Calcium Modulates Excitatory Amino Acid (EAA)- and Substance P-induced Rat Dorsal Horn Cell Responses

  • Excitatory amino acid (EAA) and substance P (SP) have been known to be primary candidates for nociceptive neurotransmitter in the spinal cord, and calcium ions are implicated in processing of the sensory informations mediated by EAA and SP in the spinal cord. In this study, we examined how $Ca^{2+}$ modified the responses of dorsal horn neurons to single or combined iontophoretical application of EAA and SP in the rat. All the LT cells tested responded to kainate, whereas about 55% of low threshold (LT) cells responded to iontophoretically applied NMDA. NMDA and kainate excited almost all wide dynamic range (WDR) cells. These NMDA- and kainate-induced WDR cell responses were augmented by iontophoretically applied EGTA, but suppressed by $Ca^{2+},\;Mn^{2+},$ verapamil and ${\omega}-conotoxin$ EVTA, effect of verapamil being more prominent and well sustained. $Ca^{2+}$ and $Mn^{2+}$ antagonized the augmenting effect of EGTA. On the other hand, prolonged spinal application of EGTA suppressed the response of WDR cell to NMDA. SP had triple effects on the spontaneous activity as well as NMDA-induced responses of WDR cells: excitation, inhibition and no change. EGTA augmented, but $Ca^{2+},\;Mn^{2+}$ and verapamil suppressed the increase in the NMDA-induced responses and spontaneous activities of WDR cells following iontophoretical application of SP. These results suggest that in the spinal cord, sensory informations mediated by single or combined action of EAA and SP can be modified by the change in calcium ion concentration.

간호사의 아동 통증 사정 및 관리에 대한 실태조사 (A status of Assessment and Management about Children in Pain)

  • This study was Performed to understand Pediatric Pain management status and nurses' knowledge and attitudes toward it. In addition. it aimed to provide basic data in order to establish effective nursing intervention strategies by confirming the barriers of effective pain management in practice. The subjects were 195 nurses working in pediatric units(general pediatric unit. oncology unit, neonatal unit. neonatal ICU, Pediatric ICU) of 8 university hospitals and one general hospital. Data was collected by the questionnaire from the 3rd of August to the 20th of September in 1999. The instrument developed by Sanna(1999) to measure nurses' knowledge and their attitudes and the other tool by Cleeland(1984) to evaluate barriers in effective Pain management was used. Results of this study are summarized as follows 1. Most nurses learn about the pain management knowledges from through regular curriculum of nursing school(62.0%) And almost nurses(90.8%) don't used to utilize pain assessment tool, but the 'Faces Rating Scale' is the most frequently used by nurses. 2. The use of pain medication(65.6%) is most frequently taken by nurses as pain management and is followed by massage (55.9%) , distraction(27.7%). 3. Nurses' knowledge level is moderate (Mn=3.07). Nurses don't seem to understand pediatric Physical development (Mn = 2.86), psychological development(Mn=2.94) well, meanwhile they seem relatively Quite knowledgeable about the way pain emerges. 4. Nurses' attitudes toward pain is based on behavioral and physiological responses to Pain. They believe 'changes in behavior are a way of assessing pain in child' and 'acute pain increases the number of respiration' Nurses are ready to accept pediatric pain, but are not positive in adopting pain intervention in practice. 5. The barriers of effective pain management are inadequate assessment of pain and pain relief(81.5%), ineffective incorporation among health professionals(80%), and lack of equipment or skills(80.0%).

