Aspergillus niger ATCC 16513과 대두(Glycine max. L) $\alpha$-galactosidase의 kinetic 성질 (Kinetic Properties of $\alpha$-Galactosidase from Aspergillus niger ATCC 16513 and Soybean(Glycine max. L))

  • Aspergillus niger ATCC 16513과 대두(Glycine max. L)의 정제 $\alpha$-galactosidase를 사용 하여 몇가지 이들의 kinetic성질을 조사 하였다. Asp. niger $\alpha$-galactosidase의 raffinose와 stachyose에 대한 Km값은 각각 37.0mM과 55.5mM, 대두 $\alpha$-galactosidase는 50.0mM과 55.5mM로서 PNPG보다 이들에 대한 친화성이 적었다. 또한 galactose는 ASP. niger와 대두 $\alpha$-galactosidase 모두의 활성을 저해 하였으나 2-mereaptoethanol과 L-cystene은 대두 $\alpha$-galatosidase의 활성만을 약간 저해 하였다. Asp. niger와 대두 $\alpha$-galactosidase의 활성에 관여하는 아미노산은 diethyl pyrocarbonate에 의한 화학수식에 의하여 histidine임이 확인 되었고 Asp. niger $\alpha$-galactosidase의 1mole당 아미노산 잔기수는 모두 902개, 대두$\alpha$-galactosidase는 286개 이었다.

  • 맨드라미의 물추출 물질의 지방산과 아미노산의 조성을 조사한 결과 지방산은 모두 14종이 검출되었으며 특히 $C_{16:2}$가 되어 있었으며 필수지방산도 모두 포함되어 있었다. 미지의 물질을 확인한 결과 tricosanoic acid, lignoceric acid, cerotic acid, 1, 1'-[3-(2-cyclopentylethylidene)-1, 5-pentanediyl] biscyclopentane의 methyl ester 등을 함유하고 있었다. 아미노산은 모두 16종이 검출되었고 산성아미노산의

  • The chiral amine auxiliary mediated stereoselective alkylation reactions of glycinamides 1-6 and 15-17 using phase transfer catalyst (PTC) for liquid-solid extraction are described. The secondary N-(diphenylmethylene) glycinamides 1, 2 and 3 give better selectivities and yields than tertiary N-(diphenylmethylene) glycinamides 4, 5 and 6. Alkylation of the glycinamide 1 and 2 using 18-Crown-6 as a PTC in toluene at $-40^{\circ}C$ gives best selectivities and yields. Alkylations of N-(4-chlorophenylmethylene)glycinamides 15, 16 and 17 under same PTC conditions give $\alpha$, $\alpha-disubstituted$ amino acid derivatives 18, 19 and 20 with low diastereoselectivities.

Temporal Changes in N Assimilation and Metabolite Composition of Nitrate-Affected Tomato Plants

  • The role of inorganic nitrogen assimilation in the production of amino acids, organic acids and soluble sugars is one of the most important biochemical processes in plants, and, in order to achieve normally, nitrate uptake and assimilation is essential. For this reason, the characterization of nitrate assimilation and metabolite composition from leaves, roots and xylem sap of tomato (Solanum lycopersicum) was investigated under different nitrate levels in media. Tomato plants were grown hydroponically in liquid culture under five different nitrate regimes: deficient (0.25 and 0.75 mM $NO_3{^-}$), normal (2.5 mM $NO_3{^-}$) and excessive (5.0 and 10.0 mM $NO_3{^-}$). All samples, leaves, roots and xylem sap, were collected after 7 and 14 days after treatment. The levels of amino acids, soluble sugars and organic acids were significantly decreased by N-deficiency whereas, interestingly, they remained higher in xylem sap as compared with N-normal and -surplus. The N-excessive condition did not exert any significant changes in metabolites composition, and thus their levels were similar with N-normal. The gene expression and enzyme activity of nitrate reductase (NR), nitrite reductase (NIR) and glutamine synthetase (GS) were greatly influenced by nitrate. The data presented here suggest that metabolites, as a signal messenger, existed in xylem sap seem to play a crucial role to acquire nitrate, and, in addition, an increase in ${\alpha}$-ketoglutarate pathway-derived amino acids under N-deficiency may help to better understand plant C/N metabolism.

Chemical Composition and Electron Donating and Nitrite Scavenging Activities of Glechoma hederacea var. longituba $N_{AKAI}$

  • This study was performed to investigate chemical and functional properties of Glechoma hederacea leaves in respect to its potential use as food material or as a medicinal herb. The chemical compositions on a dry harris were 20.38% in protein, 3.96% in fat, 59.58% in carbohydrate, 15.78% in ash, 5.36% in reducing sugar, 14.11% in total sugar and 0.26% in polyphenol, respectively. The free sugars were mainly comprised of glucose, fructose and sucrose. In fatty acids compositiosn, linolenic acid showed the highest concentration at 45%, while the ratios of saturated to unsaturated fatty acids were 1 : 1.91. Seventeen kinds of total amino acids were determined, with the highest concentration (2,465.71 mg%) of glutamic acid. Among the free amino acids, praline showed the highest concentration (260.09 mg%), followed by glutamine, $\alpha$ -amino adipic acid, glutamic acid and valine. The contents of major minerals were 647.32 mg% in Na, 597.53 mg% in K and 239.75 mg% in Ca. The antioxidative activity of 10% water extract was similar to that of 50 ppm tocopherol. The nitrite scavenging ability reached the highest bevel at pH 1.2 and the lowest at pH 6.0.

Separation of Protein and Fatty Acids from Tuna Viscera Using Supercritical Carbon Dioxide

  • Supercritical carbon dioxide extraction was investigated as a method for removing lipids and bad flavor from tuna viscera. To find the optimum conditions, different experimental variables, such as pressure, temperature, flow rate of solvent and sample size, were evaluated for the effective removal of lipids and the undesirable smell. Ethanol was used as the entrainer, with a $3\%$ by vol $CO_2$ flow rate. By increasing the pressure at constant temperature, the efficiency of the lipid removal was improved and the protein was concentrated without denaturalization. The main fatty acids extracted from the tuna viscera were palmitic acid (16:0), heptadecanoic acid (17:1), oleic acid (18:1) and docosahexaenoic acid (22:6). The major amino acids in the tuna viscera treated by supercritical carbon dioxide were glutamic acid, leucine and lysine, and the free amino acids were L-proline, taurine and L-$\alpha$-aminoadipic acid.

  • Pseudomonas syringae의 intact cell에서 에틸렌 생성을 극대화 하기 위한 전환조건은 $30^{\circ}C$. pH7.5로 조사되었고, 다양한 기질의 전환효과를 검초한 결과, Asn>Gln>Asp>Glu>$\alpha$-KG>citrate>oxalacetate의 순으로 많은양의 에틸렌을 생성하였다. 또한, arginine과 histidine을 상기 유기산과 함께 넣었을 때 에틸렌 생성에 현저한 상승효과를 나타냈다. Cell-free system 에서는 $\alpha$-KG>Glu>citrate>Gln>Ser순으로 0.5mM $\alpha$-KG에서 310.8(nl.mg $protein^[-1}.h^[-1}$)로 가장 많은 에틸렌을 생성하였고, aminotransferse 억제제인 AOA를 사용해 본 결과, Glu는 glutamate dehydrogenase에 의하여 $\alpha$-KG를 거쳐서 에틸렌으로 전환된 것이라 생각된다.
