Response of ${\beta}-Glucanases{\;}to{\;}GA_3$ in Barley Aleurone Layers (보리 호분층에서 $(1-3)-{\beta}-glucanase{\;}$${\;}(1-3,1-4)-{\beta}-glucanase$${\;}GA_3$에 대한 반응)

  • Isolated barley aleurone layers were used to examine response of (1-3)- and $(1-3,1-4)-{\beta}-glucanases{\;}to{\;}GA_3$. Protein content and levels of (1-3)- and $(1-3,1-4)-{\beta}-glucanases$ increased in the presense of added $GA_3$. However, (1-3)- and $(1-3,1-4)-{\beta}-glucanases$ showed different response to $GA_3$ in their production and secretion patterns. $(1-3,1-4)-{\beta}-glucanases$ showed higher increase in enzyme activity than $(1-3)-{\beta}-glucanase$ in the early stage of$GA_3$treatment. Secretion of enzyme by $GA_3$ into the surrounding medium was more enhanced in $(1-3,1-4)-{\beta}-glucanases$ than in $(1-3)-{\beta}-glucanase$, The differential response of the enzymes might be related to the physiological role of the enzymes in germination of barley grain.

Resistance to Chemicals and Water of Epoxy Resin Cured with Phosphazene Derivatives (포스파젠 유도체에 의해 경화된 에폭시수지의 내수.내약품성)

  • 1, 1-diamino-3, 3, 5, 5-tetrachlorocyclotriphosphazene, 1, 1-diami-no3, 3, 5, 5-tetra-(p-bromophenoxy)cyclotriphosphazene, and 1, 1-diamino-3, 3, 5, 5-tetra-(p-chlorophenoxy) cyclotri-phosphazene(ACPP) was prepared from hexachlorocyclotriphosphazene and used for the curing agent of bisphenol A type epoxy resin and phenol novolak. The resistance to chemicals and water of the cured epoxy resins were examined by DMTA and compared with those of the epoxy rosins cured with phosphazene derivatives and 4, 4'-diaminodiphenylmethane. The effect of the curing agent on resistance to chemicals and water of the cured epoxy resins were investigated. The epoxy resins cured with 1, 1-diamino-3, 3, 5, 5-tetrachlorocyclotriphosphazene and 1, 1- diamino-3, 3, 5, 5-tetra- (p-bromophenoxy)cyclotriphosphazene showed superior resistance to chemicals and water to those of 1, 1-diamino-3, 3, 5, 5-tetra- (p-chlorophenoxy)cyclotriphosphazene and 4, 4'-diaminodiphenylmethane. It is an effective curing agent for epoxy resins to enhance the resistance to chemicals, water and tome proofing.

Hydrolysis Mechanisms of Fish Oil by Lipolase-100T

  • In order to investigate the position of various fatty acids attached to glycerol and the specificity of Lipolase-100T, hydrolysis of fish oil was carried out with Lipolase-100T derived from Aspergillus oryzae. The amounts of free fatty acids produced from triglyceride, 1,2(2,3)-diglyceride, 1,3-diglyceride, and 2-monoglyceride and conversion rates of 1,2(2,3)-diglyceride to 1,3-diglyceride and 2-monoglyceride to 1(3)-monoglyceride were also calculated. The ratio of 1,2-diglyceride content to 1,3-diglyceride was higher than 70 in the early period of hydrolysis. The fatty acid content of the glyceride mixture after 72 h of hydrolysis was compared with that of fish oil, and it was found that polyunsaturated fatty acids such as C16:4, C20:4 n-3, C20:5 n-3, C21:5 n-3, C22:5 n-3 and C22:6 n-3 were located in the 2-position of glycerol. Material balance of each component in the hydrolysis system was written to obtain a set of simultaneous linear equations. The theoretical quantity of free fatty acids produced from triglyceride, 1,2-diglyceride, 1,3-diglyceride, and monoglyceride, respectively, were calculated by solving the linear equation system. The conversion rate of 1,2(2,3)-diglyceride to 1,3-diglyceride and that of 2-monoglyceride to 1(3)-monoglyceride were also obtained. The results showed that the migration rate of 1,2(2,3)-diglyceride to 1,3-diglyceride was higher than the hydrolysis rate of 1,2(2,3)-diglyceride to 2-monoglyceride and the conversion rate of 2-monoglyceride to 1(3)-monoglyceride was extremely low.

