Immediate Decrease in γ-AminoButyric Acid after Caffeine Intake in Adolescents: a Preliminary MRS Study

  • In adolescents, sleep deprivation problem is getting worse, and increased caffeine consumption is considered to relieve the stress caused by sleep deprivation and academic burden. In this study, immediate neurologic effects of caffeine intake on adolescents were evaluated in three high school students using the ${\gamma}-aminobutyric$ acid (GABA)/creatine ratio on magnetic resonance spectroscopy (MRS). MEGA-PRESS MRS and TE 135 ms single voxel MRS were performed in the anterior cingulate cortex before and after drinking a cup of coffee, which contained 104 mg of caffeine. GABA and creatine were measured on LCModel 6.3, respectively. In all three students, GABA/creatine ratios were decreased after caffeine intake. The GABA/creatine ratios obtained before caffeine intake were decreased after caffeine intake in all the three adolescents. In this preliminary study, caffeine intake caused an immediate decrease in the GABA/creatine ratio in the brain and it may be related to the neurologic effects of caffeine on an adolescent's brain.

(γ-Aminobutyric acid를 생산하는 Lactobacillus brevis AML15의 분리 및 특성 (Isolation and Characterization of Lactobacillus brevis AML15 Producing γ-Aminobutyric acid)

  • 국내해안의 젓갈과 김치류로부터 86종의 GABA 생산균주를 분리하였다. 분리된 균주들을 Thin layer chromatography를 이용하여 GABA 생성능이 우수한 AML15, AML45-1, AML72의 3종의 균주를 선발하였다. 선별된 3종의 균주의 아미노산 분석 결과 GABA 생성능이 가장 우수한 AML15 균주를 본 실험에 사용하였다. AML15의 분류학적 위치를 규명하기 위하여 16S ribosomal DNA 영역의 부분염기서열 분석을 실시하였다. 165 rDNA 분석결과 Lactobacillus brevis ATCC 367과 99%의 유사도를 나타내어 L. brevis AML15로 명명하였다. MRS 배지에 최종 전환 농도로 설정된 5%(w/v) monosodium glutamic acid를 첨가하고 배지의 초기 pH를 4.0, 5.0과 6.0으로 조정하여 배양한 결과 배지의 초기 pH가 5.0일 때 GABA 생성능이 가장 높게 조사되었다. GABA 생산배지에 GAD 효소활성에 조효소로 작용하는 PLP를 0. 10. 50과 100 ${\mu}M$의 농도로 첨가하여 아미노산 분석결과 PLP를 10${\mu}M$ 첨가하였을 때 10,424 $nM/{\mu}$l의 GABA가생산되었다. PLP를 첨가하지 않았을 때보다 PLP 첨가 후 GABA 생성이 증가됨을 확인할 수 있었다.

팽이버섯(Flammulina velutipes) 분말의 젖산발효를 통한 고농도 γ-aminobutyric acid 함유 천연 발효조미료 개발 (Development of natural fermented seasoning with Flammulina velutipes powder fortified with γ-aminobutyric acid (GABA) by lactic acid fermentation)

  • 팽이버섯 분말을 이용하여 기능성물질인 GABA 및 probiotics를 강화시킨 천연 발효조미료를 개발하기 위해서 L. plantarum EJ2014에 의한 젖산발효 최적화 연구를 수행하였다. 팽이버섯 분말에 영양성분 0.5% YE, 1% glucose, GABA 전구물질인 5% MSG를 첨가한 후 $30^{\circ}C$에서 5일간 젖산발효를 진행한 결과 발효 2일 동안 pH는 6.1에서 4.4로 감소하다가 발효 5일에 6.2로 다시 증가하는 경향을 보였으며 산도는 발효 2일 동안 0.5%에서 1.3%로 증가한 후 발효 5일에 다시 0.4%로 감소하는 경향을 보였다. 생균수는 초기 젖산균 스타터 $2.4{\times}10^7CFU/mL$에서 발효 1일째 $2.2{\times}10^9CFU/mL$로 증가한 후 5일 동안 계속 유지되었다. 전구물질인 MSG는 발효 4일에 대부분이 이용되면서 약 2.31% 농도의 GABA로 전환되었으며 DPPH radical 소거 활성은 $IC_{50}$ 값이 1.24 mg/mL로, ABTS radical 소거 활성에서 $IC_{50}$ 값은 1.53 mg/mL로 나타나면서, 발효물의 항산화 효과가 증진되는 것으로 나타났다. 또한 팽이버섯 발효물 30 g에 볶은 밀기울 1 g을 첨가하여 열풍 건조한 천연 발효조미료의 GABA 함량은 17%로 고농도의 GABA를 함유하여 probiotic 기능성이 강화되며 기호성을 갖는 발효조미료 및 건강소재의 제조가 가능하였다.

