Composition and Content of Soyasaponins and Their Interaction with Chemical Components in Different Seed-Size Soybeans

  • Soyasaponins $A_1$, DDMP-conjugated group B soyasaponins ${\alpha}g\;and\;{\beta}g$, non-DDMP counterpart soyasaponin I, II+III, and DDMP moiety were quantified in the large-, midium-, and small-seed soybean varieties. Protein contents were ranged from 38.1% to 41.8%, and oil contents were ranged from 15.5% to 18.9%, respectively. Oil contents in the large-seed varieties were significantly higher than those of medium- and small-seed varieties. Among detected soyasaponin peaks, ${\beta}g$ was a major soyasaponin in DDMP-conjugated group B soyasaponins followed by soyasaponin I, DDMP moiety and $A_1$. Soyasaponin concentration among different seed size soybean varieties. The soyasaponin concentration of mediumseed ($4014.5{\mu}g/g$) was slightly higher than those of largeseed ($3755.0{\mu}g/g$) and small-seed varieties ($3620.3{\mu}g/g$), however, the differences was statistically not significant. The composition rates of soyasaponins in the large-size seeds were 9.4% of soyasaponin $A_1$, 26.5% of DDMP-conjugated soyasaponins, 49.9% of non-DDMP counterpart soyasaponins, and 14.2% of DDMP moiety, respectively. Similar results were observed in the composition ratios of middle- and small-size seeds. Oil content and C:N ratio showed the significant positive correlations with total soyasaponin concentration, while the 100-seed weight, fiber, and ash contents showed the negative correlations with total soyasaponin but statistically not significant. It was noted that protein contents didn't have any relationship with group A, group B, DDMP moiety, and total soyasaponin. This fact suggested that protein contents are not affects the variation of soyasaponin concentration.

Saccharification and alcohol fermentation characteristics of a mixture of tapioca and hulled barley (타피오카와 겉보리 혼합원료의 당화 및 알코올 발효의 특성 연구)

  • An enzymatically saccharified tapioca and hulled barley (TB) raw mixed solution was used to examine alcohol fermentation characteristics. The TB mixture was liquefied with 0.04% ${\alpha}-amylase$ "Spezyme-Fred" and saccharified using an enzyme mixture (GPB), which consisted of glucoamylase (G), protease (P), and ${\beta}-glucanase$ (B). After the TB mixture (7:3, w/w) saccharified for 150 min at $50^{\circ}C$, its glucose content was 12.9% and viscosity was 26 cp. The use of GPB for the saccharification of TB was appropriate because the addition of ${\beta}-glucanase$ increases the glucose yield and decreases the viscosity of the saccharification liquid. The TB ratio was optimized to 7:3 (w/w) on the basis of the lower viscosity and the higher glucose content after saccharification. After TB mixture with 300% (w/w) water content was better condition than others for alcohol fermentation when it was carried out at $30^{\circ}C$. The alcohol and glucose contents of the TB mixture fermented for 72 h were 9.0 and 0.02%, respectively, and the pH and total acidity were 4.3 and 0.3%, respectively.

Study on the Copper-Arsenic Green Pigments used on Shamanic Paintings in the 19~20th century (19~20세기 무신도 등에 사용된 구리-비소 녹색 안료에 대한 연구)

  • The green pigments of shamanic paintings (83 items) in the 19~20th century were analyzed with X-ray fluorescent spectrometer(XRF), scanning electron microscopy (SEM) and fourier transform infrared spectroscopy (ATR-FTIR). In 60 items, copper and arsenic were detected in green pigments of the paintings by XRF spectra. Cu $K{\alpha}$/As $K{\beta}$ (peak intensity ratio of copper and arsenic) of shamanic paintings of Bokgaedang (shrine), solemn paintings (romance of three kingdoms) of Donggwanwangmyo and unknown enshrined place were 5.93~12.04 and higher compared to 5.67-6.26 of standard emerald greens and 4.01~7.89 of remaining shamanic paintings. The SEM images of crystal forms of copper-arsenic green pigments were various. Crystal forms were divided into oval and round spherulite with intersecting plate crystals and spherulite with agglomerate plate crystals. The crystals of the latter were found in shamanic paintings of Bokgaedang (shrine), solemn paintings (romance of three kingdoms) of Donggwanwangmyo and unknown enshrined place and the former were found in the rest of shamanic paintings. Copper-arsenic green pigments of shamanic paintings were identified as Scheele's green from shamanic paintings of Bokgaedang (shrine) and romance of three kingdoms. Emerald green from the rest of shamanic paintings by ATR-FTIR. From analytical results, it is confirmed that Scheele's green of shamanic paintings of Bokgaedang and romance of three kingdoms was used in the 1850s~1870s and emerald green had been widely used from late 19th century to 1970 in the rest of shamanic paintings.