  • Zinc chlorin 1 possessing tertiary 3$^1$_hydroxy and 13$^1$-oxo groups was synthesized as a model for the antenna chlorophylls of photosynthetic green bacteria. Self-aggregation of 1 in nonpolar organic solvents was examined and compared to 2 and 3 possessing a secondary and primary 3$^1$_hydroxy group, respectively. Zinc chlorin 1 self-aggregated in I%(v/v) CH$_2$Cl$_2$-hexane to form oligomers and showed a red-shifted Qy maximum at 704 nm compared to the monomer (648 nm in CH$_2$CI2$_2$). This red-shift is larger than that of 3$^1$S-2 (648 to 697 nm) and comparable to that of3$^1$R-2 (648 to 705 nm), but smaller than that of 1 (648 to 740 nm), indicating that while a single 3$^1$-methyl group (primary to secondary OH) suppressed tight and/or extended aggregation, the additional 3$^1$-methyl group (secondary to tertiary OH) did not further suppress aggregation. The relative stability of the aggregates was in the order 3> 3$^1$R-2∼ 1 > 3$^1$S-2 as determined by visible spectral analyses. Molecular modeling calculations on oligomers of zinc chlorins 1, 3$^1$ R-2 and 3 gave similar well-ordered energy-minimized structures, while 3 stacked more tightly than 3$^1$ R- 2 and 1. In contrast, 3$^1$S-2 gave a relatively disordered (twisted) structure. The calculated oligomeric structures could explain the visible spectral data of 1-3 in nonpolar organic solvents. Moreover, self- aggregation of synthetic zinc 13$^1$_oxo-hlorins 4-6 possessing a 2-hydroxyethyl, 3-hydroxypropyl and 3- hydroxy-I-propenyl group at the 3-position in nonpolar organic solvents was discussed.

A Novel Complement Fixation Pathway Initiated by SIGN-R1 Interacting with C1q in Innate Immunity

  • Serum complement proteins comprise an important system that is responsible for several innate and adaptive immune defence mechanisms. There were three well described pathways known to lead to the generation of a C3 convertase, which catalyses the proteolysis of complement component C3, and leads to the formation of C3 opsonins (C3b, iC3b and C3d) that fix to bacteria. A pivotal step in the complement pathway is the assembly of a C3 convertase, which digests the C3 complement component to form microbial-binding C3 fragments recognized by leukocytes. The spleen clears microorganisms from the blood. Individuals lacking this organ are more susceptible to Streptococcus pneumoniae. Innate resistance to S. pneumoniae has previously been shown to involve complement components C3 and C4, however this resistance has only a partial requirement for mediators of these three pathways, such as immunoglobulin, factor B and mannose-binding lectin. Therefore it was likely that spleen and complement system provide resistance against blood-borne S. pneumoniae infection through unknown mechanism. To better understand the mechanisms involved, we studied Specific intracellular adhesion molecule-grabbing nonintegrin (SIGN)-R1. SIGN-R1, is a C-type lectin that is expressed at high levels by spleen marginal-zone macrophages and lymph-node macrophages. SIGN-R1 has previously been shown to be the main receptor for bacterial dextrans, as well as for the capsular pneumococcal polysaccharide (CPS) of S. pneumoniae. We examined the specific role of this receptor in the activation of complement. Using a monoclonal antibody that selectively downregulates SIGN-R1 expression in vivo, we show that in response to S. pneumoniae or CPS, SIGN-R1 mediates the immediate proteolysis of C3 and fixation of C3 opsonins to S. pneumoniae or to marginal-zone macrophages that had taken up CPS. These data indicate that SIGN-R1 is largely responsible for the rapid C3 convertase formation induced by S. pneumoniae in the spleen of mice. Also, we found that SIGN-R1 directly binds C1q and that C3 fixation by SIGN-R1 requires C1q and C4 but not factor B or immunoglobulin. Traditionally C3 convertase can be formed by the classical C1q- and immunoglobulin-dependent pathway, the alternative factor-B-dependent pathway and the soluble mannose-binding lectin pathway. Furthermore Conditional SIGN-R1 knockout mice developed deficits in C3 catabolism when given S. pneumoniae or its capsular polysaccharide intravenously. There were marked reductions in proteolysis of serum C3, deposition of C3 on organisms within SIGN-$R1^+$ spleen macrophages, and formation of C3 ligands. The transmembrane lectin SIGN-R1 therefore contributes to innate resistance by an unusual C3 activation pathway. We propose that in the SIGN-R1 mediated complement activation pathway, after binding to polysaccharide, SIGN-R1 captures C1q. SIGN-R1 can then, in association with several other complement proteins including C4, lead to the formation of a C3 convertase and fixation of C3. Therefore, this new pathway for C3 fixation by SIGN-R1, which is unusual as it is a classical C1q-dependent pathway that does not require immuno globulin, contributes to innate immune resistance to certain encapsulated microorganisms.

