sucrose. The gene encoding the TcaAG was cloned, sequenced, and sequenced in E. coli. The nucleotide sequence of the gene encoded a 530 amino acid polypeptide and had a G+C content of 68.4% with a strong bias for G or C in the third position of the codons (93.6%). A sequence analysis revealed that TcaAG belonged to the $\alpha$-amylase family. We suggest that this monomeric, thermostable, and broad-acting $\alpha$-glucosidase is a departure from previously exhibited specificities. It is, therefore, a novel $\alpha$-glucosidase.

A Study on the Standardization of the Preparation Method for Waxy Rice Nochi (찹쌀 노치 제조법에 관한 연구)

  • Nochi is a kind of Korean traditional food made from glutinous rice or millet. This study attempted to examine the effects o( the various factors and to clarify the factors which affect the overall eating quality of Noch. The results were summarized as follows. 1. Starch content of waxy rice was 72.4% and IBC was 0.017%. 2. $\alpha$-amylase and $\beta$-amylase activity of malt were 40.13 and 8.94 units respectively. 3. Waxy rice soaked for 3 hours was ground to flour then sifted 20 mesh sieve. The mixture of waxy rice and malt was steamed for 20 minutes. 4. In sensory evaluation on varying the amount of malt and incubation time, the most favorite tendancy of the overall eating quality was at the level of 8hours incubation made by 5% addition of malt. 5. Total sugar contents, reducing sugar contents and the intensities of iodine stain at the incubation temperature of $60^{\circ}C$ were significantly different from those at $50^{\circ}C$.