Effects of Supplemental Levels of Bazhen on Growth Performances, Serum Traits, Immunity, Meat Quality and Antioxidant Activity of Taiwan Country Chickens

  • One hundred and sixty Taiwan country chickens (d-old chicks) were randomly assigned into four groups with four replicates and equal sex. Basal diets were supplemented with 0, 0.5, 1 and 2% of Bazhen powder, a traditional Chinese herbal medicine complex. The study was conducted for 14 wks. Experimental results indicated that Bazhen supplement did not influence feed intake, body weight gain and feed:gain ratio. Compared with control group, the percentage of serum HDL (high-density lipoprotein) linearly increased (p<0.03) and that of VLDL+LDL (very low-density+low-density lipoprotein) linearly decreased (p<0.03) in Bazhen supplemented groups, that 2% Bazhen was significantly different with control group (p<0.05). Chickens fed diets containing 2% Bazhen displayed reduced (p<0.05) serum GOT (glutamic-oxaloacetic transaminase) levels. The IgG, ${\gamma}$-globulin levels and PHA (phytohemagglutinin) skin challenge results in 1% Bazhan supplemented group were higher (p<0.05) than in the control group, the SRBC (sheep red blood cell) and ND (newcastle disease) titers in Bazhen supplemented groups were linear higher (p<0.05) than in the control group. The liver catalase activity and the capacity of scavenging DPPH (${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical were linearly increased (p<0.03) in Bazhen supplemented groups, and the 1 and 2% groups were different from the control group (p<0.05). Liver TBARS (thiobarbituric acid-reactive substances) levels in all Bazhen supplemented groups and total glutathione level in the 2% group were reduced (p<0.05) compared to the control group and displayed a linear response (p<0.05). The TBA (thiobarbituric acid) and pH value of the breast muscle after 24 h post-mortem in the Bazhen supplemented groups was linear lower (p<0.05) than in the control group. Results from this study demonstrated that Bazhen supplement in chicken had several beneficial effects, including increased SRBC and ND titers, HDL and IgG, ${\gamma}$-globulin levels, PHA skin challenge result, decreased VLDL+LDL and GOT levels, and displayed antioxidation effects in serum and carcass meat parameters.

Protective Effects of the Ethanol Extract of Viola tianshanica Maxim against Acute Lung Injury Induced by Lipopolysaccharides in Mice

  • Viola tianshanica Maxim, belonging to the Violaceae plant family, is traditionally used in Uighur medicine for treating pneumonia, headache, and fever. There is, however, a lack of basic understanding of its pharmacological activities. This study was designed to observe the effects of the ethanol extract (TSM) from Viola tianshanica Maxim on the inflammation response in acute lung injury (ALI) induced by LPS and the possible underlying mechanisms. We found that TSM (200 and 500 mg/kg) significantly decreased inflammatory cytokine production and the number of inflammatory cells, including macrophages and neutrophils, in bronchoalveolar lavage fluid. TSM also markedly inhibited the lung wet-to-dry ratio and alleviated pathological changes in lung tissues. In vitro, after TSM ($12.5-100{\mu}g/ml$) treatment to RAW 264.7 cells for 1 h, LPS ($1{\mu}g/ml$) was added and the cells were further incubated for 24 h. TSM dose-dependently inhibited the levels of proinflammatory cytokines, such as NO, $PGE_2$, $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$, and remarkably decreased the protein and mRNA expression of $TNF-{\alpha}$ and IL-6 in LPS-stimulated RAW 264.7 cells. TSM also suppressed protein expression of $p-I{\kappa}Ba$ and p-ERK1/2 and blocked nuclear translocation of $NF-{\kappa}B$ p65. The results indicate that TSM exerts anti-inflammatory effects related with inhibition on $NF-{\kappa}B$ and MAPK (p-ERK1/2) signaling pathways. In conclusion, our data demonstrate that TSM might be a potential agent for the treatment of ALI.