High Strength Nanostructured Metastable Alloys

  • Nanostructured high strength metastable Al-, Mg- and Ti-based alloys containing different amorphous, quasicrystalline and nanocrystalline phases are synthesized by non-equilibrium processing techniques. Such alloys can be prepared by quenching from the melt or by powder metallurgy techniques. This paper focuses on one hand on mechanically alloyed and ball milled powders containing different volume fractions of amorphous or nano-(quasi)crystalline phases, consolidated bulk specimens and, on the other hand. on cast specimens containing different constituent phases with different length-scale. As one example. $Mg_{55}Y_{15}Cu_{30}$- based metallic glass matrix composites are produced by mechanical alloying of elemental powder mixtures containing up to 30 vol.% $Y_2O_3$ particles. The comparison with the particle-free metallic glass reveals that the nanosized second phase oxide particles do not significantly affect the glass-forming ability upon mechanical alloying despite some limited particle dissolution. A supercooled liquid region with an extension of about 50 K can be maintained in the presence of the oxides. The distinct viscosity decrease in the supercooled liquid regime allows to consolidate the powders into bulk samples by uniaxial hot pressing. The $Y_2O_3$ additions increase the mechanical strength of the composites compared to the $Mg_{55}Y_{15}Cu_{30}$ metallic glass. The second example deals with Al-Mn-Ce and Al-Cu-Fe composites with quasicrystalline particles as reinforcements, which are prepared by quenching from the melt and by powder metallurgy. $Al_{98-x}Mn_xCe_2$ (x =5,6,7) melt-spun ribbons containing a major quasicrystalline phase coexisting with an Al-matrix on a nanometer scale are pulverized by ball milling. The powders are consolidated by hot extrusion. Grain growth during consolidation causes the formation of a micrometer-scale microstructure. Mechanical alloying of $Al_{63}Cu_{25}Fe_{12}$ leads to single-phase quasicrystalline powders. which are blended with different volume fractions of pure Al-powder and hot extruded forming $Al_{100-x}$$(Al_{0.63}Cu_{0.25}Fe_{0.12})_x$ (x = 40,50,60,80) micrometer-scale composites. Compression test data reveal a high yield strength of ${\sigma}_y{\geq}$700 MPa and a ductility of ${\varepsilon}_{pl}{\geq}$5% for than the Al-Mn-Ce bulk samples. The strength level of the Al-Cu-Fe alloys is ${\sigma}_y{\leq}$550 MPa significantly lower. By the addition of different amounts of aluminum, the mechanical properties can be tuned to a wide range. Finally, a bulk metallic glass-forming Ti-Cu-Ni-Sn alloy with in situ formed composite microstructure prepared by both centrifugal and injection casting presents more than 6% plastic strain under compressive stress at room temperature. The in situ formed composite contains dendritic hcp Ti solid solution precipitates and a few $Ti_3Sn,\;{\beta}$-(Cu, Sn) grains dispersed in a glassy matrix. The composite micro- structure can avoid the development of the highly localized shear bands typical for the room temperature defor-mation of monolithic glasses. Instead, widely developed shear bands with evident protuberance are observed. resulting in significant yielding and homogeneous plastic deformation over the entire sample.

  • 방사성 폐기물의 핵종 재고량 평가에 필요한 정량분석을 위하여 다양한 매질의 방사성 폐기물 시료로부터 $^{99}Tc,\;^{94}Nb,\;^{55}Fe,\;^{90}Sr$$^{59}Ni(^{63}Ni)$의 선택적 분리에 관한 연구가 수행되고있다. 방사성 폐기물 용해용액의 화학조성을 모사하여 만든 모의 방사성 폐기물 용해용액을 사용하여 음이온교환수지법과 Sr-Spec 추출 크로마토그래피로 Re ($^{99}Tc$ 대용원소), Nb, Fe 및 Sr을 분리해 낸 후 Ni과 함께 회수되는 Ce, Ca, Mg, Al, Cr, Ti, Mn 및 Cu와 Ni의 분리 거동을 Ni-Spec 추출크로마토그래피와 양이온교환수지법으로 조사하였다. 공존원소들로부터 Ni의 선택적 분리와 기체비례계수법에 의한 방사능 측정에 적합한 radionuclide source를 만들기 위하여 공존원소들에 대한 가리움제로 ammonium citrate 및 tartaric acid를 사용하는 dimethylglyoxime 침전법의 적용에 관하여 기술하였다.