Comparison of Yield in Aiternating Crop System Mixed of Tillering Hybrid Corn and Soybean (분얼형(分蘖型) 옥수수 교잡종(交雜種)과 대두(大豆)의 교호작(交互作)에 대한 수량(收量) 비교(比較) 시험(試驗))

  • This study was conducted to determine the effects of alternating planting systems of soybean and tillering hybrid corn on yield of both crops. The planting systems included monocultures of both crops. 1 to 1 row ratio, 1 to 2 row ratio, 1 to 3 row ratio, 2 to 1 row ratio, 2 to 3 row ratio, 3 to 1 row ratio, 3 to 2 row ratio and 3 to 3 row ratio of soybean and corn. The results obtained are summarized as follows : The plant height of soybean was increased as the number of planting rows of corn increased. But the plant, height of soybean was quite stabilized when the ratio of soybean planting rows increased. When the plant height of soybean was increased due to the increased row number of corn, the branch number and diameter of soybean decreased and lodging ratio increased. Total fresh weight of soybean per 10a was decreased as the ratio of soybean planting decreased and it was increased when the ratio of soybean planting increased over monoculture of soybean. Dry weight of soybean per 10a showed the same tendency of the fresh weight. But the highest grain yield of soybean was obtained in the monoculture of soybean and it was 276 kg/10a. Comparatively high yield of soybean was obtained in the systems of 2 to 1 and 3 to 3 ratios. The plant height, number of tillers and number of ears per plant of tillering corn were greatly affected by the alternating planting of soybean and they were 208.9cm, 2.1 and 4 per plant, respectively. The fresh and dry weights and yield of corn per 10a showed significant differences among systems. The average fresh and dry weight of corn in the systems of 3 to 1 and 2 to 1 were 1000 kg and 100 kg higher than those of monoculture of corn. The kernel yields of corn per 10a were 438 kg in the monoculture system and 530 kg in the systems of 3 to 2 and 2 to 1. The total fresh weights of corn and soybean in systems of 3 to 1, 3 to 2 and 2 to 1 were higher than 5721 kg of soybean and 4358 kg of corn in the monoculture. Combined dry weight was high in the order of monoculture, 2 to 1 and 3 to 1 row ratios. Combined grain yield was high in the systems of 2 to 1 and 3 to 1 row ratios and it was over 430 kg per 10a.

Hormonal Regulation of Leptin, Resistin, and Plasminogen Activator Inhibitor-1 Gene Expression in 3T3-L1 Adipocytes

  • Leptin, resisitn and PAI-1 (plasminogen activator inhibitor-1) are synthesized and secreted by rodent fat cells and recently postulated to be an important link to obesity. This study was conducted to characterize the hormonal regulation of leptin, resistin, and PAI-1 gene expression in the 3T3-L1 adipocytes. The cells were treated with 0.5 $\mu$M insulin, 1 $\mu$M dexamethasone (Dex), or 0.05 $\mu$M triiodothyronine (T3) for 72 hours. The mRNA levels of each peptide were measured by semi-quantitative RT-PCR. The mRNA level of the leptin-producing ob gene was significantly increased by insulin, Dex, and T3 by 3.2-, 3.1- and 2.7-fold, respectively, compared to the control (p < 0.05). The level of resistin mRNA was increased by insulin, Dex, and T3 by 2.7-, 2.5- and 2-fold, respectively, compared to the control (p < 0.05). Likewise, the level of PAI-1 mRNA was significantly increased by insulin, Dex, and T3 compared to the control (p < 0.05). Taken together, our results suggest that insulin, Dex, and T3 may regulate the gene expression of leptin, resistin, and PAI-1 in 3T3-L1 adipocytes.

  • Reaction of propane-1, 3-sultone with amines gave N-substituted aminosulphonnic acids 2a-i, Dehydration of 2a-c with $POCI_3$ gave the corresponding sultams 3a-c. Propane-1, 3-sultone 1 reacted with tertury amines to give the betaiene salts 4-11. 2-4-Dimethyl-1, 3-butadiene-1, 4-sultone 12 condensed with amines to give N-substituted-2, 4-dimethyl-1, 3-butadiene-1, 4-sultames 13a and 13b. The reaction of 3a, 13a with hydrazine hydrate gave acid hydrazides 3d or 13c. Compounds 3d, 13c reacted with isocyanates to yield urea derivatives 14a-c, 15a-c.

