Synthesis and Characterization of Amphiphilic Poly(4-vinyl benzoic acid-co-diethylacrylamide) for Hydrotropic Solubilization of Paclitaxel

This study describes the synthesis and characterization of micelle-forming amphiphilic copolymers comprised of vinylbenzoic acid (VBA) and diethylacrylamide (DEAAm) for hydrotropic solubilization of paclitaxel. For the synthesis of the copolymer, N-hydroxysuccinimide activated VBA (NSVB) and DEAAm were randomly copolymerized via reversible addition-fragmentation chain transfer (RAFT) polymerization to obtain poly(NSVB-co-DEAAm)(PND), followed by grafting of amine-terminated poly(ethylene glycol) methyl ether (mPEG) to NSVB. The resulting poly(VB-co-DEAAm)-gmPEG( PVDP) and PND were characterized by $^1H$-NMR and TGA. It was found that PVDP could significantly improve water solubility of PTX with increasing concentration of PVDP. At a low concentration, the aqueous solution of PVDP containing PTX looked opaque but became transparent with increasing PVDP concentration. The water solubility of PTX was increased up to 10000-fold at 8 wt% of PVDP, as compared with PTX alone. We measured the average diameter of PVDP at this concentration using a dynamic light scattering. The result showed that PVDP had $137{\pm}18nm$ in diameter, indicating that the improved water solubility of PTX was probably due to the micelle-forming amphiphilic nature of hydrotropic PVDP. Therefore, we expect that the amphiphilic PVDP copolymer with a hydrotropic property can be used to improve water solubility of poorly water-soluble drugs including PTX.
