Abstract
This experiment was conducted to identify the enzyme treatment time, calcium ion effect, enzyme concentration and leaf position for protoplast isolation. It was also performed to determine the adequate molarity on protoplasts, and to investigate the incubation time, pH, PEG concentration and DMSO effect for protoplast fusion. The results obtained were summarized as follows ; The optimal time of incubation in enzyme solution was 4 hours. And the protoplast releasing time was delayed by
