A Determination of the Salicylate Content of Ingredients Commonly Used in Korean foods Suggested for Allergic Patients

  • Salicylate from plant foods and histamine from animal foods have been suggested to cause various food allergic symptoms. Salicylate regarded as one of the inducing agents f3r chronic urticaria and asthma in allergic patients was studied. There is much interest about the effects of salicylate in food allergy. It it recently suggested that the intake of salicylate from foods may have contributed to chronic urticaria and asthma. The purpose of this study is to analyze the salicylate content in various foods. 153 ingredients commonly used in Korean foods were analyzed by HPLC. Among the vegetables, chard(8.22$\mu\textrm{g}$/g), soybean sprout(5.13$\mu\textrm{g}$/g), perilla lean3.03$\mu\textrm{g}$/g), Chinese cabbage(2.25$\mu\textrm{g}$/g), carrot(2.16$\mu\textrm{g}$/g), and squash(1.33$\mu\textrm{g}$/g) con-rained higher amounts of salicylate as compared to other vegetables. Among fruits, oranges(5.13$\mu\textrm{g}$/g), grapefruits(2.90$\mu\textrm{g}$/g), and plums(1.65$\mu\textrm{g}$/g) contained the most. In animal foods, almost no salicylate was found except in chocolate milk(0.32$\mu\textrm{g}$/g). It was also found in black pepper(8.2$\mu\textrm{g}$/g), coffee(can)(5.00$\mu\textrm{g}$/g), mustard(4.16$\mu\textrm{g}$/g), curry powder(3.24$\mu\textrm{g}$/g), kochujang(2.37$\mu\textrm{g}$/g), cream powder(1.19$\mu\textrm{g}$/g), brown rice(1.04$\mu\textrm{g}$/g) and sesame oil(1.00$\mu\textrm{g}$/g). All information found in this study can be used as nutritional education materials in potentially allergic people for a prudent diet plan and can also provide proper dietary guidelines for these allergic patients. Effects of Concentration of $\textrm{NO}_3^\;-$ $\textrm{K}^+$ and $\textrm{Ca}^{++}$ in Nutrient Solution on the Seedling Growth of ‘Mudeungsan’ Watermelon (배양액내 $\textrm{NO}_3^\;-$, $\textrm{K}^+$$\textrm{Ca}^{++}$ 농도가 무등산수박 유묘 생장에 미치는 영향)

  • This experiment was conducted to investigate the effects of concentration of NO$_{3}$$^{[-10]}$ , $K^{+}$ and $Ca^{++}$ in nutrient solution on the seedlings growth of ‘Mudeungsan’watermelon. Seeds were sown on March 16, 1997. NO$_{3}$$^{[-10]}$ , $K^{+}$ and $Ca^{++}$ was treated with three different levels, respectively, NO$_{3}$$^{[-10]}$ = 106, 206, 406ppm; $K^{+}$ = 150, 200, 400 ppm; $Ca^{++}$ = 150, 200, 400 ppm. Plant growth was investigated at four-days interval in 10 days after treatment. Increasing N concentration in the nutrient solution increased the plant height, leaf area, number of leaves, fresh and dry weight of shoot while high concentration of K and Ca in the nutrient solution decreased the seedling growth in terms of leaf area, leaf dry weight. The uptake of N, K and Mg in petiole sap was positively affected by the adding of 206 ppm of nitrate, 150 ppm of potassium and 200 ppm of calcium levels, respectively. The P and Ca uptake in petiole sap was not affected by the N and K concentrations in the nutrient solution while K concentration above 200 ppm decreased the N and Mg content in petiole sap. High concentration of Ca in the nutrient solution increased the Ca content in petiole sap while Ca concentration above 200 ppm decreased the N content.