녹각 추출액의 젖산발효를 통한 고농도 감마-아미노부티르산 생산 최적화 (Enrichment of gamma-aminobutyric acid (GABA) in old antler extract fermented by Lactobacillus plantarum)

  • 본 연구에서는 녹각 추출물을 이용하여 GABA 생산 최적화를 위해 추출액의 농도, 질소원인 YE, 탄소원 포도당, GABA 생성전구물질 MSG 농도에 따른 최적조건으로 YE 0.5%, 포도당 1.5%, MSG 3.5%를 첨가하여 $30^{\circ}C$에서 7일간 젖산발효를 하였다. 발효 7일 pH 6.56, 산도 0.77%로 나타났으며 $2.0{\times}10^8CFU/mL$로 높은 균수를 유지하였고 GABA를 1.4% 생성하는 것으로 나타났다. 결론적으로 녹각 추출액의 젖산균을 이용한 정치배양을 통해 고농도의 GABA 생산이 가능하였으며, 발효물은 GABA, 프로바이오틱(probiotic) 등을 함유하여 식품 및 기능성 소재로 이용가능성이 높다고 사료되었다.

Effect of Anaerobic Treatment on Carbohydrate-Hydrolytic Enzyme Activities and Free Amino Acid Contents in Barley Malt

  • Effects of acute anoxia on carbohydrate hydrolytic enzyme activities and free amino acid contents in malt were examined. Malts were prepared with barley grains germinated for 7 days which contained the highest levels of amylolytic and(1-3,1-4)-$\beta$-glucanase activities. $\alpha$-Amylase and $\beta$-amylase activities in malts were not significantly affected by anoxia for 5 or 10 h.(1-3,1-4)-$\beta$-Glucanase activity, however, decreased about 7 to 10% by anoxia for 5 or 10 h. Alanine and $\gamma$-aminobutyric acid content changed drastically. Alanine contents in malts increased by 2.2- and 2-fold, and $\gamma$-aminobutyric acid contents by 1.4- and 1.9-fold under anoxia for 5 and 10 h, respectively.

  • $\gamma$-aminobutyric acid (GABA) is a ubiquitous nonprotein amino acid that is produced primarily by $\alpha$-decarboxylation of L-glutamic acid (Glu) catalyzed by the enzyme glutamate decarboxylase (GAD). It is well known as a neurotransmitter that regulates inhibitory neurotransmission in the mammalian central nervous system. In addition, GABA has been proved to be effective for lowering blood pressure in mammals. This paper is intended to provide basic information about GABA, including the functional and biological activity of GABA, GABA production by lactic acid bacteria, and the utilization of GABA in the production of dairy products.

Effects of γ-aminobutyric acid (GABA) on growth performance, nutrient digestibility, blood profiles, meat quality, and backfat in finishing pigs

  • This experiment was conducted to estimate the influence of ${\gamma}$-aminobutyric acid (GABA) on growth performance, nutrient digestibility, blood profiles, meat quality, and backfat in finishing pigs. A total of 100 mixed finisher pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] were randomly allotted to two dietary treatments for 10 weeks, the average initial body weight was $52.03{\pm}1.08kg$. Dietary treatments consisted of supplementation of GABA to 0.05% of the basal diet (GAB) and a CON (basal diet) without supplementation. Each treatment had 10 replicates with 5 pigs per pen (three gilts and two barrows). Measurements were made at three time period: weeks 1 - 5, weeks 5 - 10 and overall experimental period. Compared to CON, the pigs fed GABA had average daily gain (ADG) increases among phases (p < 0.05). Average Daily Feed Intake ADFI increased during the overall experimental period (p < 0.05). The digestibility of Dry Matter (DM) increased. The concentration of epinephrine decreased at 5th and 10th weeks (p < 0.05), meanwhile, cortisol decreased at 10th week (p < 0.05) in GAB treatment. Otherwise, the pigs' GABA supplementation had no effect on meat quality and backfat thickness (p > 0.05). We conclude that a dietary supplementation of GABA at 0.05% improves growth performance and the digestibility of dry matter, while reducing the stress response of finishing pigs.

