Studies on Cellulolytic Enzymes Produced by Pleurotus spp. in Synthetic Medium( I ) -Effects of Carbon and Nitrogen Sources-

합성배지(合成培地)에서 Pleurots속(屬)이 생산(生産)하는 섬유소(纖維素) 분해효소(分解酵素)에 관한 연구(硏究)(제1보)(第1報) -탄소원(炭素源)과 질소원(窒素源)의 영향(影響)-

Abstract

Among the eight strains, Pleurotus sajor-caju JAFM 1017 was selected as most potent producer of cellulolytic enzymes. The avicelase and CMCase activity reached maximum levels after 10 days, and ${\beta}-glucosidase$ activity reached a maximum level after 19 days. Among the various carbon sources, cellulose powder was most effective for the production of avicelase and ${\beta}-glucosidase$, and Na-CMC (sodium carboxymethyl cellulose) was good for the production of CMCase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose (1.0%) completely depressed the production of enzymes. Nitrates were effective for the production of enzymes, but nitrites did not support growth. The production of cellulolytic enzymes increased as the concentration of urea increased. The appropriate concentration of urea was 0.054% (w/v).

8개의 Pleurotus spp.를 합성배지(合成培地)에 배양(培養)하여 섬유소분해효소(纖維素分解酵素) 생산력(生産力)이 제일 강하고 균사생육(菌絲生育)이 양호(良好)한 균사(菌絲)를 선별(選別)하여 배양기간(培養期間) 및 탄소원(炭素源), 실소원(室素源)의 영향(影響)을 검토(檢討)한 결과(結果)는 다음과 같다. 섬유소분해효소(纖維素分解酵素) 생산력(生産力)이 강한 균주(菌株)로 Pleurotus sajor-caju JAFM 1017을 선별(選別)하였다. Avicelase와 CMCase활성(活性)은 10일(日) 배양(培養)하였을때 최대치(最大値)를 보였으며 ${\beta}-glucosidase$ 활성(活性)은 19일(日) 배양(培養)하였을 때가 가장 높았다. 탄소원(炭素源)중에서 avicelase와 ${\beta}-glucosidase$ 생산(生産)은 cellulose power를, CMCase 생산(生産)은 Na-CMC를 첨가(添加)하였을 때가 가장 좋았다. 효소생산(酵素生産)은 cellulose powder 1.0%에서 가장 양호(良好))하였으며 glucose는 농도(濃度)의 증가(增加)에 따라 현저하게 감소(減少)되었다. 효소생산(酵素生産)에 양호(良好)한 실소원(室素源)은 urea였고 아실산성(亞室酸性) 실소(室素)는 효소생산(酵素生産)을 억제(抑制)하였으며 효소생산(酵素生産)에 적당(適當)한 urea의 농도(濃度)는 0.054%였다.