Isoflavone, Phytic Acid and Oligosaccharide Contents of Domestic and Imported Soybean Cultivars in Korea (국내산 콩과 수입콩의 Isoflavone, Phytic Acid 및 Oligosaccharides 함량)

  • Chemical composition, total dietary fiber(TDF), oligosaccharide, isoflavone and phytic acid contents of seven domestic and three imported soybean cultivars were determined. TDF contents were ranged from 16.83 to 21.71%(w/w) without remarkable differences among soybean cultivars. Phytic acid contents of domestic cultivars such as Geomjongkong 1 (3.02%) and Dawonkong (2.92%) were higher than imported ones such as Canadian (2.07%) and American (2.22%) soybeans for soybean sprout and US No. 1 (2.16%). The phytic acid contents of cotyledon parts were 1.5 to 2 times higher than those of hypocotyl parts. Isoflavone contents in whole seed were wide ranged from 371 to 2,398 $\mu\textrm{g}$/g among cultivars and it were existed mainly as glucoside isomers. Profiles of isoflavone aglycones were composed of 52% genistein, 36% daidzein and 11% glycitein. Hwanggumkong, Dawonkong, Geomjongkong 1 and American soybean for sprout contained lower isoflavone than others. Hypocotyl parts of soybeans contained from 6120.1 to 16921.1 $\mu\textrm{g}$/g isoflavone with ratio of 48% glycitein, 35% daidzein and 10% genistein. Isoflavone contents of cotyledon parts were ranged 375-2393 $\mu\textrm{g}$/g, composition of which had no glycitein, 55% genistein and 38% daidzein. Oligosaccharide contents were similar among cultivars. Stachyose of 3.0-3.9%, raffinose of 0.8-1.2%, sucrose of 4.5-7.8% and fructose of 0.3-0.8% were determined.

  • Typical metal oxide thin films having optical and electrical properties are widely used as inorganic functional materials. Liquid phase deposition(LPD) method, a new low temperature process, has been developed for the several advantages of no vacuum system, low cost, high throughput, and low processing temperature(<$50^{\circ}C$). Silica powder was added to 40wt% hydrofluoro-silicic acid($H_2$SiF\ulcorner) to obtain an immersing solution of silica-saturated hydrofluorosilicic acid solution. Boric acid solution was continuously added in the range from 0 to 0.05M to prepare supersaturated hydrofluorosilicic acid solution. LPD $SiL_2$film was formed with the variation of added amount of $H_2$O. The SiO$_2$thin film could be prepared from hydrofluorosilicic acid by LPD method. The thickness of LPD $_SiO2$film was influenced by the boric acid concentration and added amount of $H_2$O. Silicon in thin film existed as SiF\ulcorner by Raman spectrum.

Isolation and Identification of Antimicrobial Compound from Mokdan Bark (Paeonia suffruticosa ANDR) (목단피로부터 식품부패 미생물에 대한 항균성 물질의 분리 및 동정)

  • Antimicrobial activity of Mokdan bark (Paeonia suffruticosa $A_{NDR}$) was investigated. Methanol extract of dried Mokdan was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as L. monocytogenes, and E. coli at 500 $\mu\textrm{g}$/disc. Ethylacetate fraction was further fractionated into 3 fractions by silica gel column and thin layer chromatography (TLC). The results showed that ethylacetate fractions No. 1 and 2 had the highest antimicrobial activity. They were mixed again, reseparated, and 3 fractions were obtained. Among them, No. 1 had the highest inhibitory effect on the microorganisms, No. 1 fraction was identified as isobutyl isopentanoate by HPLC, and GC-MS.