A Study on the Status of Antioxidant Nutrients and Lipid in the Middle-aged Korean Men Living in Taegu (대구지역 중년 남성의 혈중 항산화성 영양소와 지질상태에 관한 연구)

  • Serum levels of antioxidant vitamins and lipids were determined along with anthropometric measurements in 174 healthy male subjects with mean age of 50.3$\pm$6.8 years from Taegu area in Korea. Body mass index (BMI) and waist/hip(W/H) ratio of the subjects were 23.18$\pm$2.46 and 0.88$\pm$0.04, respectively and their systolic and distolic blood pressures were 127.8$\pm$15.5 and 83.9$\pm$10.8mmHg. Twenty one percent of the subject had BMI over 25. Average seum levels of total cholesterol, LDL- and HDL-cholesterol and triglyceride were 187.7$\pm$34.9, 117.6$\pm$33.5, 41.1$\pm$9.0 and 140.7$\pm$83.6mg/dl, respectively. Sixteen percent of the subject had LDL-chole-sterol over 130mg/dl. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subject was 2.01$\pm$0.77 MDA nmoles/ml and those of $\alpha$-tocopherol, retinol. ascorbic acid and sum of $\alpha$- and $\beta$-carotene were 9.59$\pm$3.11$\mu\textrm{g}$/ml, 1.15$\pm$ 0.38$\mu\textrm{g}$/ml, 10.5$\pm$3.8$\mu\textrm{g}$/ml and 64.6$\pm$43.$\mu\textrm{g}$/dl, respectively. About 14% of the sujects had low vitamin E status of less than 7.0$\mu\textrm{g}$/ml,. while 6% had low vitamin C status of less than 4.0$\mu\textrm{g}$/ml, Serum vitamin E showed positive correlations with total cholesterol and LDL-cholesterol and triglyceride, but no correlation with TBARS. Fatty acids of serum total lipid were composed of 42.9% as saturnted. 19.3% as monounsaturated and 36.7% polyunsaturated fatty acids(PUFA). N-6 and n-3 PUFA were 27.7% and 8.3% of total fatty acids. N-6/n-3 PUFA ratios were negatively correlated both with serum total cholesterol and TBARS.

A research on the emotion classification and precision improvement of EEG(Electroencephalogram) data using machine learning algorithm (기계학습 알고리즘에 기반한 뇌파 데이터의 감정분류 및 정확도 향상에 관한 연구)

  • In this study, experiments on the improvement of the emotion classification, analysis and accuracy of EEG data were proceeded, which applied DEAP (a Database for Emotion Analysis using Physiological signals) dataset. In the experiment, total 32 of EEG channel data measured from 32 of subjects were applied. In pre-processing step, 256Hz sampling tasks of the EEG data were conducted, each wave range of the frequency (Hz); Theta, Slow-alpha, Alpha, Beta and Gamma were then extracted by using Finite Impulse Response Filter. After the extracted data were classified through Time-frequency transform, the data were purified through Independent Component Analysis to delete artifacts. The purified data were converted into CSV file format in order to conduct experiments of Machine learning algorithm and Arousal-Valence plane was used in the criteria of the emotion classification. The emotions were categorized into three-sections; 'Positive', 'Negative' and 'Neutral' meaning the tranquil (neutral) emotional condition. Data of 'Neutral' condition were classified by using Cz(Central zero) channel configured as Reference channel. To enhance the accuracy ratio, the experiment was performed by applying the attributes selected by ASC(Attribute Selected Classifier). In "Arousal" sector, the accuracy of this study's experiments was higher at "32.48%" than Koelstra's results. And the result of ASC showed higher accuracy at "8.13%" compare to the Liu's results in "Valence". In the experiment of Random Forest Classifier adapting ASC to improve accuracy, the higher accuracy rate at "2.68%" was confirmed than Total mean as the criterion compare to the existing researches.

A Study on Anti-aning of Electroacupuncture at Joksamni($ST_{36}$) in Rats (족삼리(足三里) 전침(電鍼)이 흰쥐의 노화(老化)에 미치는 영향(影響))

