Species Composition and Monthly Variation of the Organisms attached to Sargassum beds in Gamak Bay, Korea (가막만 모자반군락지내 부착생물의 종조성 및 월별변동)

  • We studied the organisms attached to Sargassum beds in Gamak Bay from November 2007 to June 2008. The water temperature ranged from 3.3 to $28.7^{\circ}C$ and salinity from 30.6 to 33.8 psu. Eighteen species $143,432\;ind./m^2$ were sampled during study periods. The biomass of the Sargassum beds ranged from a highest of $196.08\;g\;dw/m^2$ in March to a lowest of $0\;g\;dw/m^2$ in July. The major dominant species were Barleeia angustata ($134,430\;ind./m^2$), Caprellidae sp.($2,443\;ind./m^2$) and Gammaridae sp.($4,201\;ind./m^2$). The ecological indices of diversity, evenness, and richness were 0.11-0.86, 0.04-0.39, and 0.74-1.06, respectively.

Purification and Characterization of ${\beta}-Glucosidase$ from Penicillium verruculosum

  • The ${\beta}-glucosidase$ was purified to homogeneity from the culture filtrate of P. verruculosum by column chromatography. The enzyme was a glycoprotein with a relative size of approximately 220 kDa with an isoelectric point of 4.8, which was composed of dimeric protein of 105 kDa. The enzyme was stable up to $60^{\circ}C$ and the presence of glycerol significantly increased its thermostability. The enzyme was found to hydrolyze both ${\beta}-aryl$ and ${\beta}-alkyl-glucosides$ in addition to ${\beta}-glucosyl$ glucose and catalyzed glucosyl transfer to cellobiose. The enzyme attacked laminarin in an exotype-like fashion. The apparent Km's of the enzyme toward cellobiose, laminaribiose, laminarin were 0.53 mM, 0.35 mM and 1.11 mM, respectively. Glucose and glucono-${\delta}-lactone$ were competitive inhibitors for the enzyme. Copper ($Cu^{2+}$), mercury ($Hg^{2+}$) and p-chloromercuribenzoate were strong inhibitors of the enzyme. The immunoblotting result revealed that one form of ${\beta}-glucosidase$ was biosynthesized, irrespective of carbon sources used. Polyacrylamide gel electrophoresis analysis of the in vitro translated product of total RNA from avicel grown mycelium established that the P. verruculosum ${\beta}-glucosidase$ precursor was approximately 95 kDa in size.   • Intestinal tuberculosis (TB) is presented with nonspecific and variable clinical manifestations such as abdominal pain, diarrhea, fever and weight loss. Diagnosis of tuberculous enteritis may be missed or confused with many other chronic gastrointestinal disorders such as the Crohn disease and intestinal neoplasms. The diagnosis should be based on careful clinical evaluations, such as extra-intestinal signs and colonoscopic and histologic findings. Newer techniques such as PCR tests from the specimens through colonoscopic biopsy may be helpful to confirm diagnosis of tuberculous enteritis. The treatment regimens for pulmonary tuberculosis are generally effective for tuberculous enteritis as well. If not treated early, the prognosis of intestinal tuberculosis is poor. We report a case of tuberculous enteritis diagnosed by colonoscopic biopsy and TB PCR which was presented with diarrhea, abdominal pain, intermittent fever and weight loss in a 12-year-old girl with active pulmonary tuberculosis. The patient was treated successfully with antituberculosis agents for 11 months without any complications.