  • This stduy was carried out to investigate the effect of concentration of PC12 and washing of sperm of sperm motility and acrosome reaction, and the effect of sperm incubated in mTALP solution containing PC12 112.5$\mu\textrm{g}$/ml on development of follicular oocytes matured in vitro. The results obtained were as follows. 1. When fresh sperm was once washed and then incubated in mTALP solution containing 0, 75, 112.5 and 225$\mu\textrm{g}$/ml PC12 for 15minutes, 225$\mu\textrm{g}$/ml showed significantly higher percent of acrosome reacted sperm than 0, 75, 112.5 and 225$\mu\textrm{g}$/ml. 2. When once or twice washed fresh sperm was cultured in mTALP containing 112.5$\mu\textrm{g}$/ml PC12 for 15minutes, no-washing showed significantly higher percent of motile sperm than that once- and twice-washing showed significantly higher percent of acrosome reacted seprm than no- and once-washing. 3. When sperm was cultrued in mTALP containing PC12, 112.5$\mu\textrm{g}$/ml for 15minutes and then was cocultured with bovine follicular oocytes matured in vitro, 11.2 to 22.4% of the oocytes were coleaved to more than 2cell stage.

Immunostimulation Activity of the Crude Polysaccharides Fractionated from Eleutherococcus senticosus, and its Application to Prevent of Tumors by Combination Therapy with Cisplatin (오가피로부터 분리된 조다당 분획물의 면역자극활성 및 Cisplatin과의 병용에 의한 항암 상승작용의 유도)

  • In order to study the clinical usefulness of crude polysaccharides fractionated from Eleutherococcus senticosus, EN-3, in eliminating tumors, we have investigated the effect of combination therapy on the murine tumor metastasis and growth models. In experimental metastasis of colon26-M3.1 cells, prophylactic intravenous (i.v.) administration of EN-3 (0.5, 5, and 50 $\mu\textrm{g}$/mouse) inhibited tumor metastasis compared with tumor control group in 33.6, 66.8, and 81.8% respectively. The administration of EN-3 (50 $\mu\textrm{g}$/mouse) also exhibited a 66.1% therapeutic effect on lung tumor metastasis. Although EN-3 induced no toxic effect on both tumor cell and normal splenocyte in the concentration below 100 $\mu\textrm{g}$/mι in in vitro, it induced significant proliferating activity on normal splenocyte in the concentration-dependent manner. In an analysis of NK-cell activity, i.v. administration of EN-3 (4∼100 $\mu\textrm{g}$/mouse) significantly augmented NK cytotoxicity to YAC-1 tumor cells. The combination treatments of cisplatin (10 $\mu\textrm{g}$) and EN-3 (5 $\mu\textrm{g}$) induced synergistic effect on the inhibition of tumor metastasis in experimental tumor metastasis model produced by colon26-M3.1 cells. In addition, the combination treatments also exhibited prolongation of lifespan in S∼180 tumor bearing mouse for over the 60 days. Even though cisplatin (2.5 $\mu\textrm{g}$/mι) exhibited cytotoxicity to tumor cells and inhibited tumor growth over 95% in in vitro, combination treatment with EN-3 (20 $\mu\textrm{g}$/mι) was induced splenocyte proliferation and produced cytokines, such as TNF-$\alpha$, IL-1 and IL-12, from the macrophages. These results suggested that EN-3 stimulate immune system non-specifically and apply to the biological response modifiers (BRM) in chemo-immunotherapy for tumor prevention.

Y-TZP Fine Powder Preparation by Ultrasonic Spray Pyrolysis (초음파 분무 열분해법에 의한 Y-TZP 미립자의 합성)

