Quality Characteristics and Antioxidant Activity of Sikhe prepared using Hot Water Extracts of Roasted Coffee Ground Residue

커피박 열수추출물로 제조한 식혜의 품질 특성 및 항산화 활성

Abstract

This study was performed to evaluate the quality characteristics and antioxidant activity of sikhe prepared using various concentrations of hot water extracts roasted coffee ground residue (CR-sikhe; 0.2, 0.4, 0.6, 0.8, and 1.0%). The pH increased with increasing CR concentration. The reducing sugar content after 5 h saccharification was the highest when 0.8% CR extract was used. Total polyphenol and flavonoid contents increased in a concentration-dependent manner reaching maximum levels when 1.0% CR-extract was used. The antioxidant activities of CR-extracts were higher than that of the control and increased dose-dependently. The CR-0.6 showed the best taste (4.28), color (4.56), flavor (4.08), and overall acceptability (4.28). After 10 day of storage at $4^{\circ}C$, the total cell count in CR-sikhe was approximately 1-2 log cycle, which was less than that in the control.

커피 음료 제조시 생성되는 커피박의 식품자원화에 대한 기초연구로써, 전통 식품인 식혜에 적용하기 위하여, 커피박 열수 추출물을 첨가한 식혜의 당화 과정 중 품질 변화와 항산화 활성 및 저장 중 미생물 변화를 조사하였다. CR 함유 식혜는 대조구에 비해 pH가 높았으며, 당화 시간이 경과함에 따라 모든 시료의 pH는 증가하였다. 당화 완료 시점인 5시간에는 대조구와 CR 첨가구의 pH는 유의적인 차이를 나타내지 않았다. 산도는 0.06-0.08%의 범위 내에서 변동은 있었으나, CR 농도별 및 당화 시간에 따른 뚜렷한 변화는 나타나지 않았다. 동일 당화 시간에서는 CR 농도가 높을수록 환원당의 함량은 높았고, 동일 시료에서는 당화 시간이 경과할수록 증가하였다. 당화 5시간에서는 CR-0.8과 CR-1.0의 환원당은 각각 48.66, 47.24 mg/mL를 나타내어 대조구(45.10 mg/mL)에 비해 유의적으로 높았다(p<0.05). 당화 시간이 경과할수록, CR 첨가 식혜는 대조구에 비해 어둡고, 적색도 및 황색도가 증가하였으며, CR의 농도가 높을수록 이러한 경향은 뚜렷하였다. 총 폴리페놀 함량과 총 플라보노이드 함량은 CR을 함유한 식혜가 대조구에 비해 유의적으로 높았으며, CR 농도 비율이 증가될수록 높았다. 특히, CR-1.0은 대조구에 비해 총 플라보노이드 함량이 약 3배 이상 높았다. DPPH 라디칼 소거능과 아질산염 소거능은 CR 첨가 비율이 증가할수록 유의적으로 증가하였다. CR-1.0은 대조구에 비해 DPPH 라디칼 소거능은 약 3배이상, 아질산염 소거능은 약 3.8배 이상 증가하였다. CR 첨가 식혜의 종합적 기호도는 CR-1.0(3.04)을 제외한 나머지 CR 첨가구는 모두 대조구(3.32)보다 높았으며 특히, CR-0.6은 종합적 기호도 뿐만 아니라, 맛, 색, 향에서도 가장 높은 평가를 나타내었다. $4^{\circ}C$에서 20일 동안 저장한 식혜의 일반세균수를 조사한 결과, 대조군은 $10^3CFU/mL$이었고, CR을 첨가한 식혜는 $10^1-10^2CFU/mL$를 나타내었다.