The Effect of L-carnitine and Acetylcarnitine on Sperm Parameters in vitro (생체외 L-carnitine과 Acetylcarnitine의 정자지표 개선 효과)

  • Objectives: To assess the scavenging effect of carnitine derivatives on oxidative damage to sperm during sperm processing, cryopreservation and thawing. Materials and Methods: Fresh semen samples from 20 normal healthy volunteers were collected by masturbation after at least 48 hours abstinence. After liquefaction of semen samples at room temperature, the specimens were diluted with sperm wash media (Ham's F-10, Life technologics) to a uniform density of $20{\times}10^6/ml$. L-carnitine or acetylcarnitine were added with various concentration of $0{\mu}M$, $10{\mu}M$, $30{\mu}M$ in semen sample or cryoprotectant. All specimens were cryopreservated at $-196^{circ}C$ $LN_2$ for 3 days. Sperm motility, vitality, fertilizing capacity, reactive oxygen species formation and the level of lipid peroxidation were analyzed by computer assisted semen analyzer, eosin-nigrosin stain, hypoosmotic swelling test, chemiluminescence and thiobarbituric acid method, respectively, during sperm processing, cryopreservation and thawing. Results: The sperm motility was only increased in proportion to the concentration of acetylcarnitine with no statistical significance (p>0.05). The sperm vitality was also significantly improved in proportion to the concentration of acetylcarnitine with statistical significance (p<0.05). The sperm fertilizing capacity was significantly increased in proportion to the concentration of L-carnitine and acetylcarnitine and reactive oxygen species generation and lipid peroxidation were significantly decreased with same fashion (p<0.05). On comparison of effects between L-carnitine and acetylcarnitine, acetylcarnitine was superior to L-carnitine on the improvement of sperm motility and vitality as well as the suppression of reactive oxygen species generation and lipid peroxidation. Conclusions: These results suggest that carnitine derivatives have a scavenging effect against oxidative damages during sperm processing, cryopreservation and thawing. Therefore, carnitine derivatives may be useful as an oral antioxidant in patients with male infertility due to increased ROS generation.

Probing of Potential Luminous Bacteria in Bay of Bengal and Its Enzyme Characterization

  • The present study dealt with the isolation, identification and enzyme characterization of potential luminous bacteria from water, sediment, squid, and cuttle fish samples of the Karaikal coast, Bay of Bengal, India during the study period September 2007 - August 2008. Bioluminescent strains were screened in SWC agar and identified using biochemical tests. As Shewanella henadai was found to be the most common and abundant species with maximum light emission [69,702,240 photons per second (pps)], the optimum ranges of various physicochemical parameters that enhance the luciferase activity in Shewanella hanedai were worked out. The maximum luciferase activity was observed at the temperature of $25^{\circ}C$ (69,674,387 pps), pH of 8.0 (70,523,671 pps), salinity of 20 ppt (71,674,387 pps), incubation period of 16 h (69,895,714 pps), 4% peptone (70,895,152 pps) as nitrogen source, 0.9% glycerol (71,625,196 pps), and the ionic supplements of 0.3% $CaCO_3$ (73,991,591 pps), 0.3% $K_2HPO_4$ (73,919,915 pps), and 0.2% $MgSO_4$ (72,161,155 pps). Shewanella hanedai was cultured at optimum ranges for luciferase enzyme characterization. From the centrifuged supernatant, the proteins were precipitated with 60% ammonium sulfate, dialyzed, and purified using anion-exchange chromatography, and then luciferase was eluted with 500 mM phosphate of pH 7.0. The purified luciferase enzyme was subjected to SDS-PAGE and the molecular mass was determined as 78 kDa.

Biochemical Properties of Starch Granule Non-Digestive Enzyme(SGNA) of Bacillus polymyxa No.26