  • Aqueous solutions of metallic salts, ZrO(NO3)2.2H2O and Y(NO3)3.5H2O were used as raw materials to synthesize crystalline submicron spherical powders of Zr0.94Y0.06O1.97 with tetragonal crystal phase. Each aqueous solution was mixed on the magnetic stirrer to homogenize for 12 hours. The concentration of the mixed solutionwas changed from 0.01mol/$\ell$ to 0.1mol/$\ell$ calculated as the concentration of Zr0.94Y0.06O1.97. Ultrafine droplets of starting mixed solution were sprayed by the ultrasonic vibrator and carried into the furnace kept at 55$0^{\circ}C$, $650^{\circ}C$, 75$0^{\circ}C$ and 85$0^{\circ}C$ using carrier gas of air (10$\ell$/min) and pyrolysed to form Y-TZP fine powders. The results of this exeriment were as follows. 1) Synthesized powders were nonagglomerated and spherical type. 2) Particle size distribution was narrow between 0.1${\mu}{\textrm}{m}$ and 1${\mu}{\textrm}{m}$. 3) Forming reaction Y-TZP was finished above synthetic temperature 75$0^{\circ}C$. 4) As the synthetic temperature rised from 55$0^{\circ}C$ to 85$0^{\circ}C$, the mean particle size decreased from 0.35${\mu}{\textrm}{m}$ to 0.22${\mu}{\textrm}{m}$ in the concentration of starting solution with 0.02mol/$\ell$. 5) At 75$0^{\circ}C$ of synthetic temperature, the concentration changes of starting solution from 0.01mol/$\ell$ to 0.1mol/$\ell$ increased the mean particle size from 0.24${\mu}{\textrm}{m}$ to 0.38${\mu}{\textrm}{m}$. 6) Chemical compositions of each synthesized particle were homogeneous nearly.

THE EFFECT OF POLYMERIZATION TECHNIQUES ON THE MICROLEAKAGE OF COMPOMER (광중합방식이 콤포머의 변연폐쇄에 미치는 영향)

  • The aim of this study was to evaluate the effect of various polymerization techniques on the microleakage of compomer restorations. Fifty extracted human premolars and molar were used and randomly divided into 5 groups. After cavity preparation, compomer (F2000$^{\circledR}$) was filled according to the manufacturer's directions. All groups, except group 5, were filled using an incremental technique. Group 1 was polymerized for 40 seconds at a continuous 485mW/$\textrm{cm}^2$ with a VIP$^{\circledR}$(Bisco, USA) light cure unit. Group 2 was polymerized for 20 seconds at 345mW/$\textrm{cm}^2$ and then for 20 seconds at 645mW/$\textrm{cm}^2$ with the VIP equation omitted light cure unit. Group 3 was polymerized at 400mW/$\textrm{cm}^2$, gradually increased to 50mW/$\textrm{cm}^2$ 10 seconds until 550mW/$\textrm{cm}^2$ was reached; total 40 seconds with a Spectrum 800$^{\circledR}$ (Dentsply Caulk, USA) light cure unit. Group 4 was polymerized for 3 seconds using an incremental technique with a Flipo$^{\circledR}$ (LOKKi, France) light cure unit. Group 5 was polymerized for 3 seconds using a bulk fill technique with the Flipo$^{\circledR}$ light cure unit. The specimens were embedded with acrylic resin, and were sectioned with diamond saws in a mesiodistal direction along the longitudinal axis of the tooth so as to pass through the center of the restoration, and three surfaces (occlusal, pulpal, and gingival) were examined with SEM. The results were as follows ; 1. Group 5 showed a significantly larger gaps compared to other groups on the gingival, occlusal, and pulpal walls. 2. All groups except group 5 had no statistically significant gap on the gingival, occlusal, and pulpal walls. 3. There was no significant correlation between the amount of enamel on the gingival and occlusal walls and polymerization shrinkage.

  • Antimicrobial activity of Plantain(Plantago asiatica L.) was investigated. Methanol extract of dried Plantain was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, E. coli, and V. parahaemolyticus at 500 $\mu\textrm{g}$/disc. Ethylacetate fraction was further fractionated into 8 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 2 and 3 had the highest anti-microbial activity. They were mixed again, re-separated, and seven fractions were obtained. Among them, No.4 and 6 fraction had the highest inhibitory effect on the microorganisms, which were then separated into four fractions. In the 3rd fractionation, No.4 fraction was identified as hexadecanoic acid by HPLC, $^1$H-NMR and GC-MS.
