Cognitive improvement effects of Momordica charantia in amyloid beta-induced Alzheimer's disease mouse model (여주의 amyloid beta 유도 알츠하이머질환 동물 모델에서 인지능력 개선 효과)

  • Accumulation of amyloid beta (Aβ) and oxidative stress are the most common reason of Alzheimer's disease (AD). In the present study, we investigated the cognitive improvement effects of butanol (BuOH) fraction from Momordica charantia in Aβ25-35-induced AD mouse model. To develop an AD mouse model, mice were received injection of Aβ25-35, and then orally administered BuOH fraction from M. charantia at doses of 100 and 200 mg/kg/day during 14 days. In the T-maze and novel object recognition test, administration of BuOH fraction from M. charantia L. at doses of 100 and 200 mg/kg/day improved spatial ability and novel object recognition by increased explorations of novel route and new object. In addition, BuOH fraction of M. charantia-administered groups improved learning and memory abilities by decreased time to reach hidden platform in Morris water maze test. Oral administration of BuOH fraction from M. charantia significantly inhibited lipid peroxidation and nitric oxide levels in the brain, liver, and kidney compared with Aβ25-35-induced control group. These results indicated that BuOH fraction of M. charantia improved Aβ25-35-induced cognitive impairment by attenuating oxidative stress. Therefore, M. charantia could be useful for protection from Aβ25-35-induced cognitive impairment.

  • Some phytoestrogens in soy and red wine, for example, might have beneficiary rather than adverse effects. In particular, dietary soy intake seems to be highly correlated with protection of breast cancer, osteoporosis and cardiovascular disorders. However, questions persist on the potential adverse effects of the main soy constituent genistein (GS) on female reproductive physiology. Previously we found that prepubertal exposure to GS could activate the reproductive system of immature female rats leading to precocious puberty onset, and intracerebroventricularly (ICV) injected GS could directly activate hypothalamic kisspeptin-GnRH neuronal circuits in adult female rats. The present study was performed to examine the hypothalamus-specific GS effects in prepubertal female rats and which subtype of estrogen receptor is mediated in this GS effect. Prepubertal female rats (PND 30) were anaesthetized, treated with single dose of GS (3.4 ${\mu}g$/animal), and sacrificed at 2 hrs post-injection. To determine the transcriptional changes of reproductive hormone-related genes in hypothalamus, total RNAs were extracted and applied to the semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). ICV infusion of GS significantly lowered the transcriptional activities of mTOR (1:$0.361{\pm}0.058$ AU, p<0.001) but increased that of GAD67 (1:$1.285{\pm}0.099$ AU, p<0.05), which are known to act as an upstream modulator of kisspeptin and GnRH neuronal activities in the hypothalamus, respectively. GS administration enhanced significantly the mRNA levels of KiSS-1(1:$1.458{\pm}0.078$ AU, p<0.001), and exerted no effect on the mRNA level of kisspeptin receptor GPR-54 (1:$1.29{\pm}0.08$ AU). GnRH gene expression was significantly decreased in GS-treated group compared to control group (1:$0.379{\pm}0.196$ AU, p<0.05). There was no difference in the mRNA level of $ER{\alpha}$ in the GS-treated group compare to control group (1:$1.180{\pm}0.390$ AU, Fig. 3A). However, icv infusion of GS significantly increased the transcriptional activities of $ER{\beta}$ (1:$4.209{\pm}0.796$ AU, p<0.01, Fig. 3B). Taken together, the present study indicated that the acute exposure to GS could directly alter the hypothalamic GnRH modulating system in prepubertal female rats. Our study strongly suggested the involvement of $ER{\beta}$ pathway in GS's hypothalamus-specific action, and this idea is consistent with the GS's well-known $ER{\beta}$-mediated protective action in breast cancer.

  • In this study, we attempted to prepare compounds with heterocyclic replacements for metabolically unstable esters of benzopyranyl indole-2-carboxylic esters, which showed good in vitro and in vivo cardioprotective efficacies possibly through the opening of mitochondrial ATP-sensitive potassium channel ($K_{ATP}$). Initially, we tried to construct indolin-2-yl-heterocycles using unprotected indoline-2-carboxylic acid, but the cyclization was proceeded with oxidation of the indoline ring to the indole, which didn't react with benzopyranyl epoxide. Thus we introduced N-Boc group to deplete the electron density of the indoline ring. We successfully prepared various indolin-2-yl-heterocycles by the cyclization of the building blocks including carboxamide, ${\beta}$-hydroxy amide, hydrazide, nitrile starting from N-Boc-indoline-2-carboxylic acid.