  • Objectives : The purpose of this study is to investigate the effect of electroacupuncture at Joksanmi($ST_{36}$) on Rat's aging by experimental methods. Methods : The author performed several experimental procedures to observe the effects of the EC-HAS at the arthritis. First, I measured the cell survival rate of the mice lung fibroblasts. Second, the incidence rate of arthritis and arthritis index of CIA were analyzed. Third, the levels of IL-6, $INF-{\gamma}$ , $INF-{\alpha}$ , $IL-1{\beta}$, IgG, IgM and anti-collagen II were measured in serum and the level of $INF-{\gamma}$, $INF-{\gamma}$/IL-4 ratio in the CIA mouse spleen cell culture. Fourth, histological analysis of the mice joint was performed. Fifth, the expression ratio of $CD3e^+$ cells to $CD19^+$ cells, $CD4^+$ cells to $CD8^+$ cells, $CD11a^+CD19^+$ cells, $CD4^+CD25^+$ cells, $CD3^+CD69^+$ cells, and $CD11b^+Gr-1^+$ cells were checked. Results: 1. The hematocrit was meaningfully (p<0.01, p<0.05) decreased in $ST_{36}$ group in comparison with the control and sham group. 2. The glucose, ALP were meaningfully (p<0.05, p<0.05) decreased in $ST_{36}$ group in comparison with the control group. 3. In the activation of hepatic antioxidase, the catalase was meaningfully (p<0.01) increased in $ST_{36}$ group in comparison with the control group. 4. In the liver, the NO formation was meaningfully (p<0.001) decreased in $ST_{36}$ group in comparison with the nomal, control and sham group. 5. In the activation of splenetic antioxidase, the GSH was meaningfully (p<0.01) increased in $ST_{36}$ group in comparison with the sham group, and the catalase was meaningfully (p<0.001) increased in $ST_{36}$ group in comparison with the control group. 6. In the spleen, the MDA formation was meaningfully (p<0.01, p<0.05) decreased in $ST_{36}$ group in comparison with the control and sham group. 7. In the spleen cell culture, the levels of IL-6 was meaningfully (p<0.05) decreased in comparison with the sham group, $INF-{\gamma}$( was meaningfully (p<0.05) increased in comparison with the control group. Conclusions: According to the results, it is considered that electroacupuncture at Joksamni($ST_{36}$) is effective in increasing the activities of antioxidative enzyme and inhibiting lipid peroxides, and has an immunomodulatory reaction. In conclusion, it han an significant anti-aging effect and need more experimental study.

Damage Study on the Mechanical Fastening in Laminated Composites (복합적층판(復合積層板)의 기계적(機械的) 체결부(締結部)에 관한 파손연구(破損硏究))

  • A series of test was performed measuring the failure strength and failure mode of Gr/Pi, $[0^{\circ}/45^{\circ}/90^{\circ}/-45^{\circ}]_s$ laminate containing a single pin loaded hole. The finite element method is applied to calculate the stress distribution in the laminates, then the failure load and the failure mode were predicted by means of the characteristic length. 12 different geometric variations were developed to analyze the effects of the ratio of specimen width to hole diameter (W/d) and ratio of edge distance to hole diameter (L/d). X-Ray of NDE methods were utilized in finding out the initial defects, damage and the fracture mechanism, and SEM(Scanning Electron Microscopes) was used the evaluation of the fracture mechanism and crack propagation around hole under tension pin loading. $[0^{\circ}/45^{\circ}/90^{\circ}/-45^{\circ}]_s$ laminate are found to be most sensitive to W/d but not so influenced by L/d. The failure mode and tensile strength predicted by the model show agreement with experiment data for pin loading bolted jointed test except range of $L/d{\leqq}3$.

Spectral Analysis of REM Sleep EEG in Narcolepsy and REM Sleep Behavior Disorder (기면병과 렘수면행동장애에서의 렘수면 뇌파 스펙트럼 분석)

  • Introduction: It has been proposed that narcolepsy and REM sleep behavior disorder (RBD) have overlapped symptom profile and pathophysiology. This study was aimed at measuring and comparing changes in EEG frequency band of REM sleep in narcolepsy and RBD, applying EEG spectral analysis method. Methods: Nine patients diagnosed as narcolepsy and the same number of RBD patients were studied. Spectral analysis of the REM sleep EEG was performed in each patient on 9 epochs selected evenly from the first, second, and third REM periods. Then, we compared frequency band percentages of REM sleep EEG in narcolepsy and RBD. Results: Narcolepsy patients had significantly higher delta frequency ratio than RBD ones (p=0.00). In alpha and beta2 frequency bands, RBD patients showed higher percentage than narcolepsy ones. Slow wave sleep was more prevalent in narcolepsy patients. But, no difference of REM sleep percentage was found between the two groups (p=0.93). Conclusion: Higher delta frequency ratio in REM sleep of narcolepsy patients than RBD ones reflects that sleep-promoting mechanism is more dominant in narcolepsy than in RBD.
