Nam and Kim #1 Tissue-specific expression of pFV4CAT in transgenic mud loach (Misgurnus mizolepis) germ line (미꾸라지(Misgurnus mizolepis)에서 pFV4CAT 의 조직 특이적 발현)

  • The transgene, pFV4CAT, containing CAT reporter gene regulated by carp $\beta$-actin promoter, was expressed in independent transgenic mud loach germ lines, determined by reverse transcriptase-PCR (RT-PCR) and enzyme-linked immunosorbant assay (ELISA). Expression of the transmitted transgene was found to be tissue-specific in F1 and F2 generations. Tissue specificity of the expression was dependent on each transgenic line with reproducible patterns. Liver and spleen did express the transgene more frequently than other tissues tested, and muscle and heart revealed the higher amount of CAT than other tissues, while testes showed the lowest expression level. The highest level of CAT expression in muscle from a transgenic F1 line was corresponding to 68-fold compared to the basal levels of controls.

Partial Characterization of Two Cathepsin D Family Aspartic Peptidases of Clonorchis sinensis

  • Cathepsin D (CatD, EC 3.4.23.5) is a member belonging to the subfamily of aspartic endopeptidases, which are classified into the MEROPS clan AA, family A1. Helminth parasites express a large set of different peptidases that play pivotal roles in parasite biology and pathophysiology. However, CatD is less well known than the other classes of peptidases in terms of biochemical properties and biological functions. In this study, we identified 2 novel CatDs (CsCatD1 and CsCatD2) of Clonorchis sinensis and partially characterized their properties. Both CsCatDs represent typical enzymes sharing amino acid residues and motifs that are tightly conserved in the CatD superfamily of proteins. Both CsCatDs showed similar patterns of expression in different developmental stages of C. sinensis, but CsCatD2 was also expressed in metacercariae. CsCatD2 was mainly expressed in the intestines and eggs of C. sinensis. Sera obtained from rats experimentally infected with C. sinensis reacted with recombinant CsCatD2 beginning 2 weeks after infection and the antibody titers were gradually increased by maturation of the parasite. Structural analysis of CsCatD2 revealed a bilobed enzyme structure consisting of 2 antiparallel β-sheet domains packed against each other forming a homodimeric structure. These results suggested a plausible biological role of CsCatD2 in the nutrition and reproduction of parasite and its potential utility as a serodiagnostic antigen in clonorchiasis.

  • The vehicle handling performances have a close relation with the driver's safety. And the CAT(Computer aided testing) system has been introduced as a powerful tool for improving the handling performances. In this study, a CAT system for effectively measuring and analysing the handling performances is developed based on the windows GUI(Graphic user interface) environment. Especially, in order to derive a lot of significant handling performance parameters from a series of proving ground tests, the CAT system has various functions related to real time measurement, time domain analysis, frequency domain analysis, steady state analysis and so on.

Feline Gastrointestinal Eosinophilic Sclerosing Fibroplasia in a Bengal Cat

  • A 2-year-old neutered male Bengal cat presented with a 6-month history of weight loss and chronic vomiting. An abdominal ultrasound revealed increased thickness of the pylorus and ascending duodenum with concurrent enlargement of the mesenteric lymph nodes. Histologically, branching and anastomosing trabecular pattern of dense collagen was the characteristic feature, and large populations of spindle cells were also observed. These large spindle-shaped cells were positive for smooth muscle actin and vimentin on immunohistochemical examination. Based on these findings, the cat was diagnosed with feline gastrointestinal eosinophilic slcerosing fibroplasia (FGESF), and immunosuppressive therapy was initiated immediately. Unfortunately, the cat's condition deteriorated despite treatment; the cat died 56 days after initiation of therapy. This is the first report of FGESF in South Korea.

  • The electrochemistry and valence tautomerism of CoⅢ(N-N)(SQ)(Cat), [N-N=N,N,N',N'-tetramethylethylenediamine (TMEDA); SQ=3,5- or 3,6-di-tert-butyl-semiquinone, Cat=3,5- or 3,6-di-tert-butyl-catechol], have been investigated by spectroscopic, electrochemical, spectroelectrochemical methods in nonaqueous solvents under anaerobic condition. The transition temperature between tautomers is dependent upon the donation effect of substituted quinone ligand and solvent. It increases with the increase of donation effect of solvent and quinone ligand. CoⅢ(TMEDA)(SQ)(Cat) is reduced to [CoⅡ(TMEDA)(SQ)(Cat)]-, and then reduces to [CoⅡ(TMEDA)(Cat)2]2-. CoⅢ(TMEDA)(SQ)(Cat) is oxidized to [CoⅢ(TMEDA)(SQ)2]+, but the stability of the oxidized form in DMF is dependent upon the solution temperature. With the increase of solution temperatrue the oxidized form may be converted to [CoⅡ(TMEDA)(SQ)(BQ)]+ by intramolecular electron transfer from SQ ligand to CoⅢ.

  • Real-time control communication network system is important for developing defense robots because it affects environmental interaction, performance, and safety. We propose a real-time control communication network using the Xenomai real-time operating system and the open-source EtherCAT master library, SOEM. EtherCAT is an Ethernet-based industrial communication method. It has low latency and many functions such as cable redundancy and distributed clock synchronization. We use Xenomai RTOS and Intel NUC to develop the system. Experimental tests demonstrate the Real-time EtherCAT master implementation, and communication with CiA301-based slave devices. The jitter measurement was conducted to validate the real-time performance of the system. The proposed system shows possibility for real-time robotics applications in various defense robots.

  • New technologies have turned theories of education engineering into practice and also made education more efficient. One of them is the theory of 'Computer Adaptive Testing(CAT)'. This study is aimed to design and develop a web-based teaming system by making up for the weak points in the existing computer adaptive testing(CAT). The characteristics of the system are as follows: Firstly, tests can be controlled to fit the teachers' purposes by putting in different levels of both questions and samples. Secondly, this system does not test the same number of questions as students with different levels of ability do on a paper test, but a test can be taken according to their level. Thus this system is able to correctly judge learners' ability in a short time.

Cutaneous asthenia (Ehlers-Danlos syndrome) in a Korean short-haired cat

  • A 1-year-old Korean domestic short-haired cat presented with skin hyperextensibility and a severely macerated wound on the skin of the dorsal part of the neck. Diagnostic studies including histopathology and skin extensibility index revealed congenital cutaneous asthenia (Ehlers-Danlos syndrome). In this cat, the skin wounds and defects were successfully managed with standard wound management and cosmetic surgery. Although skin hyperextensibility is persistent, the cat has lived well without other complications to date. To the best of our knowledge, this is the first report of cutaneous asthenia in a cat in Korea.