  • A $\alpha$-l, 4-D-glucan maltohydrolase $(\beta$-amylase), secreted by the mesophilic aerobic bacterium Bacillus polymyxa No.26, was purified and characterized. The enzyme production was increased after a logarithmic phase of bacterial growth and paralleled with the onset of bacterial sporulation. By applying anion exchange chromatography and gel filtration the enzyme was purified 16.7-fold and had a specific activity of 285.7 units/mg. Two enzyme activities were eluted on a column of DEAE-Sephadex chromatography, and they were designated as E-I for a major enzyme peak and E-II for a minor peak. Of them, E-I enzyme peak was further purified by using gel chromatography. The molecular mass of this enzyme was determined to be 64, 000 daltons and consisted of a single subunit, showing an isoelectric point of 8.9. The enzyme was able to attack specifically the $\alpha$-l, 4-glycosidic linkages in soluble starch and caused its complete hydrolysis to maltose and $\beta$-limited dextrin. This amylolytic enzyme displayed a temperature optimum at $45^\circ{C}$ and a pH optimum at 7.0. The amino acid composition of the purified enzyme was quite similar to the other bacterial $\beta$-amylases reported. Surprisingly, the purified enzyme from this aerobe only exhibited hydrolytic activity on soluble starch, not on starch granules. The degradation of from starch by $\beta$-amylase was greatly stimulated by pullulanase addition. These results differentiated from other $\beta$-amylases reported. Based on a previous result that showed the enzyme system involves in effective degradation of raw starch granules, this result strongly suggested that the purified enzyme (E-I) can be a synergistic part of starch granule-digestion and E-II plays a crucial role in digestion of starch granules.

Current Application of Embryo Cryopreservation for Farm Animals (가축 수정란 동결보존의 최근 이용방법)

  • This paper reviews the most important steps that have generated consistent progress in principles and developmental progress of embryo cryopreservation, and also study on freezing procedure and its application by conventional method and current improved method for freezing procedure and its appilcation of embryo cryopreservation in farm animals. Four were of particular interest: 1.The transport of water across the ccli membrane (zona pellucida) during freezing and thawing accordinglyplays a role in determing whether the celi survives. This movement of water is controlied mainly by extracellular phase changes and by the nature and concentration of any cryoprotective agent present. Therates of cooling, freezing and warming, and the intervals over which they are applied are further decisi've factors in determining whether a cryopreservation procedure allows survival after thawing. 2.The first successful deep freezing experiments with sheep morula and blastocysts during the seventies were based on the early procedures used for mouse embryos.Current research during the eighties is developed with the aim of simplifying and improving current procedures such as one-step dilution and rapid or ultra-rapid cooling by using the model of laboratory animals. 3.The conventional method for the embryo cryopreservation is described. An alternative to this method which may result in high survival and also in reducing of the freezing and thawing time is done by combing a permeable cryoprotectant such as glycerol, DMSO or propanediol and a non-permeable compound such as sucrose, trehalose, raffinose or lactose. 4.Finally a different approach to the preservation of embryos, named vitrification, is introduced. This procedure depends upon the ability of concentrated solutions of cryoprotective agents such as glycerol and propanediol to supercool to very low temperature (-196$^{\circ}C$) during rapid cooling before solidifying without formation of ice. However, more complete data are necessary for successful vitrification of blastocysts.

  • To assess its suitability as a prodrug of 5-fluorouracil (5-FU), 1-glycyloxymethyl-5-FU HCl (GFU), a 5-fluorouracil derivative having a glycyloxymethyl group at the N-l position was synthetized. Its physicochemical properties and hydrolysis kinetics, in aqueous solution of pH $1{\sim}10$ and in the presence of human plasma or rat liver homogenate were studied. Its acute toxicity and antitumor activity against sarcoma 180 were also examined, GFU showed higher lipid/water partition coefficient than 5-FU. The calculated $pK_{\alpha}$ values of 5-FU and GFU were 8.02 and 7,20, respectively. The decomposition rates of GFU in aqueous solution showed a pH-dependence over the pH range used, which could be ascribed to solvent catalysed hydrolysis reaction at pH lower than 4,16 and to specific hydroxide ion hydrolysis reaction at pH higher than 4,16, The half-life of GFU was 6,9 min in 80% human plasma solution and less than 3 min in rat liver homogenate at $37^{\circ}C$, The $LD_{50}$ value of 5-FU was 240 mg/kg while that of GFU was 440.6 mg/kg (226 mg as 5-FU). Both of 5FU and GFU showed a strong antitumor activity, Therapeutic ratios of 5-FU and GFU were 3.07 and 3.55, respectively.