  • In the problem of some sequential estimation, the stopping times may be written in the form $N(c) = inf{n \geq n_0; n \geq c^2 S^2_n/\delta^2 (\bar{X}_n)}$ where ${s^2_n}$ and ${\bar{X}_n}$ are the sequences of sample variance and sample mean of the independently and identically distributed (i.i.d.) random variables with distribution $F_{\theta}(x), \theta \in \Theta$, respectively, and $\delta$ is either constant or any given positive real valued function. We obtain some asymptotic normality and asymptotic expectation of the N(c) in various limiting situations. Specially, uniform asymptotic normality and uniform asymptotic expectation of the N(c) are given.

  • Alternaria from different Allium plants was characterized by multilocus sequence analysis. Based on sequences of the ${\beta}$-tubulin (BT2b), the Alternaria allergen a1 (Alt a1), and the RNA polymerase II second largest subunit (RPB2) genes and phylogenetic data analysis, isolates were divided into two groups. The two groups were identical to representative isolates of A. porri (EGS48-147) and A. vanuatuensis (EGS45-018). The conidial characteristics and pathogenicity of A. vanuatuensis also well supported the molecular characteristics. This is the first record of A. vanuatuensis E. G. Simmons & C. F. Hill from Korea and China.

  • Twenty Penicillium isolates were recovered during the investigation of fungal community in the soil samples collected from Wando (Jeonnam Province, Korea). Among them, one species was identified and described as P. cainii based on phylogentic analysis of internal transcribed spacer and ${\beta}$-tubulin (BT2) genes and morphological characteristics. This is a first report of P. cainii in Korea.

  • Regiospecific total syntheses of (±)-11-deoxy-4-methoxy-10-oxo-βrhodomycinone (21a) and (±)-11-deoxy-1-methoxy-10-oxo-β-rbodomycinone (21b) are described. 2-(2-Bromoethyl)-1,3-dioxane (6) was transformed to naphthalenone 12, which was condensed with (phenylsulfonyl)-isobenzofuranone 13 to afford 7,8-dihydro-9-ethyl-6-hydroxy-4-methoxynaphthacen-5,12-dione (15). Epoxide 16 prepared from olefinic compound 15, reacted with HF/Pyr (7:3) to give 17. Dihydroxylation of 17 with t-BuOK/P(OMe)3/O2, selective cis-diol protection of mixed compounds 18 with phenylboronic acid in toluene, separation of cis-boronate 19 and trans-diol 20 by column chromatography on silica gel, and cleavage of the boronate group of 19 with 2-methylpentane-2,4-diol in acetic acid completed the construction of 21.

  • trans-Metanicotine, a subtype (${\alpha}_4{\beta}_2$)-selective ligand for neuronal nicotinic acetylcholine receptor, is under clinical phase for Alzheimer's disease. An efficient synthetic route for ($\pm$)-methyl-(1-aryl-4-pyridin-3-yl-but-3-enyl)-am ices, derivatives of tracts-metanicotine, was explored. Allylation reaction of aryl aldimines with allylmagnesium bromide in THF gave ($\pm$)-methyl-(1-aryl-but-3-enyl)-amines. Protection of the amines with the Boc group and following Heck reaction of the N-Boc amines with 3-bromopyridine gave ($\pm$)-methyl-(1-aryl-4-pyridin-3-yl-but-3-enyl)-carbamic acid tert-butyl esters. Deprotection of the N-Boc group in aqueous 1 N-HCI solution gave the titled amines in good yields. Thus, trans-metanicotine analogues modified at the ${\alpha}-position$ of the methylamino group with amyl groups were obtained in 5 steps.

  • Biphenyl dimethyl dicarboxylate (DDB) is a by-product produced in process of synthesizing Schizandrin-C. Generally, DDB has known to protect hepatocytes and to decrease the index of liver enzyme (e.g. GOT and GPT) in chronic hepatitis. The present study was aimed to demonstrate whether DDB can protect the brain cell, especially the Alzheimer brain in vitro. A Convergence Study on the Factors Influencing Nursing Students' Attitudes toward Standard Precautions: Focusing on the Health Belief Model (간호대학생의 표준주의 태도에 미치는 영향 요인에 관한 융합적 연구 : 건강신념모델 기반)

  • "Standard Precautions" (SP) is important for nursing students as it will lead to the protection of health professionals from infectious diseases and prevention of the wide spread of epidemic illnesses. Therefore, this study investigated factors influencing nursing students' attitudes toward SP. Data were collected from 291 nursing students from May to June, in 2016. According to the results, perceived benefit (4.26) was the highest among sub factors of the health belief model, test score of SP was 78.8%, and attitude score (4 point scale) was 3.63. Perceived sensitivity, perceived benefits, perceived barriers, self-efficacy, knowledge about SP showed significant correlations with the attitudes towards SP. Among sub-factors of the health beliefs, perceived sensitivity (${\beta}=.152$, p<.05) and perceived barrier (${\beta}=-.125$, p<.05) were found to influence nursing students' attitudes toward SP (F= 5.680, p<.001). Hence, it can be concluded that a convergence education program promoting health belief is needed for improving attitudes toward SP among nursing students.