청신화담전 투여가 경련방생시 뇌조직 중 경련 및 산화 관련물질에 미치는 영향 (Anticonvulsant, Antioxidant Effect of Cheongsinhwadam-Jeon(청신화담전))

  • Objective and Methods : This study was performed to evaluate the anticonvulsant, antioxidant effect of modified formulas Korean traditional medicine Cheongsinhwadam-Jeon(CSHDJ). The extract of CSHDJ was administered (p.o.) to mice for 14 days in anticonvulsant and antioxidant tests. Results : The pretreatment of CSHDJ extract prohibited pentylenetrazol (PTZ)-induced convulsion. In PTZ-induced convulsion, lowered level of brain ${\gamma}$-aminobutyric acid(GABA) was restored normal state by the pretreatment of CSHDJ. Increased level of brain glutamic acid was lowered to normal state by CSHDJ, and increased activity of brain ${\gamma}-aminobutyric$ acid transaminase(GABA-T) was reduced to normal state by CSHDJ. In PTZ-induced convulsion, increased level of brain lipid peroxide was lowered to normal state by the pretreatment of CSHDJ. Increased activity of brain xanthine oxidase(XOD) was lowered by CSHDJ, and increased activity of brain aldehyde oxidase lowered to normal state by CSHDJ. In PTZ-induced convulsion, increased activities of superoxide dismutase(SOD) and catalase in brain were lowered by the pretreatment of CSHDJ, whereas increased level of glutathione and increased activity of gluthathione peroxidase in brain were not changed significantly. Conclusions : Above results suggest that CSHDJ has anticonvulsant, antioxidant effect. That seems to be strongly related with the levels of GABA, glutamate, lipid peroxide and the activities of GABA-T, XOD, aldehyde oxidase, SOD, catalase in brain tissue. From these results, CSHDJ could be applied to various convulsive disorders.

Rutin 및 GABA 첨식이 누에 성장에 미치는 영향 (Effect of Mulberry Leaf Sprayed with Rutin and GABA on Silkworm Growth)

  • Flavonol glycoside의 일종인 Rutin과 free amino acid의 일종인 GABA(${\gamma}$-aminobutyric acid)가 누에 성장에 어떤 영향을 미치는가를 알기 위해 뽕잎에 각각 0.1%의 수용액을 분무하여 3령 기잠부터 상족까지 누에를 사육한 결과 다음과 같았다. 1. rutin과 GABA(0.1%를 분무한 뽕잎의 rutin과 GABA 함량은 대조구 86mg/100g FW, 16mg/100g FW에 비해 2, 2.8배 높았다. 2. rutin 및 GABA를 첨식한 뽕잎으로 사육한 누에의 잠체중은 대조구에 비해 최구 25%까지 감소하였으며 5령 5일에 대조구 대비 각각 7%, 15%의 감소를 보여 rutin보다는 GABA가 잠체중 증가에 더 큰 악영향을 주는 것으로 나타났다. 3. 처리간에 식하량, 배분량, 소화량 등은 거의 차를 보이지 않았으나 rutin 처리구에서 소화량이 대조구 대비 6% 정도 높은 것으로 나타났다. 4. rutin 및 GABA 처리는 유충의 경과일수를 연장시켜 상족의 개시가 대조구에 비해 3일 정도 늦었으며, 상족기간도 대조구에 비해 1일이 길었으며, 상족마리수도 대조구 대비 각각 15.4%, 9.1% 떨어지는 것으로 나타났다.

Effects of poly-$\gamma$-glutamic acid on serum and brain concentrations of glutamate and GABA in diet-induced obese rats

  • Poly-gamma-glutamic acid ($\gamma$-PGA) is a mucilaginous and biodegradable compound produced by Bacillus subtilis from fermented soybeans, and is found in the traditional Korean soy product, cheongkukjang. This study was carried out to evaluate the effects of $\gamma$-PGA from a food source on the concentration of the neurotransmitter GABA and its metabolic precursor glutamate in diet-induced obese rats. Eight-week old male Sprague-Dawley rats (n=60) were used. The rats were divided into two groups and obesity was induced by providing either a 10% control fat or 45% high fat diet for 5 weeks. The rats were then blocked into 6 groups and supplemented with a 0.1% $\gamma$-PGA diet for 4 weeks. After sacrifice, brain and serum GABA and glutamate concentrations were analyzed by high performance liquid chromatography with fluorometric detection. The rats fed the high fat diet had significantly increased body weights. $\gamma$-PGA supplementation significantly increased serum concentrations of glutamate and GABA in the control fat diet groups while this effect was not found in the high fat groups. In the brain, glutamate concentrations were significantly higher in the $\gamma$-PGA supplemented groups both in rats fed the normal and high fat diets than in the no $\gamma$-PGA controls. GABA concentrations showed the same tendency. The results indicated that $\gamma$-PGA intake increased GABA concentrations in the serum and brain. However, the effects were not shown in obese rats.