Electrochemical Properties of a Zirconia Membrane with a Lanthanum Manganate-Zirconia Composite Electrode and its Oxygen Permeation Characteristics by Applied Currents

  • An electrochemical oxygen permeating membrane (OPM) is fabricated using Zr0.895Sc0.095Ce0.005Gd0.005O2-δ (ScCeGdZ) as the solid electrolyte and aLa0.7Sr0.3MnO3-bScCeGdZ composite (LZab, electrode) as the electrode. The crystal phase of the electrode and the microstructure of the membrane is investigated with X-ray diffraction and scanning electron microscopy. The electrochemical resistance of the membrane is examined using 2-p ac impedance spectroscopy, and LZ55 shows the lowest electrode resistance among LZ82, LZ55 and LZ37. The oxygen permeation is studied with an oxygen permeation cell with a zirconia oxygen sensor. The oxygen flux of the OPM with LZ55 is nearly consistent with the theoretical value calculated from Faraday's Law below a critical current. However, it becomes saturated above the critical current due to the limit of the oxygen ionic conduction of the OPM. The OPM with LZ55 has a very high oxygen permeation flux of ~ 3.5 × 10-6 mol/㎠s in I = 1.4 A/㎠.

팽이버섯으로부터 Lectin의 정제와 특성 (Purification and Characterization of the Lectins from Mushroom Flammulina velutipes)

  • 식용버섯인 팽이버섯(Flammulina velutipes)으로부터 적혈구 응집능력을 갖는 렉틴(flammulina velutipes lectin, FVL) FVL-1과 FVL-2을 정제하였다. FVL-1과 FVL-2의 분자량은 각각 10.6 kDa과 37 kDa으로 추정되었다. FVL-2는 사람적혈구의 경우 모든 혈액형에서 응집현상을 나타내었으나, FVL-1은 O형의 적혈구에 대하여 높은 상대활성도를 나타내었다. 렉틴의 hemagglutination inhibition test 결과 FVL-1과 FVL-2는 fetuin, bovine submaxillary mucin, asialofetuin, porcine stomach mucin에 의해 적혈구 응집력이 저해되었다. FVL-1은 1.56 mM $Cu^{2+}$에서 적혈구 응집력이 저해되었으며, $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, $Fe^{2+}$ 등에서도 저해되었다. 반면 FVL-2는 금속 이온에 대한 저해가 나타나지 않았다. pH 안정성에 있어서 FVL-1은 pH $5{\sim}11$에서 안정하였으며, FVL-2는 pH 4까지의 범위에서 안정하였다. 온도 안정성에 있어서는 FVL-1과 FVL-2 각각 $55^{\circ}C$, $45^{\circ}C$까지 안정하였다. FVL-1은 단백질이었으며 FVL-2는 당 단백질이었다.

포도주에서 분리한 Saccharomyces cerevisiae JS59가 생성하는 Invertase의 정제 및 특성 (Purification and Characterization of an Invertase Produced with Saccharomyces cerevisiae JS59 Isolated from Home-made Wine)

  • 전화당(invert sugar)을 생산하기 위하여 포도주로부터 분리한 효모가 생산하는 invertase의 특성을 조사한 결과는 다음과 같다. 포도주로부터 분리한 효모는 지방산 분석을 통하여 Saccharomyces cerevisiae JS59로 잠정 동정되었다. 본 균주가 생성하는 invertase를 ammonium sulfate 침전, DEAE-Sephadex A-50, Sephadex G-200 column chromatography 법으로 정제하였을 때 단일성을 보였으며, specific activity가 7620.9 unit/mg, 최종 회수율은 13.9로 약 14배 정제된 효소를 얻었다. 본 효소의 $K_m$ 값은 11.5 mM이었다. SDS-PAGE로부터 분자량은 38.5 kDa으로 나타났다. 정제된 invertase의 최적 pH는 5였고, pH 4에서도 94%의 높은 효소활성을 나타냈으며 4에서 6까지의 pH영역에서 안정하였고, $55^{\circ}C$에서 최적 활성을 나타내었으며 $50^{\circ}C$까지는 안정하였다. $Ag^{2+}$$Hg^{2+}$에 의해서 저해를 받았고, $Co^{2+}$, $Mn^{2+}$에 의해서는 효소활성이 증가되었으며, 기질과 효소 반응물을 thin layer chromatography로 분석한 결과, 본 효소는 기질인 sucrose를 완전히 분해하여 환원당을 생성함이 확인되었다.