The Study on Anti-galling Characteristics of 304 Stainless Steel by Sn-Al Thermal Diffusion Coating (Sn-Al 열 확산 코팅에 따른 304 스테인리스강의 고착방지성능 연구)

  • 볼트, 너트 등의 파스너는 건축 재료나 기계부품을 고정하는 데 사용하는 기계요소로, 건축, 철도, 조선 등 전 산업분야에 걸쳐 사용되고 있다. 그 중 스테인리스 소재의 볼트, 너트는 뛰어난 내식성과 저렴한 가격으로 널리 사용되고 있는데, 소재의 특성 및 작업현장의 상황, 온도의 변화 등의 원인에 의해 고착현상(galling)이 발생한다. 고착현상이란 성분 혹은 표면경도가 비슷한 금속의 나사산을 조이는 과정에서 발생하는 압력의 증가 및 마찰력에 의해 냉간 용접(cold welding)이 일어나는 것으로 나사산의 표면이 눌어붙게 된다. 이러한 고착현상은 스테인리스 소재에서 많이 발생하는데, 한번 발생한 후에는 비파괴 해소가 불가능한 상태가 되어 경제적 손실을 야기한다. 이러한 고착현상의 해소를 위해 본 연구에서는 주석과 알루미늄을 사용한 새로운 열 확산 코팅 기술을 개발하고 이를 304 스테인리스강에 적용하여 열처리 온도에 따른 특성변화를 확인하였다. 열 확산 코팅을 위해 팩 세멘테이션 방법을 이용하여 아르곤 분위기 하에서 열처리 하였고, 온도는 $200{\sim}250^{\circ}C$에서 코팅을 수행하였다. 이에 따른 코팅 전과 후의 표면 및 단면 분석을 통해 성공적으로 코팅층이 형성됨을 확인하였고, 온도가 증가함에 따라 코팅성분의 양이 증가하는 현상을 보임을 알 수 있었다. 또한, 고착방지성능을 확인하기 위하여 ASTM G196-08 시험을 통해 코팅조건에 따른 고착현상을 분석하였으며, 그 결과 기존에 코팅되지 않은 304 스테인리스강보다 고착현상이 개선됨을 확인하였다. 따라서 304 스테인리스강 소재의 볼트, 너트 제품에 주석-알루미늄 코팅층을 적용시키면 기존의 고착현상을 개선하고 서비스 품질을 향상시킬 수 있을 것으로 판단된다.

A STUDY ON THE AGING DEGRADATION OF ETHYLENE-PROPYLENE-DIENE MONOMER (EPDM) UNDER LOCA CONDITION

  • The aging degradation and lifetime assessment of a domestic class 1E Ethylene-Propylene-Diene-Monomer (EPDM), which is a popular insulating elastomer for electrical cables in the nuclear power plants, were studied for equipment qualification verification under the Loss of Coolant Accident (LOCA) conditions. The specimens were acceleratively aged, underwent a LOCA environment, as well as tested mechanically, thermo-gravimetrically, and spectroscopically according to the American Society of the Testing of Materials (ASTM) procedures. The tensile test results revealed that the elongation at break gradually decreased with an increasing aging temperature. The lifetime of EPDM aged isothermally at $140^{\circ}C$ was 1,316 hours and reduced to 1,120 hours after experiencing the severe accident test. The activation energies of the elongation reduction were $1.10{\pm}0.196$ eV and $0.93{\pm}0.191$ eV before and after the LOCA condition, respectively. The TGA test results also showed that the activation energy of the aging decomposition decreased from 1.35 eV to 1.02 eV after undergoing the LOCA environment. Although the mechanical property changes were discernibly observed during the aging process, along with the LOCA simulation, the FT-IR analysis showed that the spectroscopic peaks and their intensities did not alter significantly. Therefore, it can be concluded that the degradation of the domestic class 1E EPDM due to aging can be tolerable, even in severe accident conditions such as LOCA, and thus it qualifies as a suitable insulating material for electrical cables in the nuclear power plants.