Fabrication of Stack-Structured Gas Sensor of LaCrxCo1-xO3/Li1.3Al0.3Ti1.7(PO4)3 and Its NOx Sensing Properties (LaCrxCo1-xO3/Li1.3Al0.3Ti1.7(PO4)3의 적층구조를 가지는 가스센서 제조와 그의 NOx 검지특성)

  • Impedancemetric $NO_x$ (NO and $NO_2$) gas sensors were designed with a stacked-layer structure and fabricated using $LaCr_xCo_{1-x}O_3$ (x = 0, 0.2, 0.5, 0.8 and 1) as the receptor material and $Li_{1.3}Al_{0.3}Ti_{1.7}(PO_4)_3$ plates as the solid-electrolyte transducer material. The $LaCr_xCo_{1-x}O_3$ layers were prepared with a polymeric precursor method that used ethylene glycol as the solvent, acetyl acetone as the chelating agent, and polyvinylpyrrolidone as the polymer additive. The effects of the Co concentration on the structural, morphological, and $NO_x$ sensing properties of the $LaCr_xCo_{1-x}O_3$ powders were investigated with powder X-ray diffraction, field emission scanning electron microscopy, and its response to 20~250 ppm of $NO_x$ at $400^{\circ}C$ (for 1 kHz and 0.5 V), respectively. When the as-prepared precursors were calcined at $700^{\circ}C$, only a single phase was detected, which corresponded to a perovskite-type structure. The XRD results showed that as the Co concentration of the $LaCr_xCo_{1-x}O_3$powders increased, the crystal structure was transformed from an orthorhombic phase to a rhombohedral phase. Moreover, the $LaCr_xCo_{1-x}O_3$ powders with $0{\leq}x<0.8$ had a rhombohedral symmetry. The size of the particles in the $LaCr_xCo_{1-x}O_3$powders increased from 0.1 to $0.5{\mu}m$ as the Co concentration increased. The sensing performance of the stack-structured $LaCr_xCo_{1-x}O_3/Li_{1.3}Al_{0.3}Ti_{1.7}(PO_4)_3$ sensors was found to divide the impedance component between the resistance and capacitance. The response of these sensors to NO gas was more sensitive than that to $NO_2$ gas. Compared to other impedancemetric sensors, the $LaCr_{0.8}Co_{0.2}O_3/Li_{1.3}Al_{0.3}Ti_{1.7}(PO_4)_3$ sensor exhibited good reversibility and reliable sensingresponse properties for $NO_x$ gases.

Inhibitory Effect of Dihydroartemisinin, An Active Ingredient of Artemisia annua, on Lipid Accumulation in Differentiating 3T3-L1 Preadipocytes

  • Objectives: Artemisinin and its derivatives extracted from Artemisia annua, a Chinese herbal medicine, have variable biological effects due to structural differences. Up to date, the anti-obesity effect of dihydroartemisinin (DHA), a derivative of artemisinin, is unknown. The purpose of this study was to investigate the anti-adipogenic and lipolytic effects of DHA on 3T3-L1 preadipocytes. Methods: Oil Red O staining and AdipoRed assay were used to measure lipid accumulation and triglyceride (TG) content in 3T3-L1 cells, respectively. Cell count analysis was used to determine the cytotoxicity of 3T3-L1 cells. Western blot and real-time reverse transcription polymerase chain reaction analyses were used to analyze the expression of protein and mRNA in 3T3-L1 cells, respectively. Results: DHA at 5 μM markedly inhibited lipid accumulation and reduced TG content in differentiating 3T3-L1 cells with no cytotoxicity. Furthermore, DHA at 5 μM inhibited the expression of CCAAT/enhancer-binding protein-α (C/EBP-α), peroxisome proliferator-activated receptor-γ (PPAR-γ), fatty acid synthase (FAS), and perilipin A as well as the phosphorylation of signal transducer and activator of transcription-3 (STAT-3) in differentiating 3T3-L1 cells. Moreover, while DHA at 5 μM had no effect on the mRNA expression of adiponectin, it strongly suppressed that of leptin in differentiating 3T3-L1 cells. However, DHA at 5 μM had no lipolytic effect on differentiated 3T3-L1 cells, as assessed by no enhancement of glycerol release. Conclusions: These results demonstrate that DHA at 5 μM has a strong anti-adipogenic effect on differentiating 3T3-L1 cells through the reduced expression and phosphorylation of C/EBP-α, PPAR-γ, FAS, perilipin A, and STAT-3.