  • 생물학적 난분해성 물질인 폴리비닐 알콜(PVA : Polyvinyl alcohol)을 탄소원 및 에너지원으로 이용하는 Xanthomonas campestris J2Y로부터 PVA oxidase를 생산하여 정제하기 위하여 PVA가 탄소원으로 첨가된 배지에서 진탕배양한 배양액을 원심분리한 후 상등액을 10 mM phosphate buffer(pH 7.5)로 평형시킨 DEAE -cellulose를 통과시켜 얻은 분획을 사용하여 DEAE-cellulose와 Sephadex G-150을 이용한 Gel filtration을 통하여 PVA oxidase를 정제하였다. 정제된 PVA oxidase는 polyacrylamide gel 전기영동으로 단일밴드로 확인되었으며 SDS-polyacrylamide gel 전기영동과 Sephadex G-150 column chromatography를 통해 측정한 결과 55,000 daltons 이었다. PVA oxidase의 최적 pH는 7이고 최적온도는 $37^{\circ}C$였다. 열 안정성은 $50^{\circ}C$까지는 70% 이상의 안정성을 나타내었고, pH에 대한 안정성은 5-11에서 비교적 안정하였다. 금속이온에 대한 영향은 $Ag^{2+},\;Hg^{2+},\;Sn^{2+}$ 등에서는 아주 강한 저해를 받았고, $Co^{2+},\;Fe^{2+},\;Zn^{2+},\;Pb^{2+}$에서는 50% 정도의 저해를 받았다. 반면에 $Mn^{2+},\;Cu^{2+}$는 효소활성을 증가시켰다. PVA에 정제 PVA oxidase의 Km치는 $7.04{\times}10\;^2mmol/{\ell}$이었다.

  • 기존의 무선랜 모델에서는 현재 접속중인 AP가 송신하는 비콘 프레임의 수신이 일정 회수 이상 실패해야 새로운 AP를 찾는 스캔 절차가 시작된다. 이 경우 비콘 보다 길이가 큰 데이터 패킷은 비콘 손실이 발생하기 이전부터 이미 손실이 발생함으로 이동노드의 핸드오버 기간 중 많은 데이터 손실이 발생하게 된다. 본 논문에서는 비콘 프레임이 정상적으로 수신되는 경우에도 수신전력 레벨이 정해진 임계치보다 작고 동시에 이전 비콘 수신전력보다 작은 경우에 카운터 값을 증가시키고, 이 카운터 값이 정해진 값 이상이 되면 새로운 AP를 찾는 스캔 절차를 수행하도록 수정 모델을 개발하였다. 또한 비콘 수신전력 임계치 설정의 타당성을 검토하고, UDP와 TCP 트래픽 환경에서 성능향상 정도를 분석하였다.

Study of Thermostable Chitinase Enzymes from Indonesian Bacillus K29-14

  • Thermophilic microorganisms capable of producing chitinase enzymes were screened from samples collected from several crater and geothermal areas. The chitinolytic microorganisms were grown in a selective medium containing colloidal chitin. The Bacillus K29-14 isolate was found to exhibit the highest chitinase and chitin deacetylase activities. When grown in a chitin-containing medium, the isolate produced extracellular chitinase after 24 h of incubation. The optimum temperature and pH for the chitinase were $55^\circ{C}$ and pH 7, respectively, while those for the chitin deacetylase were $55^\circ{C}$ and pH 8, respectively. The thermostable chitinase and chitin deacetylase also retained 80- 90% of their activity after incubation for 5 h at $70^\circ{C}$. The divalent cations $CoCl_2\;and\;NiCl_2$, increased the chitinase activity, while $ZnCl_2$, inhibited the enzyme. The chitin deacetylase was also activated by the presence of $MgCl_2$ and inhibited by $MnCl_2,\;NiCl_2,\;and\;CaCl_2$. A zymogram analysis revealed several forms of chitinase, with a 67 kDa form being the major enzyme.
